The Right Key, the Wrong Lock: TIGIT Checkpoint Blockade and the Road to Precision Immunotherapy.

T-cell immunoreceptor with immunoglobulin and immunoreceptor tyrosine-based inhibitory motif (ITIM) domains (TIGIT) emerged as one of the most promising next-generation immune checkpoint targets following the success of programmed cell death protein 1 (PD-1), programmed death-ligand 1 (PD-L1), and cytotoxic T-lymphocyte-associated protein 4 (CTLA-4) blockade. TIGIT suppresses antitumor immunity through interaction with cluster of differentiation 155 (CD155), inhibition of CD226-mediated co-stimulation, and promotion of immunosuppressive regulatory T-cell (Treg) activity within the tumor microe

Amniotic Mesenchymal Stromal Cell Administration Prevents and Stops Lung Fibrosis and Is Associated with Distinct Macrophage Signatures.

Background/Objectives: Mesenchymal stromal cells from the amniotic membrane (hAMSCs) counteract fibrosis progression, primarily via anti-inflammatory effects like promoting macrophage polarization toward an anti-inflammatory, pro-regenerative phenotype. Methods: We investigated hAMSCs' ability to prevent and halt lung fibrosis in a bleomycin-induced fibrosis murine model. We focused on their impact on recruitment and polarization of different macrophage populations, including SPARC- and CD169-expressing macrophages, implicated in resolving pulmonary inflammation and fibrosis. Results: hAMSCs,

Biopsied Cutaneous Adverse Effects of PD-1 and PD-L1 Checkpoint Inhibitors and Progression-free Survival: A Retrospective Case Series of 75 Patients.

Programmed cell death-1 checkpoint inhibitors have significantly improved cancer survival outcomes. More than half of patients on programmed cell death-1 checkpoint inhibitors develop cutaneous immune-related complications. The prognostic significance of eruptions biopsied in routine clinical practice remains underexplored. We aimed to characterize biopsy-confirmed cutaneous toxicities associated with checkpoint inhibitors and assess their association with survival. We conducted a retrospective study of biopsy-confirmed checkpoint inhibitor-associated dermatoses at our single institution betwe

Transcriptomics and single-cell RNA sequencing uncover prognostic characteristics and immune regulatory mechanisms of lactylation in cervical cancer.

INTRODUCTION: Lactylation, an emerging post-translational modification, modulates tumor metabolism and gene expression, thereby influencing the initiation and progression of cervical squamous cell carcinoma (CESC). This study aims to identify lactylation-associated prognostic features in CESC through the integration of transcriptomic data and single-cell RNA sequencing (scRNA-seq). METHODS: Publicly available datasets were utilized for this study. Lactylation-related genes (LRGs) linked to CESC prognosis were found through analysis. Key prognostic genes include PGK1, PFKP, DDX39A, RFC4, WAS,

Neuromedin U 'innervation' reprograms myeloid immunosuppression in neuroendocrine prostate cancer.

Neuroendocrine prostate cancer (NEPC) is an increasingly recognized, highly aggressive disease variant with no actionable therapeutic targets and a life expectancy of 7 months or less. Using a transgenic mouse model, we now show that early stages of NEPC are associated with increased intraprostatic recruitment of Ly6G+ polymorphonuclear neutrophils (PMN) and reduced infiltration of CD8+ T cells. This coincided with expansive transcriptional changes of increased cell viability and cell migration, as well as upregulation of multiple neuronal mediators with the neuropeptide, Neuromedin U (NMU) as

Parkinson's disease-linked D620N mutation selectively alters the brain-specific protein interactome of VPS35.

Mutations in several genes are known to cause familial forms of Parkinson's disease (PD), including mutations in the vacuolar protein sorting 35 ortholog (VPS35) gene linked to late-onset, autosomal dominant PD. VPS35 encodes a core subunit of the retromer complex which functions in endosomal sorting and recycling. It remains unclear how the pathogenic D620N mutation in VPS35 disrupts retromer function to induce neurodegeneration in PD. Using cell- and rodent-based models expressing D620N VPS35, we performed interactome proteomics to identify alterations underlying the pathogenic effects of D6

Peripheral γδ T-cell remodeling in psoriasis is associated with effector-biased transcriptional organization and reduced representation of a circulating CSMD1-marked state.

BACKGROUND: Despite the prominent immune dysregulation in psoriasis, the organization of peripheral blood γδ T cell states remains incompletely defined. METHODS: Here, we performed single-cell transcriptomic profiling of enriched peripheral blood-derived γδ T cells from healthy donors and patients with psoriasis. By integrating state annotation, inferred transcriptional ordering, ligand-receptor communication, transcriptome-inferred metabolite-sensor communication, and regulatory network analyses, we constructed a circulating γδ T-cell state atlas. RESULTS: We identified eight γδ T-cell stat

Mixed T-cell chimerism at 3 months predicts late relapse in acute myeloid leukemia after allogeneic stem cell transplantation.

Relapse remains the leading cause of treatment failure after allogeneic stem cell transplantation (HCT) in acute myeloid leukemia (AML), yet predictors of late relapse remain poorly defined. We conducted a multicenter retrospective study including 388 patients with AML, representing a disease-specific cohort undergoing first HCT across six Scandinavian centers to evaluate the prognostic role of early T-cell chimerism assessed at three months post-HCT. Mixed T-cell chimerism (MTC) was defined as < 95% donor-derived CD3 + T cells. Among patients alive and relapse-free at 12 months, three-month M

Immune monitoring by Torque Teno virus load in addition to virus-specific T cells after pediatric kidney transplantation.

Pharmacokinetic monitoring is insufficient to estimate the intensity of immunosuppression after kidney transplantation (Tx). The randomized controlled IVIST trial demonstrated that additional steering of immunosuppressive therapy by virus-specific CD4+ T cells (Tvis) is safe and reduces exposure to immunosuppressants. The adenovirus-specific CD4+ T cells (ADV-Tvis) proved to be particularly suitable due to their stability and high prevalence. Another promising biomarker for post-Tx immunomonitoring is the Torque Teno virus (TTV) but pediatric data are very limited. This descriptive longitudina

Stage-specific ROMO1 in rheumatoid arthritis: predictive immune insights into the MIF pathway and HLA-DR/IL2RA axis via integrated GWAS, transcriptomic, single-cell, and spatial profiling.

Emerging evidence links reactive oxygen species modulator 1 (ROMO1), a key mitochondrial ROS regulator, to rheumatoid arthritis (RA) pathogenesis. However, its exact mechanism remains elusive given the conflicting evidence about its specific function. We used a four-level integrative framework combining multi-omics data and literature‑supported mechanistic inference. At the genetic level, Mendelian randomization (MR) was performed to explore potential causal relationships between ROMO1, IL2RA, HLA-DR, MIF, and RA risk, followed by differential expression analysis and machine learning-based fea

Myositis with nemaline rods: Clinical features and treatment outcomes.

BACKGROUND: Several idiopathic inflammatory myopathies (IIM), originally classified under polymyositis, evolved into distinct entities. Herein, we describe clinical features and treatment outcomes of a potentially novel clinico-histopathological entity: myositis with nemaline rods (MNR). METHODS: We performed retrospective chart review of the Mayo Clinic electronic medical records to identify patients with nemaline rods and muscle inflammation on biopsy, not fitting into any IIM subgroup, and extracted clinical and laboratory data. Additional immunohistochemical studies were performed on musc

p97-mediated proteostasis is a checkpoint for late-stage thymocyte positive selection.

AAA+ ATPase p97 is a central regulator of protein homeostasis, yet its role in late-stage thymocyte development remains undefined. Here, we demonstrate that T-cell-specific ablation of p97 in mice severely blocks the double-positive (DP) to single-positive (SP) transition, with a pronounced defect in CD8+ lineage commitment. Using both genetic deletion and acute pharmacological inhibition, we revealed a stage- and lineage-specific requirement for p97, with DP thymocytes being most sensitive to p97 loss. This failure in late-stage positive selection leads to intrathymic developmental arrest of

A serpin-myeloid axis in pancreatic cancer heterogeneity and immune evasion.

Pancreatic ductal carcinoma (PDAC) is characterized by a highly immunosuppressive, extracellular matrix-rich microenvironment, yet tumours display marked heterogeneity1-4. This raises the question of whether immune resistance is a global tumour property or is organized within spatially restricted niches. Here, using Perturb-map spatial functional genomics, we determine how different genes shape the growth and cellular environments of PDAC clones across space and time. This analysis revealed early gene-driven remodelling of local immune neighbourhoods preceding late-stage spatial clonal dominan

Characterization of the occurrence and distribution of specific immune cells throughout the esophageal squamous cell carcinoma progression.

Esophageal squamous cell carcinoma (ESCC) is recognized as one of the most prevalent malignancies globally, with a notably high incidence in China. The disease often presents with subtle symptoms in its early stages, resulting in diagnoses at mid to late stages, which diminishes the possibility for radical surgical intervention. Additionally, the effectiveness, side effects, and resistance associated with radiotherapy and chemotherapy further constrain their use in treating ESCC. The advent of immune checkpoint inhibitors has expanded the array of immunotherapeutic strategies available for man

Recognition and management of early complications in CAR-T cell therapy and virus-specific T-lymphocyte therapy: a practical clinical reference.

BACKGROUND: Cellular immunotherapies-including chimeric antigen receptor T-cell (CAR-T) therapies and adoptive transfer of virus-specific T lymphocytes (VSTs) - have transformed the treatment of refractory hematological malignancies and post-transplant infectious complications. Eight products are currently authorized by the European Medicines Agency, encompassing seven autologous CAR-T products targeting CD19 or BCMA and tabelecleucel (Ebvallo), the first approved allogeneic off-the-shelf EBV-specific T-cell product for EBV-positive post-transplant lymphoproliferative disease. The β-hCG-TNF-α-T cell axis in threatened abortion: unraveling the pathogenic cascade from aberrant glycosylation to inflammatory demise.

Threatened abortion (TA), vaginal bleeding before 20 weeks in a viable pregnancy, affects 20-25% of early pregnancies. Its unpredictable nature is a major clinical challenge. Current diagnostics (β-hCG and ultrasound) are retrospective and fail to reveal the dynamic pathology. This review presents the "Danger Signal" Hypothesis, focusing on the β-hCG-TNF-α-T cell axis, to explain TA pathogenesis. The process involves three stages: 1) Initiation: Stressed trophoblasts secrete aberrantly glycosylated β-hCG, which acts as a DAMP. 2) Amplification: This DAMP triggers a TNF-α-dominated cytokine sto

Placental calcineurin/NFATc1 expression and angiogenic profiles in isolated fetal growth restriction: A prospective longitudinal study.

INTRODUCTION: The calcineurin/nuclear factor of activated T-cells (NFAT) signaling pathway regulates soluble fms-like tyrosine kinase-1 (sFlt-1) expression in placental tissue; however, its role in isolated fetal growth restriction (FGR) has not been investigated. METHODS: This prospective longitudinal cohort study enrolled 39 women with isolated FGR (15 early-onset <32 weeks, 24 late-onset ≥32 weeks). Placental calcineurin and NFATc1 were quantified by ELISA at delivery. Serum sFlt-1, placental growth factor (PlGF), and vascular endothelial growth factor (VEGF), together with urinary sFlt-1

T-Cell Engagers in Lung Cancer: A Comprehensive Literature Review from Tarlatamab Approval to Next-Generation Strategies.

BACKGROUND: Lung cancer remains the leading cause of cancer-related mortality worldwide, with five-year survival below 5% for metastatic small cell lung cancer (SCLC) and below 10% for metastatic non-small cell lung cancer (NSCLC). Immune checkpoint inhibitors have improved outcomes, but primary and acquired resistance, driven by tumor microenvironment immunosuppression, antigen heterogeneity, and T-cell exhaustion, leaves a substantial unmet need. T-cell engagers (TCEs), bispecific antibodies that redirect cytotoxic T-cells to tumor cells independent of MHC-I-restricted antigen presentation,
