Turning cold tumors hot: Strategies to enhance antitumor immunity.

Immunologically cold tumors remain a major obstacle to effective cancer immunotherapy because they lack sufficient immune infiltration and fail to sustain productive antitumor immune responses. Although substantial progress has been made in understanding the mechanisms underlying immune evasion, only a minority of patients achieve durable benefit from current immunotherapeutic approaches. In this review, we discuss emerging strategies aimed at converting cold tumors into immune-responsive lesions by enhancing tumor antigenicity and promoting effective immune priming. We examine how these appro

Hyaluronan fragments modulate inflammation in preeclampsia: a focus on Th17/Treg balance.

Pregnant women with preeclampsia (PE) exhibit an exacerbated inflammatory immune response. Hyaluronan (HA), a damage-associated molecular pattern (DAMP), is increased in the plasma of women with PE and may differentially modulate immune responses depending on its molecular weight. Forty-five women were investigated, including 20 with PE, 20 normotensive (NT) pregnant women, and 5 healthy non-pregnant (NP) women. Plasma samples were obtained for TNF, IL-10, HA, and soluble CD44 (sCD44) analyses by ELISA. Expression of the CD44 receptor, as well as intracytoplasmic transcription factors T-bet (T

Transcriptomic analysis reveals AURKB promotes malignant progression of hepatocellular carcinoma.

Due to the poor prognosis of hepatocellular carcinoma (HCC), there is an urgent need to identify novel therapeutic targets. This study aims to evaluate the potential of Aurora kinase B (AURKB) as a therapeutic target for HCC and provide a theoretical basis for immunotherapy in HCC. In this study, 374 patients were included from The Cancer Genome Atlas (TCGA) database, and single-sample gene set enrichment analysis was performed to classify them into high-immunity and low-immunity groups. Through differential gene expression analysis and univariate Cox regression analysis, AURKB was screened ou

Allergenicity reduction of silkworm pupae allergen Bomb m 6 via ultrasound-assisted xylosylation: Structural disruption and epitope-masking effects.

Silkworm pupae (Bombyx mori) are a promising sustainable protein, but their use is limited by potent allergens like Bomb m 6. This study developed an ultrasound-assisted xylosylation method to mitigate its allergenicity. Ultrasound-assisted xylosylation effectively altered the structure of Bomb m 6 and introduced 8 new glycation sites within known B-cell epitopes, suggesting a potential for epitope masking. These modifications reduced IgE-binding capacity by nearly 2-fold, which remained sustained even after simulated gastrointestinal digestion. In vivo, mice sensitized with ultrasound-xylosyl

Association of peripheral CD4⁺/CD8⁺ T-cell ratio with SYNTAX score and coronary stenosis severity in patients with coronary artery disease.

BACKGROUND: Coronary Artery Disease (CAD) is characterized by chronic immune-inflammatory activation. The relationship between the peripheral CD4+/CD8+ T-cell ratio and coronary stenosis severity remains unclear. We investigated its association with SYNTAX score and potential as a complementary marker. METHODS: CD4+ and CD8+ T-cell counts were measured by flow cytometry in 297 patients with CAD and 181 controls. Coronary stenosis was assessed by angiography and SYNTAX score. Multivariable linear regression was used for SYNTAX score, and logistic regression for moderate-to-severe stenosis (SYN

Association of early CD4⁺ T-lymphocyte depletion with 28 day mortality in sepsis: a retrospective single-center cohort study.

INTRODUCTION: Sepsis-induced immune dysregulation, characterized by T-cell loss, is associated with poor outcomes. This study investigated the association between early peripheral T-lymphocyte subset profiles and 28 day mortality in patients with sepsis. METHODS: In this single-center retrospective cohort study, 60 critically ill patients with sepsis and 30 healthy controls were included. Patients with sepsis were classified as 28 day survivors (n = 30) or non-survivors (n = 30). Peripheral T-lymphocyte subset percentages (CD3⁺, CD4⁺, and CD8⁺) were measured by flow cytometry within 24 h of I

Successful conservative management of hepatic portal venous gas complicating severe immune-related gastrointestinal injury during pembrolizumab-based concurrent chemoradiotherapy for cervical cancer: a mechanistic case report.

Concurrent chemoradiotherapy (CCRT) combined with pembrolizumab has become the standard treatment for high-risk locally advanced cervical cancer, increasing the need for prompt recognition of immune-related adverse events (irAEs). We report a rare case of severe gastrointestinal irAE complicated by hepatic portal venous gas (HPVG) during pembrolizumab-based CCRT. A 72-year-old woman with FIGO 2018 stage IIIC1r cervical squamous cell carcinoma developed persistent diarrhea and anorexia during treatment, followed by sudden hematemesis after the first brachytherapy session. Upper gastrointestinal

Suppressive activity and regulatory T-cell phenotypes of cells generated from human peripheral blood mononuclear cells under CD80/CD86 costimulation blockade.

BACKGROUND: Costimulation blockade targeting CD80/CD86 has been widely investigated as an immunomodulatory strategy in transplantation. However, its effects on human peripheral immune cells and the induction of regulatory phenotypes remain incompletely understood. METHODS: Human peripheral blood mononuclear cells (PBMCs) were cultured under CD80/CD86 costimulation blockade. Suppressive activity was evaluated using mixed lymphocyte reactions (MLRs). The contribution of T cells was assessed by CD3 depletion and reconstitution experiments. Regulatory T-cell phenotypes and cytokine production wer

[18F]F-AraG PET Imaging of Immunometabolic Activity in Individuals Without Malignancy.

[18F]F-AraG is a clinical-stage PET tracer in immuno-oncology that accumulates in activated T cells and tissues with high mitochondrial biogenesis. Characterizing its distribution in individuals without malignancy is essential for interpreting immunometabolic states relevant to cancer biology. Methods: Twenty adults without active malignancy underwent a single [18F]F-AraG PET/CT scan, with tracer uptake assessed in relation to clinical history, medications, and demographic and physiologic factors. Results: Bilateral brown adipose tissue activation was identified in 3 participants receiving pot

Spatiotemporal multi-omics reveals lineage, spatial and functional heterogeneity of tumor-associated macrophages and therapeutic vulnerabilities in the tumor microenvironment.

Tumor-associated macrophages (TAMs) are central orchestrators of immune evasion, therapeutic resistance, and clinical outcome across diverse solid tumors. However, the classical M1/M2 paradigm fails to capture the lineage diversity, spatial heterogeneity, and functional plasticity of TAMs within the tumor microenvironment. Unlike previous reviews that mainly summarize macrophage polarization or TAM subtype classification, this review focuses on how emerging single-cell and spatial multi-omics technologies redefine TAM ontogeny, phenotypic diversity, spatial organization, and therapeutic vulner

Antigen-Presentation Competence in Gynecologic Cancers: Mechanisms, Evidence and Translational Priorities.

Immune-checkpoint biomarkers in gynecologic cancers primarily estimate antigenic potential or characterize an inhibited immune response. However, they do not establish whether malignant cells are visible to antigen-specific lymphocytes. We define antigen-presentation competence as the capacity of the tumor-immune ecosystem to generate antigenic peptides, display them through functional human leukocyte antigen molecules, prime cognate T cells through professional antigen-presenting cells, and permit recognition of malignant cells by effector lymphocytes. Recurrent defects have been reported at

Evaluation of MRI Radiomic Features as Surrogates of Tumor Immune Status in Breast Cancer: A Pilot Study of MRI-Derived Radiomic Features for Non-Invasive Immune Profiling in Breast Cancer.

BACKGROUND: Tumor-infiltrating lymphocytes (TILs) and CD8+ T cell density are important predictors of immunotherapy response in breast cancer, but their evaluation requires invasive tissue biopsies. Radiomics, which enables high-throughput extraction of quantitative imaging features, offers a promising non-invasive approach to characterize the tumor immune microenvironment (TIME). This study evaluates whether MRI-derived radiomic features could predict immune infiltration status in breast cancer. Additionally, we have developed an on-line browser-based tool that can predict the immune status o

STAT6 Deficiency Accelerates DMBA/TPA-Induced Skin Papilloma Development and Is Associated with Regulatory T-Cell Accumulation and Reduced CD8+IFN-γ+ Responses.

Inflammation and the balance between antitumor surveillance and immunoregulatory mechanisms influence chemically induced skin tumor development. STAT6 is a central mediator of IL-4/IL-13 signaling, but its contribution to early skin papilloma development remains unclear. Here, we examined papilloma onset and burden, histopathological alterations, Treg accumulation, and CD8+ T-cell-associated responses in WT and STAT6-deficient mice subjected to a two-stage DMBA/TPA protocol. STAT6-/- mice developed papillomas significantly earlier and exhibited a markedly greater lesion burden than WT mice. Th

Maternal-Fetal Exposome and Early-Life Microbiota-Pattern Recognition Receptors Signaling in Inflammatory Bowel Disease Susceptibility: A Mechanistic Review.

Inflammatory bowel disease (IBD) develops through interactions among genetic susceptibility, intestinal microbiota, environmental exposures, and dysregulated mucosal immunity. Increasing evidence suggests that these interactions may begin before diagnosis-and possibly before birth-when maternal signals, microbial assembly, epithelial maturation, and immune education intersect. This narrative review synthesizes findings from human observational and cohort studies, experimental animal and cellular models, and mechanistic literature to examine how prenatal and early-life exposures may influence l

Differential Expression of Cytotoxic-Associated Markers in Circulating CD4+CD28- T Cells and NK Cells in Rheumatoid Arthritis.

Rheumatoid arthritis (RA) involves alterations in cytotoxic lymphoid differentiation, yet the precise phenotypic remodeling of circulating helper T cells and natural killer (NK) cells remains incompletely understood. We evaluated circulating CD4+CD28+ and CD4+CD28- T-cell subsets and NKG2A+CD56+ NK cells, alongside granule-mediated cytotoxic markers (granzyme B and perforin) and the surface-bound death receptor Fas ligand (FasL), in 28 female patients with RA and 12 age-matched healthy female controls using multiparameter flow cytometry. CD4+CD28- cells were increased in RA compared with contr

Regulatory T Cells Across Oral Diseases: A Narrative Review on a Functional Perspective.

Regulatory T cells (Tregs) maintain peripheral immune tolerance and are involved in various oral diseases with markedly different characteristics. In the oral field, Tregs have largely been discussed through reports on their distribution and numbers, and interpretations of whether their role is protective or pathogenic. Current identification markers do not directly assess the suppressive function, and reports on this function remain limited. This review focuses on the suppressive function of Tregs in the organization of oral diseases. Cells may express FOXP3 without exerting stable suppressiv

Cell Tropism-Related Gene Signatures in MDD Identified by Transcriptomic Analysis and Rat Model Validation.

Major depressive disorder (MDD) is associated with immune dysregulation, but the contribution of chemotaxis-related genes remains incompletely understood. This study aimed to identify chemotaxis-related signatures in MDD and evaluate their diagnostic relevance. Two peripheral-blood microarray datasets (GSE76826 and GSE38206) were integrated after batch correction, and differentially expressed genes were intersected with the GOBP_TAXIS gene set. Hub genes were selected using random forest and SVM-RFE and evaluated by logistic regression, ROC, calibration, and decision curve analyses. Immune-cel

Effects of Cd8a Gene Knockout on Gut Microbiota and Liver Metabolism.

OBJECTIVE: Evaluate the effects of CD8+ T cell defects caused by Cd8a gene knockout on gut microbiota and hepatic metabolism, and preliminarily explore the underlying mechanisms. METHODS: The Cd8a gene was knocked out in mice to generate Cd8a knockout mice. Genotypes were identified using polymerase chain reaction (PCR) and agarose gel electrophoresis. Mice were divided into three groups: normal control, CD8+ T cell-deficient, and CD8+ T cell reinfusion, with six mice per group. CD8+ T cells from normal control mice were isolated via magnetic bead sorting and reinfused into CD8+ T cell-defici
