Skeletal Muscle Index, Tumor and Systemic Immunity, and Survival in Advanced Pancreatic Ductal Adenocarcinoma.

BACKGROUND: This study investigated relationships between the skeletal muscle index (SMI) and the tumor immune microenvironment (TIME), peripheral immune phenotypes, and serum biomarkers in advanced pancreatic ductal adenocarcinoma (PDAC) and evaluated the prognostic significance of these parameters. METHODS: A retrospective analysis of 119 advanced PDAC patients was performed. Sarcopenia was diagnosed using sex-specific SMI cut-offs from baseline L3-level computed tomography (CT) images. The TIME was characterized by multiplex immunofluorescence (mIF; n = 31), peripheral immunity by flow cyt

[Clinical and morphological characteristics and prognostic significance of the immune microenvironment and the level of PD-L1 expression in squamous cell carcinoma of the tongue and oropharynx].

Squamous cell carcinoma of the tongue and oropharynx is characterized by significant clinical and biological heterogeneity, highlighting the need for reliable prognostic and predictive biomarkers. This review analyzes current evidence regarding the significance of programmed death-ligand 1 (PD-L1) expression and immune microenvironment characteristics in these malignancies. PD-L1 has been shown to play a central role in tumor immune evasion; however, its prognostic value remains controversial and appears to depend on human papillomavirus (HPV) status, tumor immune contexture, and methodologica

Synthetic Notch CD123 and CLL1 chimeric antigen receptor T-cells (CAR-T) target acute myeloid leukemia and reduce off-tumor effects associated with CLL1 CAR-T cells.

Chimeric antigen receptor T-cell (CAR-T) therapy targeting the C-type lectin-like molecule-1 (CLL1) antigen has emerged as a promising therapeutic approach for acute myeloid leukemia (AML). However, its clinical translation is hindered by on-target, off-tumor toxicity, particularly severe neutropenia caused by CLL1 expression on normal neutrophils. To address this limitation, we constructed a synthetic Notch (SynNotch) dual-antigen AND-gated CAR-T system targeting CD123 and CLL1 to improve tumor specificity and reduce off-tumor toxicity. CD123 and CLL1 expression was first assessed in AML cell

Lymphangiogenesis driven by VEGF-C reshapes tumor immune landscape and enables tumor eradication by viral immunotherapy in mouse melanoma models.

Vascular Endothelial Growth Factor C (VEGF-C), a key lymphangiogenic growth factor, has been implicated in improving responses to cancer immunotherapy. We investigated the impact of VEGF-C on the efficacy of oncolytic virotherapy with avian paramyxoviruses APMV-1 (NDV) and APMV-4. Treatment of B16F10 tumors lacking VEGF-C delays tumor growth, and complete responses (CRs) are rare. In contrast, virotherapy of VEGF-C-expressing tumors leads to CRs in most mice. Rechallenged mice remain tumor- and metastasis-free for over two years, indicating durable immunity. VEGF-C-induced lymphangiogenesis co

Single-cell analyses reveal lymph node-tumor ecosystem remodeling during lymph node metastasis of gastric cancer.

Gastric cancer (GC) is a highly aggressive malignancy with a strong propensity for lymph node metastasis. To characterize the metastatic ecosystem, we perform integrated single-cell transcriptomic and T cell receptor profiling of 156,871 cells from 30 primary tumors and matched sentinel lymph nodes (SLNs), spanning non-metastatic and metastatic tumors and SLNs from non-metastatic to micro- and macro-metastatic states. We identify LN-enriched progenitor exhausted T cells as a potential reservoir for tumor-infiltrating exhausted CD8+ T cells and reveal dynamic remodeling of the lymph node microe

Proteomic Landscape and Clinical Implications of Ovarian Carcinosarcoma.

Ovarian carcinosarcoma (OCS) is a rare and aggressive ovarian cancer subtype with poor prognosis and limited treatment options. This study aimed to provide an in-depth proteomic analysis of OCS to uncover molecular features associated with tumor progression, prognosis, and immune microenvironment characteristics. Using mass spectrometry-based proteomic profiling and bioinformatics, we analyzed 14 OCS and nine paired normal ovarian (NO) tissue samples. We identified significant proteomic alterations, including the upregulation of proteins involved in angiogenesis, cell proliferation, DNA repair

Loss of Acid Ceramidase in Myeloid Cells Protects From Chronic Colitis in IL10-Deficient Mice.

Patients with inflammatory bowel disease exhibit elevated expression of acid ceramidase (AC), an enzyme in sphingolipid metabolism. Recent studies have shown that myeloid cells contribute to the elevated expression of AC, such that the conditional loss in these cell types protects from acute colitis. We crossed conditional ASAH1 LyzMCRE knockout mice with IL10 knockout mice to determine the role of AC in a model of spontaneous, chronic colitis. Colon tissues were analyzed for lipids, mRNA, and protein. We performed flow cytometry to determine the role of myeloid AC in recruiting immune cells i

CTLA4 rs3087243-A-linked haplotype is associated with increased CTLA-4 expression and suppressive function in human regulatory T cells.

CTLA-4 is an inhibitory receptor on T cells that limits immune activation by competing with the co-stimulatory receptor CD28 for binding to CD80 and CD86 on antigen-presenting cells. Regulatory T cells (Tregs) employ CTLA-4-mediated transendocytosis as a key suppressive mechanism. Several CTLA4 polymorphisms are associated with autoimmune disease risk and may modulate CTLA-4 expression and function. Here, we analyzed samples from 155 genotyped subjects using flow cytometry and functional assays to determine how haplotypes formed by 3 CTLA4 single-nucleotide polymorphisms shape T-cell phenotype

Glycome remodeling programs pathogenic γδ T17 cell responses in autoimmunity.

Glycans play fundamental roles in T cell biology; however, the glycome landscape of γδ T cells remains to be unlocked. Despite the growing importance of γδ T cells in host defense, tissue homeostasis, and diseases, the glycosylation mechanisms guiding their activation, commitment, and function remain poorly defined. By performing single-cell RNA sequencing, we revealed that type 3/γδ T17/interleukin-17A (IL-17A)-committed γδ T cells rely on (N-acetylglucosaminyltransferase 1) MGAT1-MGAT5-mediated branched N-glycosylation. We further demonstrated that the absence of branched N-glycans on γδ T c

Immunosenescence in colorectal cancer: Interplay of microbiota, inflammaging, and metabolic reprogramming.

Colorectal cancer (CRC) represents a major cause of cancer-associated morbidity and mortality worldwide, with significantly higher incidence and lethality in elderly individuals. Growing evidence implicates immunosenescence, the age-dependent functional decline of the immune system, as a key contributor to CRC initiation, progression, and therapeutic resistance. In this review, telomere attrition, impaired DNA repair, mitochondrial dysfunction, oxidative stress, and epigenetic remodeling are discussed as interconnected molecular processes that contribute to immunosenescence. These processes di

Treg-associated oligomannose N-glycans trigger immune dysregulation in myasthenia gravis.

Myasthenia gravis (MG) is an autoimmune neurological disease driven by the production of autoantibodies against components of the neuromuscular junction, resulting in fatigable muscle weakness, and in more severe cases, leading to life-threatening complications. This breakdown of immune tolerance has been associated with an impaired suppressive function and altered frequency of Regulatory T cells (Tregs) in autoimmune diseases, including MG. As key regulators of immune tolerance, Tregs play a pivotal role in maintaining immune homeostasis through their ability to suppress other immune cells, c

REG family proteins and immune regulation: current evidence and perspectives.

As a group of small secretory proteins widely expressed in digestive and nondigestive tissues, the regenerating islet-derived (REG) family members have been highlighted for their multifunctional properties, such as pro-proliferative, mitogenic, anti-apoptotic, differentiation-inducing, anti-inflammatory, bactericidal, anti-oxidation and pro-tissue-repair activities. Emerging evidence indicates context-dependent immunomodulatory potentials of specific REG proteins. This review focused on the involvement and potential roles of REG family proteins in regulating immune cells. Available data sugges

Peripheral FOXP3+ regulatory T cells as potential predictive markers of concurrent chemoradiation therapy response in locally advanced cervical cancer.

Concurrent chemoradiation therapy (CCRT) is the standard treatment for locally advanced cervical cancer (LACC); however, approximately 30% of patients respond poorly, and alternative therapeutic options remain limited. We aimed to identify peripheral immune biomarkers associated with treatment response and characterize the immune mechanisms underlying clinical outcomes. Thirty patients with LACC undergoing CCRT were enrolled and classified three months after treatment as responders (n = 15) or non-responders (n = 15) according to Response Evaluation Criteria in Solid Tumors. In primary human lymphocytes, the triazole fungicide cyproconazole impairs regulatory B cell interleukin-10 expression and induces T cell senescence.

Triazole fungicides are applied worldwide, yet their effects on human lymphocytes remain poorly characterised. We examined whether cyproconazole impairs regulatory B cell function and induces T cell senescence. Purified B and T lymphocytes were exposed to cyproconazole (30-200µM, 48-72h). B cells were more susceptible to apoptosis than T cells, their regulatory output collapsing disproportionately to cell loss: IL-10 in viable CD19⁺ cells fell by 87% at 30µM, granzyme B declining only at higher concentrations. Cyproconazole drove T cells out of the cell cycle into G0 at 48h, before any viabili

Polymeric Nanoparticle Gene Delivery to Reprogram the Tumor Immune Microenvironment in Ovarian Cancer for an Anti-Tumor Response.

The tumor immune microenvironment (TIME) in ovarian cancer is highly immunosuppressive, and existing immunotherapies are not effective in 85-90% of patients. To address this, we fabricated nanoparticles (NPs) composed of biodegradable poly(β-amino ester) (PBAE) complexed with plasmids encoding 4-1BB ligand and interleukin-12, for delivery into ovarian cancer cells. Collectively with the intrinsic major histocompatibility complex (MHC):antigen signals in tumor cells, these NPs generate tumor-associated antigen-presenting cells (tAPCs), leading to a T-cell-mediated anti-tumor response in an anti

HSP70-Mediated Macrophage-Stromal Crosstalk is an Immunopathogenic Driver of Recurrent Implantation Failure.

BACKGROUND: Recurrent implantation failure (RIF) remains a major challenge in assisted reproductive technology (ART), affecting a considerable proportion of patients despite multiple transfers of high-quality embryos. Although impaired endometrial receptivity is recognized as a critical contributor to RIF, the molecular mechanisms responsible for establishing a non-receptive endometrial microenvironment remain incompletely understood. The proliferative phase precedes decidualization and subsequent embryo implantation, representing a critical period for endometrial preparation. The endometrium

The Triad of Vascular Ageing: Cellular Senescence, Inflammaging, and Epigenetic Remodelling in Atherosclerosis.

Atherosclerotic cardiovascular disease remains a leading global cause of the cardiovascular mortality burden, yet traditional lipid-centric models fail to explain the residual cardiovascular risk observed in older populations. This review, synthesising contemporary literature from 2020 onwards alongside foundational studies, explores the paradigm shift towards the 'geroscience hypothesis', positing that intrinsic biological ageing of the vascular wall is a potent amplifier and primary driver of atherogenesis acting in synergy with circulating lipids. We delineate a pathogenic triad: cellular s

Prospective phase I study of nivolumab to promote graft-versus-tumour in relapsed or refractory haematological malignancy post allogeneic haematopoietic stem cell transplantation.

Allogeneic haematopoietic stem cell transplantation (alloHSCT) relies on donor-derived T cells to mediate the graft-versus-tumour (GVT) effect, but relapse may result from impaired immune reconstitution, tumour antigen downregulation or T-cell exhaustion. This prospective phase I study evaluated the safety and efficacy of the PD-1 inhibitor, nivolumab, in enhancing GVT post-alloHSCT and explored the impact of baseline T-cell phenotypes on clinical outcomes. Fourteen patients with relapsed/refractory haematological malignancies post-alloHSCT received nivolumab every 2 weeks for up to 48 weeks.

Targeting FEN1 disrupts EIF4A3-mediated MUC1-C autoregulation: a novel strategy to suppress lung adenocarcinoma progression.

Flap endonuclease 1 (FEN1), a key enzyme in DNA metabolism, exhibits oncogenic properties in various cancers. However, its functional role and underlying mechanisms in lung adenocarcinoma (LUAD) progression and tumor immunity remain poorly defined. Here, we demonstrate that FEN1 is significantly upregulated in LUAD tissues and serves as an independent prognostic factor for poor survival. Genetic depletion of FEN1 suppressed LUAD cell proliferation by inducing G0/G1 cell cycle arrest and cellular senescence, while significantly impairing migration and invasion capabilities. Mechanistically, int

Platelet-derived microparticles induce CD8+ T cell senescence in chronic kidney disease via the transfer of microRNA-550a-5p.

Infection is a leading cause of mortality in patients with chronic kidney disease (CKD), in which T cell dysfunction is considered a typical feature. Although platelets are active participants in immune response, their role in regulating T cell function in patients with CKD remains unclear. In this study, we demonstrate that CD8+ T cell senescence may be a major contributor to the immunosuppressed state of patients with CKD, and that platelets derived from patients with CKD can induce premature CD8+ T cell senescence, potentially through mediating mitochondrial dysfunction. Further investigati
