Two Novel SLC5A5 Variants (Q263L and G350D) Causing Congenital Hypothyroidism.

SLC5A5 encodes sodium-iodide symporter (NIS), which transports inorganic iodide into thyroid cells. Biallelic loss-of-function variants in SLC5A5 cause thyroid dyshormonogenesis due to iodide transport defect (ITD). We report a Japanese sibling with ITD carrying novel compound heterozygous SLC5A5 variants (p. [Gln263Leu]; [Gly350Asp]). The elder brother was diagnosed with congenital hypothyroidism (CH) through newborn screening (NBS), while the younger brother, with a negative NBS result, developed CH-related symptoms at age 3 months. We characterized the two variant NIS proteins in vitro and

Common and Rare DUOX Variants in Patients With Congenital Hypothyroidism: Case-control Study and Family-based Analysis.

CONTEXT: Dual oxidases (DUOXs) are essential for thyroid hormone synthesis. Rare DUOX variations have been detected in patients with congenital hypothyroidism (CH); however, their mode of inheritance and genotype-phenotype correlations remain unclear. Additionally, no study has determined whether common DUOX variants confer a risk of CH. OBJECTIVE: To elucidate the molecular and clinical characteristics of CH caused by rare and common DUOX variants. METHODS: Targeted next-generation sequencing was performed on 203 trios (parents and their child with CH) to screen for rare DUOX variants. For

Drosophila AK-3, a homolog of human CKMT1B, is essential for spermiogenesis.

IN BRIEF: The assembly and maintenance of axonemal microtubules during spermatogenesis is essential for production of functional sperm. This study shows that arginine kinase 3 (AK-3) (human ortholog CKMT1B) is necessary for Drosophila spermiogenesis and its biological function is conserved from flies to humans. ABSTRACT: Spermatogenesis is a conserved process across animals, involving the proliferation and maintenance of germ stem cells, haploid spermatid production via meiosis and the generation of mature sperm with unique shapes. Our previous studies revealed that after ocnus (ocn) knockdow

Genetic and functional analysis of TUBB1 variants in congenital hypothyroidism.

BACKGROUND: Congenital hypothyroidism (CH) is the most common neonatal disorder, primarily caused by thyroid dysgenesis (TD). While the genetic cause has been identified in less than 5% of TD cases, there is an urgent need to investigate additional gene mutations that may be responsible. In 2018, TUBB1 was identified as a novel candidate gene associated with TD. Nevertheless, further research is required to confirm the role of TUBB1 in TD pathogenesis and the association between TUBB1 mutations and TD in humans. Based on the previous genetic analysis of TUBB1 in 289 Chinese TD patients, this s

Metabolite biomarkers of screening neonatal congenital hypothyroidism based on dried blood spot metabolomics.

Congenital hypothyroidism (CH) can be detected early during thyroid-stimulating hormone (TSH) screening based on dried blood spot (DBS) samples. However, it falls short in differentiating between neonates with elevated TSH levels and those with CH, which leads to many neonates undergoing secondary diagnosis through venous blood sampling. Metabolomics was used to analyze metabolic alterations in neonates with CH, and identify DBS-based metabolite biomarkers for CH screening to reduce secondary diagnosis. Based on non-targeted metabolomics, the metabolic alterations in neonates with CH were anal

CONGENITAL HYPOTHYROIDISM: FROM THEORY TO PRACTICE - A CLINICAL CASE.

Hypothyroidism (HT) is one of the common diseases in endocrinology, caused by a long-term, persistent deficiency of thyroid hormones or a deficiency of their effect on tissues. The deficiency of these hormones leads not only to a slowdown in the biochemical processes of growth and development of the child's body, disruption of metabolic processes, including lipid peroxidation and the blood clotting mechanism, but also to a delay in intellectual development and inhibition of the formation of personality structure and thinking. In this regard, early diagnosis, as well as timely and adequate ther

Screening and Functional Analysis of TPO Gene Mutations in Patients With Congenital Hypothyroidism.

Thyroid peroxidase (TPO) gene mutations have been reported as the most commonly reported pathogenic variants in congenital hypothyroidism (CH) caused by thyroid hormone (TH) synthesis disorders, the significance of some mutations remains unclear. The study analyzed 54 children diagnosed with CH who underwent whole-exome sequencing (WES). Functional analysis of the TPO p.K78Q variant, a novel variant of uncertain significance (VUS), revealed that it caused developmental defects in zebrafish and also disrupted thyroid axis gene expression, decreasing tg, dio1, dio2, trβ, nis, ttr and increasing

Perinatal thyroid hormone deficiency leads to oxidative stress-induced neuronal damage and activation of astrocytes in rat hippocampus: Neuroprotective effect of exercise.

Thyroid hormones play a crucial role in proper brain development. This study examined the effects of developmental thyroid hormone deficiency on neuronal survival, glial fibrillary acidic protein (GFAP)-positive cells, and oxidative stress biomarkers in the hippocampus of congenital hypothyroid rats. The effectiveness of treadmill exercise in attenuating oxidative stress-induced neuronal damage and astrocyte activation was also evaluated. Pregnant Wistar rats in the hypothyroid group received propylthiouracil in their drinking water from gestational day 6 until weaning, while control dams rece

Investigating TSHR gene variants in consanguineous families: novel insights into variable expression in familial congenital hypothyroidism.

BACKGROUND: A defective thyroid-stimulating hormone receptor (TSHR) gene is one of the main known genetic factors leading to congenital hypothyroidism (CH). However, the relationship between TSHR genotypes and phenotype and the underlying reason for the broad spectrum of phenotypes in the patients carrying TSHR gene defects have not yet been clearly established. This study aimed to investigate the genetics of patients with CH to identify TSHR defects and to explore the specific extrathyroidal defects and other phenotypic features in these patients to establish a genotype-phenotype correlation.

Mechanistic insights into TSH-mediated macrophage mitochondrial dysfunction via TSHR signaling in metabolic disorders.

The role of thyroid-stimulating hormone receptor (TSHR) in macrophages on adipose tissue metabolic disorders remains unclear. We generated macrophage-specific TSHR knockout mice (LysM-Cre Tshrfl/fl) using the Cre/LoxP system and induced subclinical hypothyroidism (SCH) via methimazole treatment. Metabolic/inflammatory phenotypes were assessed via glucose/insulin tolerance tests, histology, and molecular analyses. Mitochondrial function and proinflammatory polarization were examined in primary peritoneal macrophages and RAW264.7 cells under TSH stimulation. The results showed macrophage TSHR de

Toll-like Receptor 9 Mediates Epstein-Barr Virus-Aggravated Inflammation in a Mouse Model of Inflammatory Bowel Disease.

Background/Objectives: Inflammatory bowel disease (IBD) is a chronic inflammatory condition encompassing ulcerative colitis (UC) and Crohn's disease (CD). The role of environmental factors in the pathogenesis of IBD remains elusive. Nevertheless, evidence suggests a pivotal role of viruses, specifically Epstein-Barr virus (EBV), in the progression of IBD through mechanisms such as molecular mimicry and bystander activation. Our previous findings demonstrate EBV DNA's significant role in exacerbating colitis symptoms and elevating the levels of the pro-autoimmune cytokine interleukin-17A (IL-17

Harnessing the CD16-PI3Kγ axis in peritoneal CD14hi macrophages: A transformative approach to endometriosis management.

Endometriosis (EMS) is a prevalent estrogen-dependent gynecological disorder characterized by ectopic growth of endometrial tissue. Central to its pathophysiology, peritoneal CD14hi macrophages exhibit a pro-EMS phenotype and drive disease progression. These macrophages originate from CD16-CD14+ monocytes and co-express functional surface markers critical for EMS development. We demonstrate that endometrial stromal cells (ESCs) exert significantly stronger regulatory effects on CD14hi macrophage polarization than estrogen alone. Crucially, CD16 maintains peritoneal CD14hi macrophage homeostasi

Amiodarone-induced ocular and extra-ocular toxicity: a retrospective cohort study.

Amiodarone (AMD) is a largely employed anti-arrhythmic agent for the treatment of recurrent supraventricular and ventricular tachyarrhythmias. Because of its lipophilic properties, prolonged half-life and prevailing biliary excretion, it is not rarely responsible for potentially severe adverse events that can involve one or more organs with a prevalence ranging from 15% in the first year of drug intake to 50% in patients treated for a longer time. In addition to pro-arrhythmia effects, AMD toxicity may result in a variable combination of clinical manifestations, including visual impairment, th

Role of SerpinA3 in the Pathogenesis of Graves' Orbitopathy in Orbital Fibroblasts.

IMPORTANCE: Categorization systems for adverse events are not standardized across care settings and specialties and do not always include near miss events (events where there was potential for patient harm, but where no actual harm occurred), making it difficult to effectively assess patient safety for quality improvement. OBJECTIVE: To develop and assess interrater agreement on a classification system for adverse events reporting that incorporates events in both inpatient and outpatient settings across medical and surgical subspecialties including near miss events. Sequencing in over 50,000 cases identifies coding and structural variation underlying atrial fibrillation risk.

Atrial fibrillation (AF) is a prevalent and morbid abnormality of the heart rhythm with a strong genetic component. Here, we meta-analyzed genome and exome sequencing data from 36 studies that included 52,416 AF cases and 277,762 controls. In burden tests of rare coding variation, we identified novel associations between AF and the genes MYBPC3, LMNA, PKP2, FAM189A2 and KDM5B. We further identified associations between AF and rare structural variants owing to deletions in CTNNA3 and duplications of GATA4. We broadly replicated our findings in independent samples from MyCode, deCODE and UK Biob
