Exploiting subtractive genomics to identify novel drug targets and new immunogenic candidates against Bordetella pertussis: an in silico study.

BACKGROUND: Bordetella pertussis, the causative agent of whooping cough, remains a significant global health concern despite the widespread availability of vaccines. The persistent reemergence of pertussis is driven by the bacterium's ongoing genomic evolution, shifting epidemiological patterns, and limitations in current vaccine strategies. These challenges highlight the urgent need to identify novel drug targets and immunogenic candidates to enhance therapeutic and preventive measures against B. pertussis. METHODS: Identification of novel drug targets and the detection of immunogenic factor

In vivo RNA-seq and infection model reveal the different infection and immune characteristics of B. pertussis strains in China.

INTRODUCTION: Various strains emerged in B. pertussis re-emergence, the pathogenic characteristics and mechanisms remain elusive. We aimed to explore the relationship between the in vivo transcriptome and colonization advantage of various pertussis clinical strains during the B. pertussis re-emergence. METHODS: Four pertussis strains were isolated from clinically suspected cases by active surveillance. The phylogenetic relationships of clinical strains and global isolates were compared by a genome-wide SNP-based phylogenetic tree and allele genotyping. LC-MS/MS analysis and binding affinity d

Atypical Presentation of Bordetella pertussis in a Six-Month-Old with Cleaved Lymphocytes and Mild Respiratory Distress.

Bordetella pertussis, the causative agent of whooping cough, typically presents in infants with paroxysmal coughing, inspiratory whoop, and post-tussive vomiting; however, atypical presentations are increasingly recognized, particularly in partially or recently immunized individuals. We present the case of a six-month-old previously healthy, fully immunized female infant who was evaluated for a three-day history of low-grade fever, feeding difficulties, increased irritability, and mild respiratory symptoms, in the absence of the characteristic pertussis cough or respiratory distress. A highly conserved sRNA downregulates multiple genes, including a σ54 transcriptional activator, in the virulence mode of Bordetella pertussis.

Bacterial small non-coding RNA (sRNAs), together with the RNA chaperone Hfq, post-transcriptionally regulate gene expression by affecting ribosome binding or mRNA stability. In the human pathogen Bordetella pertussis, the causative agent of whooping cough, hundreds of sRNAs have been identified, but their roles in B. pertussis biology are mostly unknown. Here we characterize the Hfq-dependent sRNABpsA, whose level is dramatically higher in the virulence (Bvg+) mode. We show that transcription from a σA-dependent promoter yields a long form of 190 nucleotides (nt) that is processed by RNase E t

Bordetella pertussis infection and antibody dynamics in household cohorts in two South African communities, 2016 - 2018: findings from the PHIRST study.

INTRODUCTION AND METHODS: In a cohort study, enrolling new households annually during 2016-2018, we estimated the B. pertussis attack rate using serology and PCR, identified factors associated with seroconversion, and described antibody changes over time. Nasopharyngeal swabs were collected biweekly for 6-10 months annually, with cases defined as individuals testing PCR-positive at least once. Seroconversion was defined as a ≥4-fold increase in anti-pertussis toxin IgG concentration between consecutive blood draws. Logistic regression was used to identify factors associated with seroconversion

Limited response of primary nasal epithelial cells to Bordetella pertussis infection.

Bordetella pertussis is a Gram-negative coccobacillus that causes whooping cough or pertussis, a respiratory disease that has recently experienced a resurgence. Upon entering the respiratory tract, B. pertussis colonizes the airway epithelium and attaches to ciliated cells. Here, we used primary human nasal epithelial cells (hNECs) cultured at the air-liquid interface and investigated their interaction with B. pertussis B1917, focusing on the role of the type III secretion system effector protein BteA. In this model, which resembles the epithelial cells of nasal epithelium in vivo, B. pertussi

Is It Time to Consider Hydroxyurea as a Potential Therapeutic Option in the Management of Malignant Pertussis - A Case Report and Literature Review.

Malignant pertussis is a life-threatening condition in infants, for which no effective evidence-based treatment strategies have been established, owing to the rarity of the condition. We present the case of a four-month-old infant with paroxysmal coughing spells followed by a whooping sound and shortness of breath with a leukocyte count of 41.82 × 109/L and lymphocytosis (32.25 × 109/L). A diagnosis of malignant pertussis, based on the severe paroxysmal cough, significant lymphocytosis, and progressive respiratory symptoms, was established, and treatment was initiated with azithromycin and hyd

Establishment of a rapid Bordetella pertussis detection method based on RPA-CRISPR-Cas12a technology.

Whooping cough (pertussis) is an acute respiratory infectious disease caused by Bordetella pertussis (BP). It poses a risk to infants and young children. This investigation aimed to construct a simple, rapid, and accurate diagnostic protocol for BP detection that does not depend on complex equipment or large-scale instruments. This study combines Recombinase Polymerase Amplification (RPA) technology with the CRISPR/Cas12a system, utilizing immunochromatographic lateral flow strips (ILFS) test and fluorescence curves to observe data. This diagnostic strategy does not require complex equipment u

Resurgence of pertussis: whopping the '100-day cough'.

PURPOSE OF REVIEW: Against the WHO's report of 84% diphtheria-pertussis-tetanus (DPT) primary vaccination coverage globally, the resurgence of pertussis (whooping cough), contributing factors and measures to control it are described. RECENT FINDINGS: USA and China, with 94-97% primary DPT immunization uptake, reported a 6-fold and 65-fold increase in pertussis between two time periods in 2023 and 2024. The global post-COVID-19 pertussis epidemic is trending towards a shift from infants towards older persons. Macrolide resistance is prevalent in 98% of Bordetella pertussis strains in China and

Macrolide-Resistant Bordetella pertussis in Hong Kong: Evidence for Post-COVID-19 Emergence of ptxP3-Lineage MT28 Clone from a Hospital-Based Surveillance Study.

A post-COVID surge of Bordetella pertussis was observed globally. China has reported a high level of macrolide-resistant Bordetella pertussis (MRBP) in recent years; however, the epidemiology of MRBP in Hong Kong remains unknown. We retrieved archived B. pertussis isolates from respiratory samples collected at five regional public hospitals in Hong Kong between 2015 and 2024 and tested their minimum inhibitory concentration (MIC) for macrolides and other non-macrolide antibiotics using the Etest method. All isolates were also subjected to whole genome sequencing for genotypic resistance, Multi

Absence of Macrolide-Resistant Mutations in Bordetella pertussis in Antananarivo (Madagascar) and Cambodia During the Last Pertussis Cycle Before the COVID-19 Pandemic.

Macrolides are the first-line treatment against pertussis. The high prevalence of macrolide-resistant Bordetella pertussis (MRBP) strains reported in China is concerning. MBRP might be underestimated in other countries. Looking for the mutation causative for MRBP in retrospective studies conducted in Madagascar and Cambodia, we found no MRBP.

A dual role for PGLYRP1 in host defense and immune regulation during B. pertussis infection.

Bordetella pertussis, the etiologic agent of whooping cough, remains a serious public health concern despite widespread vaccination. Improved therapeutics and vaccines are urgently needed to treat and prevent pertussis disease. Host recognition of bacterial peptidoglycan (PGN), including B. pertussis extracellular PGN fragment tracheal cytotoxin (TCT), shapes the immune response to infection. Peptidoglycan recognition proteins (PGLYRPs) are a conserved family of innate immune molecules which bind bacterial PGN. While they function as immune signaling receptors in arthropods, PGLYRPs in mammals

Natural immune boosting biases pertussis infection estimates in seroprevalence studies.

Seroepidemiology has significant potential for uncovering the unreported burden of infectious diseases. However, for diseases without well-defined serological correlates of protection, natural immune boosting-whereby pathogen exposure triggers a detectable immune response without causing a transmissible infection-can complicate the interpretation of serosurveys. This issue is relevant to pertussis, a vaccine-preventable disease that remains a significant public health concern worldwide. Here, we aimed to evaluate the reliability of pertussis serosurveys using a transmission model that tracked
