A Principled Statistical Framework for Analyzing Spatial Patterns in Spatially Resolved Multi-Omics.

Emerging spatial multi-omics technologies enable the profiling of molecular variation within its tissue context, yet existing methods for identifying spatially variable features lack principled approaches to experimental design and cross-sample inference. Here, we present STORM, a principled S tatistical TO ol for spatially R esolved M ulti-omics, for rigorously analyzing spatial patterns in spatial multi-omics research. STORM incorporates a robust and efficient nonparametric test that quantifies local deviations in molecular feature measurements to detect spatial dependence across transcripto

Flooding with brackish water disrupts Liriodendron tulipifera sap flux in an ecosystem-scale experiment.

Coastal forests are retreating due to sea‑level rise and extreme precipitation events, yet the physiological mechanisms that precede woody plant decline remain poorly constrained. We used the ecosystem‑scale TEMPEST (Terrestrial Ecosystem Manipulation to Probe the Effects of Storm Treatments) experiment; annual, plot‑level freshwater and saltwater flooding of a mid‑Atlantic deciduous forest, to quantify short‑ and long‑term responses of sap flux in three tree species: Acer rubrum, Fagus grandifolia, and Liriodendron tulipifera. We analyzed five years of sap flux density measurements (2021-2025

Microplastic pollution in Greek seas: Abundance, spatio-temporal dynamics, potential sources and post-storm amplification.

Microplastic (MP) pollution represents an increasing threat to coastal and marine ecosystems, yet data from Greek waters remain scarce. This study provides a comprehensive assessment of MP abundance, spatial distribution, polymer composition, and seasonal variability in surface waters and sediments collected from 27 coastal sites in Greece between 2021 and 2024. Sampling was combined with Fourier - Transform Infrared (FT-IR) spectroscopy polymer identification and satellite-derived Suspended Particulate Matter (SPM) data, available for a subset of sites, to explore hydrodynamic influences on M

Simultaneous immune checkpoint blockade-associated diabetes mellitus and thyrotoxicosis.

Immune checkpoint inhibitors (ICIs) have become an increasingly prominent therapeutic option in the management of a variety of cancers. As their clinical use expands, so too should our understanding of immune-related adverse events (irAEs) associated with these agents. By enhancing immune activation, ICIs can cause off-target inflammation in various tissues, including endocrine. We present a case illustrating a rare but life-threatening manifestation of ICI toxicity: simultaneous severe diabetes mellitus and thyrotoxicosis after just a single dose of ICI. This case highlights the importance of

A new immunological paradigm for diseases: the protein-homeostasis-system hypothesis.

Contemporary immunology models are based primarily on reductionist concepts. However, they face limitations in reconciling clinical discordances such as the 'viral load paradox' in severe infections and the teleological enigma of the 'autoimmune paradox'. To bridge these systemic conceptual gaps, this paper proposes the protein-homeostasis-system (PHS) hypothesis, a novel paradigm based on organicism that views the living organism as a unified, self-regulating biosystem. It is asserted that all diseases, including autoimmune disorders and malignancy, are triggered by specific etiological subst

Nebulized Polygonum cuspidatum-derived nanovesicles loaded with dexamethasone synergistically ameliorate acute lung injury via metabolic-immune reprogramming.

BACKGROUND: Systemic glucocorticoid therapy for acute lung injury (ALI) is frequently limited by poor pulmonary targeting and severe systemic side effects. To address these challenges, we developed a bio-inspired, inhalable nanoplatform using naturally derived nanovesicles from the medicinal plant Polygonum cuspidatum (PcDNs) to actively deliver dexamethasone (PcDNs@Dex) via nebulization. METHODS: We assessed the post-nebulization stability of the nanosystem and evaluated its therapeutic efficacy in vitro and in vivo. In vitro, we investigated macrophage polarization (M1/M2) and the associate

Desert dust storm, ambient temperature, and pulmonary function among children with asthma in the Eastern Mediterranean.

BACKGROUND: Climate change has contributed to increased dust events and rising ambient temperature, posing potential health risk for susceptible populations including children with asthma. The study aims to examine associations between exposure to desert dust storm (DDS) and higher ambient temperature with pulmonary function indicators, including forced expiratory volume in 1s (FEV1), forced vital capacity (FVC) and FEV1/FVC% among asthmatic children during the dust season in the Eastern Mediterranean. METHODS: We conducted exploratory secondary analyses of repeated pulmonary function measure

Mechanical Thrombectomy Versus Anticoagulation Alone in Intermediate-High Risk PE: Improved Functional 90-Day Outcomes in STORM-PE RCT.

BACKGROUND: Up to 50% of pulmonary embolism (PE) survivors experience long-term functional limitations and reduced quality of life (QoL). Reperfusion strategies can rapidly relieve right-ventricular (RV) dysfunction and prevent clinical deterioration or hemodynamic collapse; however, these approaches have never been evaluated in a randomized trial against anticoagulation alone using patient-centered outcomes. OBJECTIVES: This study sought to evaluate whether mechanical thrombectomy with anticoagulation improves functional, QoL, and safety outcomes compared with anticoagulation alone in patien

A Health-Protective Screening Threshold for Personal Care Products: Metabolomic and Endocrine Mechanisms of Adolescent Reproductive Toxicity in Zebrafish.

Adolescents are uniquely vulnerable to endocrine-disrupting chemicals (EDCs) due to the critical maturation of the hypothalamic-pituitary-gonadal (HPG) axis, yet current safety assessments overlook whole-formulation risks during this developmental window. Using an adolescent zebrafish model, we established a biologically anchored screening threshold and elucidated the molecular mechanisms of formulation toxicity. Direct exposure to personal care products at unadjusted Scientific Committee on Consumer Safety (SCCS)-derived concentrations caused severe systemic overload (F1 survival ≤ 16.87%). H

Decoupling metabolic defense: Mutational and recombinational chaos in Sod1-deficient cells.

Superoxide dismutase 1 (Sod1) is long recognized as a frontline antioxidant, yet its broader role in safeguarding the eukaryotic genome remains under-explored. Here, using a yeast model, we reveal that Sod1 is an indispensable guardian of genomic integrity. Its absence unleashes a genomic storm: whole-genome sequencing uncovered a dramatic surge in single nucleotide variations (13-fold), loss of heterozygosity (12-fold), chromosomal rearrangements (4-fold), and a staggering 48-fold increase in aneuploidy. Remarkably, this genomic instability was completely abolished under anaerobic conditions,

DHODH is the key target of the KDM4 inhibitor QC6352 in glioblastoma stem cells.

Histone lysine demethylase 4 (KDM4) enzymes are frequently overexpressed in cancer and are considered to be promising therapeutic targets. QC6352, which is widely used as a best-in-class KDM4 chemical probe, shows potent antitumor activity across cancer models. Using comprehensive chemical biology approaches in glioblastoma models, we demonstrate that the efficacy of QC6352 and that of its clinical analog zavondemstat are independent of KDM4 inhibition. Instead, we identify dihydroorotate dehydrogenase (DHODH), a key enzyme in de novo pyrimidine biosynthesis, as the functional target driving t

Generation of DFNA5-deficient chickens reveals a species-specific role of pyroptosis in avian antiviral immunity.

The global burden of viral diseases in both humans and agricultural animals is exacerbated by dysregulated inflammatory cell death and cytokine storms. Pyroptosis, executed by gasdermin family proteins, is a key driver of this pathology, yet its functional conservation and therapeutic targeting in avian species remain critically unexplored. Here, we establish the first germline-edited heterozygous DFNA5-mutant chicken model using primordial germ cell mediated genome editing. We demonstrate that genetic disruption of DFNA5 markedly attenuates Raptinal-induced pyroptosis and reduces IL-1β and IL

β-Lactam antibiotics trigger mouse TLR2/4- and human TLR2/5-dependent Jarisch-Herxheimer reaction in leptospirosis.

Leptospirosis is a neglected zoonotic disease causing ~1 million cases and 60,000 deaths annually. Antibiotic therapy can trigger a detrimental inflammatory Jarisch-Herxheimer reaction (JHR). We investigated antibiotic effects in leptospirosis, using a mouse model of severe disease and human whole blood from healthy donors infected ex vivo with bioluminescent Leptospira interrogans. We tested the effect of amoxicillin and ceftriaxone, two bactericidal β-Lactams antibiotics and assessed bacterial survival, cytokines, pathophysiology and JHR mechanisms upon amoxicillin. β-lactams induced profoun

Novel (Azolo)Pyrimidine Derivatives as Dual Modulators of NO and IL-6 in Murine Macrophages and Human Monocytes.

Small molecules that simultaneously suppress nitric oxide (NO) and interleukin‑6 (IL‑6) offer a strategy to alleviate pathological inflammation. Building on our previous work with aryl‑substituted azaheterocycles, we synthesized 33 new pyrimidine and azolopyrimidine derivatives bearing amino, amide, hydroxyl, or halide groups to enhance hydrogen‑bonding capacity and improve inhibitory activity against both mediators. Screening in C57BL/6J mouse peritoneal macrophages identified nine compounds that markedly inhibited NO production at 50 µM. Two leads-triazolo[1,5‑a]pyrimidine 12bc and dihydropy

[A case of paroxysmal sympathetic hyperactivity following hypoxic brain injury with ‍difficult treatment].

The patient was a woman in her 20s. In June of a certain year, she experienced cardiopulmonary arrest due to acute epiglottitis and was admitted to the ICU and required mechanical ventilation. From the first day of hospitalization, she exhibited generalized seizures and was diagnosed with acute symptomatic seizures following hypoxic brain injury. Treatment with diazepam and levetiracetam was initiated, leading to symptom resolution. However, around the seventh day of hospitalization, she began experiencing episodic limb muscle hypertonia and fever. Based on diagnostic criteria, she was diagnos

Mitral valve regurgitation and emotional dysregulation secondary to long-standing Graves' thyrotoxicosis.

SUMMARY: Graves' disease is common, with an estimated prevalence of 0.2-2% and multiple associated complications. We present a case of severe mitral regurgitation secondary to poorly controlled Graves' disease for 11 years, highlighting the need for definitive treatment. A 30-year-old female attended hospital with palpitations, agitation and generalised pain. Her past medical history included Graves' disease diagnosed in 2014, with poor compliance and engagement with secondary care and medical treatment. Biochemical investigations and clinical examination confirmed thyrotoxicosis (TSH < 0.05 m
