Thyroid hormone excess targeted to oxytocin neurons disrupts oxytocin physiology without impacting behavior.

Deficiencies in oxytocin are associated with neurodevelopmental and behavioral disorders, and the oxytocin gene (Oxt) can be regulated by thyroid hormone. We have previously shown that a systemic excess of thyroid hormone during development causes deficits in oxytocin that are associated with major abnormalities in mood-related and social behaviors. To determine whether the disruption in oxytocin physiology is responsible for these aberrant behaviors we targeted thyroid hormone excess to oxytocin neurons using a mouse model of conditional deficiency of the type 3 deiodinase (DIO3), the neurona

Increased Risk of Thyrotoxicosis After COVID-19 in Adolescents and Young Adults During the Omicron-Predominant Period: A Nationwide Matched Study.

OBJECTIVE: Thyroid dysfunction has been increasingly reported after the coronavirus disease 2019 (COVID-19) pandemic. To date, no population-based study has examined this association using more than 10 million matched pairs. We compared the risk of documented thyroid outcomes after COVID-19 with that after influenza during the Omicron-predominant period. METHODS: We conducted a nationwide retrospective cohort study using the Japanese National Database of Health Insurance Claims, including patients with COVID-19 between January 2022 and March 2023, and those with influenza between September 20

A Case of Acute Liver Failure Secondary to Thyrotoxicosis.

Thyroid storm is a rare, life-threatening endocrine emergency that can cause multiorgan dysfunction, although acute liver failure (ALF) remains an uncommon manifestation. A 52-year-old woman with Graves' disease who had discontinued methimazole presented with three days of nausea, vomiting, diarrhea, melena, generalized weakness, and chest pain. She was lethargic and tachycardic, with a diffusely enlarged thyroid gland, warm skin, bilateral hand tremors, and hypoglycemia. Laboratory testing demonstrated severe thyrotoxicosis, marked hepatocellular injury with an aspartate aminotransferase conc

Association of Thyroid-stimulating Immunoglobulin in Early Pregnancy with Adverse Obstetric Outcomes: A Retrospective Cohort Study.

INTRODUCTION: The prevalence of TSI positivity in pregnant women and its association with pregnancy complications remain unclear. METHODS: This study evaluated 3090 pregnant women with stored serum samples during early pregnancy at Peking University First Hospital from October 2017 to August 2018. The risks of maternal and fetal adverse outcomes were compared across three groups: group A (TSI < 0.1 IU/L), group B (0.1≤ TSI < 0.55 IU/L), and group C (TSI ≥ 0.55 IU/L), using logistic regression models. RESULTS: The positivity rate of TSI (≥0.55 IU/L) was 1.33%, whereas 96.31% of women had TSI

Simultaneous immune checkpoint blockade-associated diabetes mellitus and thyrotoxicosis.

Immune checkpoint inhibitors (ICIs) have become an increasingly prominent therapeutic option in the management of a variety of cancers. As their clinical use expands, so too should our understanding of immune-related adverse events (irAEs) associated with these agents. By enhancing immune activation, ICIs can cause off-target inflammation in various tissues, including endocrine. We present a case illustrating a rare but life-threatening manifestation of ICI toxicity: simultaneous severe diabetes mellitus and thyrotoxicosis after just a single dose of ICI. This case highlights the importance of

Mitral valve regurgitation and emotional dysregulation secondary to long-standing Graves' thyrotoxicosis.

SUMMARY: Graves' disease is common, with an estimated prevalence of 0.2-2% and multiple associated complications. We present a case of severe mitral regurgitation secondary to poorly controlled Graves' disease for 11 years, highlighting the need for definitive treatment. A 30-year-old female attended hospital with palpitations, agitation and generalised pain. Her past medical history included Graves' disease diagnosed in 2014, with poor compliance and engagement with secondary care and medical treatment. Biochemical investigations and clinical examination confirmed thyrotoxicosis (TSH < 0.05 m

Refractory amiodarone-induced thyrotoxicosis causing malignant arrhythmias, treated with therapeutic plasma exchange-bridged thyroidectomy.

Amiodarone-induced thyrotoxicosis (AIT) is a potentially life-threatening complication of amiodarone therapy, particularly hazardous in patients with underlying structural heart disease. We report the case of a man in his mid-60s with severe cardiomyopathy and recurrent ventricular tachycardia who developed refractory thyrotoxicosis following amiodarone exposure. Overlapping features of type 1 and 2 AIT suggested mixed-type disease, posing a significant diagnostic challenge. Despite aggressive conventional therapy with high-dose thionamides and glucocorticoids, supplemented by adjunctive agent

Traditional Chinese Medicine for Graves' Disease: Clinical Efficacy, Pharmacological Mechanisms, and Challenges.

ETHNOPHARMACOLOGICAL RELEVANCE: Graves' disease (GD) is a common autoimmune disorder affecting the thyroid gland and is the most common cause of hyperthyroidism and thyrotoxicosis. Given the limitations of current conventional antithyroid therapy for GD, including side effects and relapse concerns, exploring alternative therapeutic options, such as Traditional Chinese Medicine (TCM), is of clinical interest. TCM, with its advantages of multi-component, multi-target actions and relatively mild side effects, has become a promising alternative therapy for GD. AIM OF THE STUDY: This review aims t

Thyrotoxicosis-Associated Cholestatic Jaundice in a Patient With Incidental Gallstone Disease: A Case Report and Literature Review.

BACKGROUND Thyrotoxicosis is associated with well-recognized and clinically significant complications, including congestive heart failure and cardiac arrhythmias. The relationship between hyperthyroidism and hepatic dysfunction is complex, with several proposed pathophysiological mechanisms; however, direct cholestatic injury is frequently overlooked. Although mild liver enzyme elevation is common in hyperthyroidism, clinically overt jaundice is rare. Nevertheless, increasing evidence from case reports and case series suggests that thyrotoxicosis, especially Graves disease, can result in chole

Case Report: Intracranial hypertension occurring in association with subacute thyroiditis.

Subacute thyroiditis (SAT) is a self-limited inflammatory thyroid disorder that typically produces a transient thyrotoxic phase. Intracranial hypertension (IH) is a rare but potentially sight-threatening complication of thyrotoxicosis, reported almost exclusively in hyperthyroidism, with only a single previous report linking it to SAT. We report the case of a 29-year-old woman who developed IH during the thyrotoxic phase of subacute thyroiditis, temporally related to dental manipulation. She presented with headache, nausea, vomiting, and fever, followed by a syncopal episode; clinical, laborat

From storm to stability: The role of plasmapheresis in the stabilization of patients with high-risk thyrotoxicosis.

INTRODUCTION: Thyrotoxicosis in high-risk patients is a severe condition requiring prompt management, although conventional therapeutic approaches often present limitations. Therapeutic plasmapheresis has emerged as a potential option to achieve rapid clinical stabilization and facilitate a safe transition to definitive treatment by effectively reducing circulating thyroid hormone levels. The objective of this study was to evaluate the safety and efficacy profile of plasmapheresis in patients with high-risk thyrotoxicosis. MATERIALS AND METHODS: We conducted a retrospective multicenter study

Syzygium aromaticum leaf extract promoted biogenic CuO nanoparticles for the anti-apoptotic efficacies of NCI-H661 human lung adenocarcinoma cell line and following the PI3K/AKT/mTOR signaling pathway.

BACKGROUND: A lower rate of permanent pacemaker (PPM) has been linked to a target aortic implantation height (AIH) >0.70, following transcatheter aortic valve replacement (TAVR) with the SAPIEN 3 valve. Based on clinical experience, it was hypothesized that a higher AIH (≥0.85) would lower the rate of PPM implantation. METHODS: A total of 127 patients (66 females, 61 males; mean age 82 ± 8 years) underwent TAVR with the SAPIEN 3 valve between May 2015 and July 2016. AIH was defined as the proportion of the valve frame above the aortic annulus in the post-deployment aortogram. A target AIH (≥0

Finite-time synchronization of quaternion-valued fuzzy memristive neural networks by an non-separation method via event-triggered control.

In this paper, the event-triggered control method is applied to study a class of generalized delayed quaternion-valued fuzzy Memristive Neural Networks. By constructing a novel Lyapunov function and designing an appropriate event-triggered controller, new event-triggered conditions and synchronization criteria are established. To handle the inherent complexity of quaternion operations, a nonlinear scaling technique is employed to compare distinct quaternion components, while an improved quaternion norm together with a symbolic function enables a holistic analysis of the quaternion-valued fuzzy

Traditional Chinese Medicine as an Adjunct in Hyperthyroidism Management: A Narrative Review of Pathophysiology, Proposed Mechanisms, and Clinical Evidence.

Hyperthyroidism, characterized by elevated serum thyroxine levels, can be classified into overt and subclinical forms. Untreated hyperthyroidism poses substantial health risks. Antithyroid drugs, radioactive iodine, and surgery are established conventional therapeutic options; however, their clinical application may be limited by relapse, adverse effects, contraindications, or patient preference. This narrative review examines the epidemiology of hyperthyroidism, particularly its association with iodine status, strong genetic predisposition, clinical manifestations predominantly seen in women

Peripheral manifestations of monocarboxylate transporter 8 deficiency: hepatic and skeletal muscle involvement.

Monocarboxylate transporter 8 (MCT8) deficiency, also known as Allan-Herndon-Dudley syndrome, is a rare X-linked disorder caused by pathogenic variants in the SLC16A2 gene. It leads to impaired transport of thyroid hormones, particularly triiodothyronine (T3) and thyroxine (T4), across the blood-brain barrier and into neural cells. Consequently, the brain experiences reduced thyroid hormone availability, resulting in central hypothyroidism and severe neurodevelopmental impairment. In contrast, peripheral tissues remain exposed to elevated circulating T3, leading to chronic peripheral thyrotoxi
