BRAFV600E-Informed TI-RADS Reclassification of Biopsy Eligibility in Thyroid Nodules: A Retrospective Oracle Decision-Simulation Study.

Background/Objectives: We evaluated a hypothetical oracle strategy that assumes BRAFV600E status is known before fine-needle aspiration (FNA), although testing usually requires FNA-derived material. Methods: This retrospective simulation included 4944 nodules from 4560 patients with linked ultrasound and molecular records following FNA evaluation. The cohort was selected for cytology and molecular testing and enriched for BRAFV600E positivity (42.4%) and TI-RADS 4C nodules (66.7%). The predefined rule combined BRAFV600E-positive upgrading with one-level downgrading of negative TI-RADS 4A/4B no

Causal effects of antihypertensive drugs on thyroid diseases: A trans-ancestry drug-target Mendelian randomization study.

Thyroid disorders frequently coexist with cardiovascular disease, including hypertension, but whether long-term modulation of antihypertensive drug targets influences thyroid disease risk remains uncertain. We aimed to evaluate the associations of 7 antihypertensive drug-target classes with thyroid diseases in European and East Asian populations using a trans-ancestry drug-target Mendelian randomization (MR) framework. We performed a two-sample, trans-ancestry drug-target MR analysis of 7 antihypertensive target classes. Target-proximal single-nucleotide polymorphisms associated with systolic

LncRNA AP003469.2 promotes hepatocellular carcinoma progression by regulating cuproptosis via the miR-4732-3p/LIPT1 axis.

BACKGROUND: Hepatocellular carcinoma (HCC) is a malignant tumor characterized by high recurrence and metastasis rates, as well as a poor prognosis. Cuproptosis is a form of programmed cell death driven by abnormal copper-ion metabolism. Existing studies have shown that its occurrence is closely regulated and associated with the progression of HCC. However, it is still unclear how long non-coding RNAs (lncRNAs) control the process in HCC. The purpose of this work is to clarify how the lncRNA AP003469.2 influences cuproptosis and promotes the development of HCC. METHODS: This study utilized RNA

A novel framework for identifying cellular signature LncRNAs and their functional coregulatory networks with prognostic implications at single-cell resolution.

INTRODUCTION: Long non-coding RNAs have been implicated in crucial roles in developing malignant tumors. However, comprehensive analyses of lncRNA within the tumor microenvironment, particularly at the single-cell level across various tumor types, remain limited. OBJECTIVES: This study aimed to establish a comprehensive methodological framework for prioritizing and identifying novel cancer-associated lncRNA biomarkers and potential therapeutic targets at the single-cell level, and to integrate these findings into a unified platform. METHODS: We developed SIGLNCOS, a computational framework f

Association of ABO and Rh blood groups with clinicopathological features and lymph node involvement in surgically treated thyroid malignancies: a retrospective single-center study.

This study investigated the association between ABO and Rh blood group antigens and the clinicopathological features of 418 patients who underwent thyroidectomy for primary thyroid malignancy between December 2016 and June 2023. Since continuous variables did not follow a normal distribution (p < 0.05), data were presented as median and interquartile range (IQR). The distribution of ABO blood groups was A (44.5%), O (31.8%), B (16.0%), and AB (7.7%), with 90.9% of patients being Rh-positive. The median age of the cohort was 48 years (IQR: 19). While ABO groups showed no significant difference

An integrated single-cell transcriptomic and microbiome dataset of orbital adipose tissue in thyroid eye disease.

Thyroid eye disease (TED), also historically referred to as thyroid-associated ophthalmopathy (TAO) or Graves' orbitopathy (GO), is an autoimmune ophthalmopathy characterized by inflammation and adipogenesis within orbital adipose (OA) tissue. While the contribution of immune cell infiltration and fibroblast activation is recognized, the cellular heterogeneity and the potential role of local microbial dysbiosis in the orbital microenvironment remain poorly defined. Here, we present a multi-omics dataset derived from OA of patients with TED and healthy controls collected at Beijing Tongren Hosp

Machine learning and penalized logistic regression-based models for the identification of potential characteristic genes in primary hyperparathyroidism.

PURPOSE: Primary hyperparathyroidism (PHPT) is an endocrine disorder in which one or more parathyroid glands oversecrete parathyroid hormone, leading to hypercalcemia and potential complications such as kidney stones, bone fractures, and other systemic issues. Machine learning (ML) is extensively applied to diagnose medical diseases due to its high accuracy and ease of implementation. METHODS: The microarray expression dataset (GSE83421) was selected from the Gene Expression Omnibus Database (GEO) for analysis. Differential expression analysis was performed with false discovery rate (FDR) cor

Cardiovascular risk factors in autoimmune thyroiditis: role of adiponectin, resistin, inflammation and auto-immunity - a cross-sectional study.

OBJECTIVE: Autoimmune thyroiditis (AIT) has been associated with an increased cardiovascular risk. Whether adipokines, inflammation and auto-immunity contribute to this increased risk is uncertain. Our aim was to evaluate the interrelationships of adipokines, inflammation, auto-immunity, and thyroid function with cardiovascular risk in autoimmune thyroiditis. METHODS: Sixty-two patients with AIT, with normal free T4 (FT4) and free T3 (FT3), without other comorbidities were studied. Patients were divided into two groups: AIT with thyroid-stimulating hormone (TSH) ≤2.5 mU/L and AIT with TSH >2.

The role of microbiota on thyroid cancer: from carcinogenicity, treatment and predictability.

In recent years, more and more attention has been paid to the mechanism of microbiota in thyroid cancer (TC). Gut microbiota (GM) and its metabolites can regulate the immune system and affect thyroid function through the gut-thyroid axis. When the GM is dysregulated, the intestinal barrier function is impaired, resulting in immune dysfunction, chronic inflammation, oncogene activation, and eventually leading to the occurrence of TC. In addition, intratumoral microbiota (ITM) is also directly involved in the occurrence, progression and invasion of TC. However, the mechanism of action between th

How I Do It: An Imaging Approach and Review of Autoimmune Encephalitis.

Encephalitis is a medical emergency characterized by inflammation of the brain parenchyma due to an infective cause or an immune-mediated process. Autoimmune encephalitis (AE) is characterized by a misdirected immune attack against neuronal structures, resulting in an inflammatory response. Triggers for the development of antibodies are often paraneoplastic or parainfectious. AE may occur in children or adults with symptoms including subacute onset of altered mentation, cognitive issues, drug-resistant epilepsy, movement disorders, and/or autonomic dysfunction. Limbic encephalitis is a common

IL1RAP promotes EMT and metastatic phenotypes in papillary thyroid carcinoma and is associated with PI3K/AKT signaling activation.

Although papillary thyroid carcinoma (PTC) usually follows an indolent course, lymph node metastasis and locally advanced disease remain clinically important challenges. This study evaluated interleukin-1 receptor accessory protein (IL1RAP) in PTC using pan-cancer datasets, TCGA/GEO thyroid cancer cohorts, and in vitro validation. IL1RAP was upregulated in PTC tissues and cell lines, and higher IL1RAP expression was associated with advanced pathologic stage, lymph node metastasis, extrathyroidal extension, and unfavorable prognostic indicators. Functional assays showed that IL1RAP overexpressi

Thyroid and Parathyroid Dysfunction Following Larynx-Trachea-Thyroid-Parathyroid Allotransplantation.

Larynx-trachea-thyroid-parathyroid allotransplantation can restore essential laryngeal functions, but postoperative endocrine changes remain poorly understood. This study assessed thyroid and parathyroid function in four recipients. All developed early thyroid hormone alterations consistent with euthyroid sick syndrome, followed by variable endocrine outcomes. Two experienced thyrotoxicosis associated temporally with graft rejection, while another developed hypothyroidism and elevated parathyroid hormone during late rejection. Transient parathyroid hormone suppression was also observed. In con

Psychosocial stress and autoimmune thyroid disease: a systematic review of clinical phenotypes and vulnerability factors.

Psychological stress has been proposed as a potential trigger or modulator of autoimmune thyroid diseases, but evidence remains fragmented. This systematic review examined the relationship between stress and thyroid autoimmunity, with particular attention to Hashimoto's thyroiditis and Graves' disease, highlighting the critical methodological distinction between stressor exposure (objective events) and the subjective stress response (psychological burden). PubMed, Embase, Scopus, and PsycINFO were searched for controlled human studies; 25 studies involving a total of N = 117,034 participants m

The dual IGF-1R/IR blocker, Linsitinib, modulates human orbital fibroblast activity and proliferation.

AIM: The insulin-like growth factor-1 receptor (IGF-1R) and its downstream signaling pathways drive proliferation, survival, and extracellular matrix production, representing key therapeutic targets in thyroid eye disease (TED). This novel in vitro study investigated the inhibitory effects of the dual IGF-1R/insulin receptor small-molecule inhibitor Linsitinib on orbital target cell activity, proliferation, and survival. METHODS: Human orbital fibroblasts (OF) were pre-treated with increasing doses of Linsitinib and stimulated with 100 ng/mL IGF-1 to quantify hyaluronic acid (HA) and pro-coll

Sacral Neural Crest-Derived Extracellular Vesicles Promote Pelvic Ganglion Repair Through Anti-Pyroptotic Effects Involving ANXA1-FPR2 Signaling.

Pelvic autonomic nerve injury is a major cause of postoperative bowel dysfunction, yet effective strategies for restoring damaged pelvic ganglia remain lacking. Here, we demonstrate that extracellular vesicles derived from human pluripotent stem cell-derived sacral neural crest cells (Sacral NC-EVs) promote pelvic neuroregeneration by suppressing oxidative stress-induced pyroptosis. In a rat model of major pelvic ganglion (MPG) injury, local administration of Sacral NC-EVs improved gastrointestinal motility and partially restored rectal neural innervation and epithelial barrier integrity.
