Molecular mechanisms underlying the effectiveness of triiodothyroacetic acid (Triac) in patients with resistance to thyroid hormone β (RTHβ): a multicentre, international, retrospective cohort study.

BACKGROUND: Disease features in resistance to thyroid hormone beta (RTHβ), caused by THRB mutations, arise from insensitivity to thyroid hormone in tissues expressing thyroid hormone receptor (TR)β and from lifelong thyrotoxic effects in TRα1-expressing tissues, posing substantial cardiovascular morbidity/mortality. The T3-analogue triiodothyroacetic acid (Triac) holds potential as an effective treatment. Systematic data on Triac therapy in RTHβ are lacking, and the mechanisms through which Triac acts are unclear. This study aims to elucidate these knowledge gaps. METHODS: Clinical and bioche

Prediction of radiation-induced hypothyroidism after proton-based supraclavicular nodal irradiation in breast cancer.

BACKGROUND: The thyroid gland lies adjacent to the supraclavicular target and is highly radiosensitive, placing patients at risk for radiation-induced hypothyroidism (RIHT). While RIHT incidence and predictive models have been well described after photon-based irradiation, evidence in the proton therapy setting remains limited. PURPOSE: To evaluate the incidence of RIHT in breast cancer patients undergoing proton-based supraclavicular nodal irradiation and to develop proton-specific normal tissue complication probability (NTCP) models incorporating dose-volume, dose-averaged linear energy tra

[Successful treatment of myocardial infarction complicated with thyroid crisis using extracorporeal membrane oxygenation: a case report and literature review].

Thyroid crisis is caused by an excess of thyroid hormones in the body, leading to a hypermetabolic state and a series of pathological and physiological changes. Clinical manifestations are often non-specific, and in severe cases, it can manifest as multiple organ dysfunction. Acute myocardial infarction combined with thyroid crisis is dangerous and has a high mortality rate. Timely identification and combination of mechanical circulatory support and antithyroid drug treatment are key to improving prognosis. On January 28, 2021, a 49-year-old female patient was admitted to Yanbian University Af

Symptom Management Following Radioactive Iodine Treatment in Adults With Thyroid Cancer: Usability Evaluation and Proof-of-Concept Study.

BACKGROUND: Thyroid cancer is often labeled a "good cancer" due to high survival rates. However, many patients diagnosed with differentiated thyroid cancer (DTC) treated with radioactive iodine (RAI) experience short- and long-term symptoms as a result of treatment. These include salivary dysfunction, dry mouth, dry eyes, and nausea. Despite these challenges, patients report limited knowledge and unmet informational needs regarding RAI symptoms and symptom management. Evidence-based, patient-centered education to support symptom management during DTC survivorship is needed. Our team developed

Prediction of gross extrathyroidal extension in papillary thyroid carcinoma using a deep learning radiomics model based on post-processing techniques for multi-phase contrast-enhanced MRI images.

BACKGROUND: Gross extrathyroidal extension (ETE) affects surgical management and prognosis in papillary thyroid carcinoma (PTC), yet noninvasive assessment is difficult. METHODS: In this retrospective exploratory feasibility study, 101 patients with pathologically confirmed PTC who underwent multi-phase contrast-enhanced MRI (MCE MRI) were analyzed. From the second and seventh contrast phases, four derived image types (ADD, ADD/SUB, Max, Min) were generated. Deep features from six image modalities were extracted using fine-tuned ResNet101, and radiomics classifiers were trained to predict gro

Management of pregnancy in patients with Graves' disease secondary to alemtuzumab: a case series and literature review.

INTRODUCTION: Alemtuzumab treatment for multiple sclerosis frequently induces Graves' disease (GD), characterized by common coexistence of stimulating and blocking anti-thyrotropin receptor autoantibodies (TRAb), causing fluctuations of thyroid function. Alemtuzumab-induced TRAb-positive hypothyroidism is also common. These conditions are challenging to manage during pregnancy, considering the imperative necessity to maintain euthyroidism to minimize risks for both pregnancy and fetal/neonatal outcomes. METHODS: We reviewed the clinical course and therapeutic management of four pregnant women

Exceptional and Durable Response to Selpercatinib in a Patient with Apparently RET-Negative, HRAS-, and NF1-Mutated Metastatic Medullary Thyroid Carcinoma.

BACKGROUND: Selpercatinib is approved for RET-mutated medullary thyroid carcinoma (MTC); its activity in tumors without detectable RET alterations is uncertain. PATIENT FINDINGS: A 50-year-old woman developed rapidly progressive MTC (Ki-67 = 30%) with extensive liver and bone metastases. Because of severe symptomatic progression, selpercatinib was started urgently, off-label, before molecular testing results were available. She had rapid clinical improvement, marked calcitonin and CEA decline, and a remarkable and durable radiographic partial response until the last follow-up (29 months). SU

Metabolic syndrome and incident thyroid cancer risk in adults: A systematic review and meta-analysis.

The global incidence of thyroid cancer (TC) has increased, with metabolic syndrome (MetS) identified as a potential modifiable risk factor. However, epidemiological evidence regarding this association remains inconsistent. We conducted a meta-analysis to quantify the relationship between MetS and the risk of incident TC in adult populations. We searched PubMed, Embase, and Web of Science for longitudinal observational studies assessing MetS in relation to incident TC. Risk ratios (RRs) with 95% confidence intervals (CIs) were pooled using random-effects models to account for heterogeneity. Our

Label-Free Detection of Thyroid Cancer Biomarkers Using Heterojunction GAA Ferroelectric p-n-i-n TFET Biosensor.

The global rise in thyroid cancer incidence has intensified the demand for reliable, rapid and minimally invasive diagnostic tools as current clinical practice often result in unnecessary surgical procedures for benign thyroid nodules. In this context, this work proposes a highly sensitive label-free biosensor, based on a heterojunction gate-all-around ferroelectric p-n-i-n tunnel field effect transistor (HJ-GAA-Fe p-n-i-n TFET) for early thyroid cancer detection. The sensing mechanism leverages variations of the dielectric properties and charge states of thyroid cells, which modulates electro
