Investigating the impact of intestinal glucagon-like peptide-1 on hypoglycemia in type 1 diabetes.

Recent advances in understanding type 1 diabetes (T1D) highlight the complexity of managing hypoglycemia, a frequent and perilous complication of diabetes therapy. This letter delves into a novel study by Jin et al, which elucidates the role of intestinal glucagon-like peptide-1 (GLP-1) in the counterregulatory response to hypoglycemia in T1D models. The study employed immunofluorescence, Western blotting, and enzyme-linked immunosorbent assay to track changes in GLP-1 and its receptor expression in diabetic mice subjected to recurrent hypoglycemic episodes. Findings indicate a significant inc

Low-carbohydrate diet in type 1 diabetes: A systematic review.

BACKGROUND: The treatment of type 1 diabetes mellitus (T1D) consists of applying insulins with different action profiles and ingesting known amounts of nutrients from the diet. Although there is no specific recommendation regarding the distribution of these macronutrients, low-carbohydrate (LC) diets seem to show promise in some types of diabetes, but their role in T1D is still controversial, given the potential glycemic improvement, but with the concurrent possibility of changes in other parameters such as lipids. This systematic review aimed to evaluate the metabolic effects of these diets o

Integrating machine learning and single-cell sequencing to identify shared biomarkers in type 1 diabetes mellitus and clear cell renal cell carcinoma.

PURPOSE: Type 1 diabetes mellitus (T1DM), as an autoimmune disease, can increase susceptibility to clear cell renal cell carcinoma (ccRCC) due to its proinflammatory effects. ccRCC is characterized by its subtle onset and unfavorable prognosis. Thus, the aim of this study was to highlight prevention and early detection opportunities in high-risk populations by identifying common biomarkers for T1DM and ccRCC. METHODS: Based on multiple publicly available datasets, WGCNA was applied to identify gene modules closely associated with T1DM, which were then integrated with prognostic DEGs in ccRCC.

Tregs in pregnancy with type 1 diabetes mellitus: pilot study.

OBJECTIVE: Pregnancy in women with type 1 diabetes mellitus has been associated with adverse outcomes due to persistent hyperglycemia and impaired maternal-fetal interactions. Regulatory T cells seem to exert a critical role in this process. Pregnancy can change the profile of Treg cells and affect the outcome of pregnancy; therefore, our purpose was to characterize the profile of regulatory T cells in the peripheral blood of pregnant and nonpregnant (controls) women with type 1 diabetes mellitus. METHODS: This prospective case-control study recruited 36 women with type 1 diabetes mellitus, 1

Single sural nerve response: A reliable and practical method for diagnosis of diabetic peripheral neuropathy in children with type 1 diabetes.

OBJECTIVES: Type 1 diabetes mellitus (T1DM) is a significant global health issue, particularly due to its association with microvascular complications such as diabetic peripheral neuropathy (DPN). Sensory nerves in the lower extremities are primarily affected by DPN, with the sural nerve being particularly impacted. The conventional method for diagnosing DPN involves evaluating four motor and four sensory nerves in the upper and lower extremities. Motor tests use dual-point high-intensity stimulation to elicit a compound muscle action potential, while sensory tests apply a single, lower-intens

Screening for autoimmune atrophic gastritis by serum gastrin measurement in subjects with type 1 diabetes.

INTRODUCTION: Despite associated risk of anemia and gastric cancer, screening for autoimmune atrophic gastritis (AAG) is underperformed in subjects with type 1 diabetes mellitus (T1DM). We measured the predictive value of serum gastrin as a biomarker of gastric atrophy in subjects with T1DM and parietal cell autoantibodies (PCA). SUBJECTS AND METHODS: PCA measurements were retrospectively retrieved in 1,425 consecutive subjects with T1DM between 2014 and 2018. Screening for AAG was conducted in PCA+ subjects by measuring blood counts, serum ferritin, vitamin B12 and gastrin; and by performing

Nanocellulose dysregulated glucose homeostasis in female mice on a Western diet: The role of gut microbiome.

There is currently increased interest in nanocellulose as a food emulsifier and dietary supplement. It was hypothesized that nanocellulose could modulate behaviors and glucose homeostasis in female mice using mechanisms of altered gut microbiome and immune modulation. An initial experiment was conducted with the objective of examining whether three common types of nanocellulose affected the gut microbiome of female C57BL/6 mice on a Western diet. Cellulose nanofibrils (CNF), TEMPO-CNF and cellulose nanocrystals were administered at the physiologically relevant dose of 30 mg/kg/day for 30 days

Recent advances in stem cell-based therapies for type 1 diabetes: A glimpse into the future.

Type 1 diabetes mellitus (T1DM) is a serious, chronic metabolic and autoimmune disease that affects millions globally. While insulin administration remains the most effective treatment, it is not a cure. Long-term therapies, such as immunotherapy, can be effective for some patients, but they have notable limitations and do not provide a permanent solution. As a result, current research has shifted towards stem cell-based therapies, which offer a potentially expandable and scalable source of pancreatic beta cells. These therapies aim to restore long-term endogenous β-cell function in all T1DM p

Implication of the HLA-DQA1, HLA-DQB1 and CTLA-4 alleles in the susceptibility to type 1 diabetes in Jordanian population.

BACKGROUND: Type 1 diabetes mellitus (T1D) is a chronic autoimmune disease caused by the selective destruction of pancreatic beta cells, leading to insulin deficiency. Both genetic and environmental factors contribute to disease susceptibility. Among genetic factors, human leukocyte antigen (HLA) class II molecules, particulary DQA1 and DQB1 haplotypes, have been associated with T1D risk. This study aimed to identify haplotypes that increase susceptibility to or provide protection against T1D in Jordanian population. METHODS: A total of 200 healthy individuals and 200 T1D patients were includ

Reversing Neuronal Klotho Dysfunction-Mediated Diabetic Neuropathy Through 16:8 Intermittent Fasting.

Insulin supply is the golden standard for type 1 diabetes mellitus (T1DM) therapy. Is there a drug-reduction application for reversing glucose metabolism disabled and diabetic neuropathy (DN), and is it suitable for the young and elderly populations? Reducing T1DM-associated DN, and maintaining glucose metabolism require using the anti-aging gene Klotho to regulate specific signaling cascades. This study applied five 16:8 intermittent fasting (16-h fasting, 8-h eating; 168if) protocols by different executing times to young and elderly diabetic mice to evaluate whether 168if is age-dependent an

Causal associations between gut microbiota and type 2 diabetes mellitus subtypes: a mendelian randomization analysis.

PURPOSE: To investigate the causal relationships between gut microbiota and novel adult-onset type 2 diabetes mellitus(T2DM) subtypes. METHODS: We conducted Mendelian randomization (MR) analyses using genome-wide association data from European populations. Initial MR analyses examined associations between gut microbiota and four T2DM subtypes, followed by validation analyses using type 1 diabetes mellitus(T1DM) and T2DM GWAS data. We also performed bidirectional MR analyses and tested for heterogeneity and pleiotropy across all analyses. RESULTS: Our MR analyses revealed distinctive associat

A cost-utility analysis of long-acting insulin analogues (detemir, glargine and degludec) for the treatment of adult type 1 diabetes in South Africa.

BACKGROUND: Type 1 Diabetes Mellitus (T1DM) is a life-threatening condition that is managed with administered insulin. Intermediate- to long-acting insulin represents the basal insulin constituent of the total insulin used in treating T1DM. In South Africa, intermediate-acting Neutral Protamine Hagedorn (NPH) insulin has been the mainstay basal insulin recommended in the public sector, despite the availability of newer (ultra) long-acting insulin analogues. A cost-utility analysis of the newer long-acting insulin analogues insulins degludec, glargine U100, glargine U300 and detemir in comparis
