Gamma-Oryzanol Solid Dispersion Attenuates Dexamethasone-Induced Depressive-like Behaviors in Rats Associated with Calbindin-1 and Calcium Signaling.

Depression is associated with dysfunction of the hypothalamic-pituitary-adrenal axis, neurotransmitter imbalance, and calcium signaling deficits. Although gamma-oryzanol (GO) has shown antidepressant effects, its underlying molecular mechanism remains elusive, while its poor aqueous solubility limits its nutraceutical application. To overcome this obstacle, we successfully developed GO solid dispersion (GOSD) with enhanced water solubility for nutraceutical formulations. This study examined the antidepressant effect of GOSD in dexamethasone (Dx)-induced depressive-like behaviors in rats using

Behind the Sella: A Rare Case of Pituicytoma Mimicking a Pituitary Adenoma.

BACKGROUND Pituicytoma is a rare central nervous system (CNS) World Health Organization (WHO) grade I glioma originating from pituicytes, the specialized glial cells of the neurohypophysis or infundibulum. It accounts for less than 0.1% of all sellar and suprasellar masses and often presents a diagnostic challenge because of its clinical and radiological resemblance to other pituitary lesions. CASE REPORT A 48-year-old man was admitted to the endocrinology clinic because of clinical symptoms of hypogonadism, headaches, and visual field defects. Hormonal evaluation revealed hypogonadotropic hyp

Neuroimmune and Neuroendocrine Mechanisms of the Brain-Heart Axis in Cardiovascular Diseases: A Review.

The brain-heart axis, a core bidirectional communication network linking the central nervous system and cardiovascular system, plays a pivotal role in the onset, progression, and prognosis of cardiovascular diseases when its regulatory homeostasis is disrupted. This review systematically examines the core regulatory pathways and molecular mechanisms of the brain-heart axis and highlights its pathological roles in major cardiovascular diseases, such as hypertension, coronary artery disease, heart failure, and arrhythmias. Particular emphasis is placed on recent advances in key pathways, includi

[Patients with papillary craniopharyngioma successfully treated with BRAF and MEK inhibitors].

Craniopharyngioma is a rare brain epithelial tumor infiltrating the hypothalamus causing visual loss, panhypopituitarism, cognitive deficits and obesity. 94% of papillary craniopharyngiomas harbour a BRAF mutation. Two patients with PCP and BRAF V600E mutation, with different tumor status, were treated with BRAF and MEK inhibitors. Case I is a male diagnosed with biopsy and treated for 16 months with BRAF and MEK inhibitors. After 12 months of treatment the tumor volume decreased from 2 220 mm3 to 90 mm3. The patient was treated with fractionated cranial irradiation (54 Gy) and has been withou

Exploring the role of histone deacetylases and histone deacetylase inhibitors in obesity and emerging evidence from Anorexia Nervosa.

Histone deacetylases (HDACs) are key epigenetic regulators that modulate chromatin accessibility and transcriptional programs controlling metabolism, energy balance, and reward-related behaviors. Aberrant HDACs activity contributes to metabolic dysfunction by altering adipogenesis, thermogenic capacity, lipid profile, and hypothalamic circuits governing appetite. In obesity, specific HDAC isoforms promote adipocyte hypertrophy, impair browning of white adipose tissue, disrupt mitochondrial function, and exacerbate inflammation and insulin resistance. HDAC inhibitors (HDACi) counteract these ef

Depression and bone loss: associations, skeletal features and mechanisms from clinical and preclinical studies.

Depression has been associated with negative skeletal outcomes, but the specific characteristics and underlying mechanisms remain incompletely understood. This narrative review summarizes clinical and preclinical studies in this field. Current findings suggest that depression may be associated with lower bone mineral density, altered bone remodeling and microarchitecture, and increased risks of falls and fractures. The magnitude and pattern of these associations may vary according to depression severity and chronicity, skeletal site, sex, and life stage. Experimental studies indicate that skel

The hypothalamic orexin-medial prefrontal cortex pathway modulates susceptibility to depression in the learned helplessness paradigm in male mice.

Depression is a prevalent psychiatric disorder primarily characterized by behavioral despair and helplessness. The orexinergic system plays a crucial role in stress-induced depression; however, the precise mechanisms remain unclear. The medial prefrontal cortex (mPFC), a major target of orexinergic projections, is essential for emotional regulation. This study suggests that impaired orexinergic input to the mPFC underlies depression-like behaviors in murine models. In the learned helplessness (LH) paradigm, the majority of male mice subjected to inescapable foot shocks developed depression-lik

Eating Behavior in Menopausal Women: From Molecular to Clinical Aspects-What Changes?

The menopausal transition represents a critical period in women's lives characterized by profound hormonal, metabolic, and behavioral changes. Estrogen deficiency during menopause triggers complex alterations in eating behavior through multiple molecular mechanisms involving hypothalamic neuropeptide systems, neurotransmitter pathways, and peripheral hormonal signals. This narrative review examines the molecular basis of eating behavior changes in menopausal women, integrating evidence from basic science and clinical research. At the molecular level, declining estradiol disrupts the hypothalam

Functional hypothalamic amenorrhea: A call for global awareness and research.

Functional hypothalamic amenorrhea (FHA) is a common yet under-recognized cause of secondary amenorrhea resulting from reversible suppression of the hypothalamic-pituitary-ovarian axis. Triggered by chronic stress, low energy availability, disordered eating, and/or excessive exercise, FHA leads not only to anovulation and hypoestrogenism but also to broader neuroendocrine disruption involving the hypothalamic-pituitary-adrenal and thyroid axes. Although historically associated with extreme trauma and the "female athlete triad," FHA occurs worldwide and remains significantly underdiagnosed acro

Investigating the effect of diet-induced prediabetes on the HPA-axis, sympathetic nervous system and endogenous opioids in male Sprague-Dawley rats.

BACKGROUND: Prediabetes is associated with metabolic dysfunction that may disrupt neuroendocrine regulation before the onset of overt type 2 diabetes mellitus. Although previous studies have independently examined alterations in the hypothalamic-pituitary-adrenal (HPA) axis, sympathetic nervous system (SNS), and endogenous opioid system during prediabetes, the temporal interactions among these systems during disease progression remain poorly understood. This study aimed to investigate changes in these interconnected neuroendocrine pathways in a diet-induced prediabetic rats. METHODS: Male Spr

The Many Faces of Acromegaly: Understanding Its Clinical and Biological Heterogeneity.

BACKGROUND: Acromegaly, in 95% of cases resulting from a PitNET that hypersecretes growth hormone (GH), is constantly revisited in the literature due to the repercussions on morbidity and mortality and difficulties that persist in early diagnosis. SUMMARY: To shorten the interval between the onset of acromegaly manifestations and the diagnosis, our goal was to summarize the main situations that deviate from the typical presentation of the disease, highlighting the variability in phenotypic features, biochemical findings, and GH tumor expression. These non-classic findings contribute to the di

Palatable-Food-Driven Top-Down Circuit Inhibits PVNCRF Activity to Mitigate Stress Via Peri-PVNCRFR1 Neurons.

Stress is a major precipitating factor for emotional disorders, including anxiety. To cope with stress, individuals frequently engage in hedonic behaviors, such as eating palatable food, which provide transient relief from psychological distress and may protect against the development of pathology. However, the neural mechanisms by which hedonic experience counteracts stress-induced anxiety remain poorly understood. Here, we identify a neural circuit functionally connecting the prefrontal cortex (PFC) to the paraventricular nucleus (PVN) of the hypothalamus that mediates stress mitigation thro

A MYCN-GAL-SREBP1 lipogenic axis drives proliferation in silent corticotroph adenomas.

BACKGROUND: Silent corticotroph adenomas (SCAs) are an aggressive pituitary neuroendocrine tumor (PitNET) subtype lacking effective medical therapies and showing a high rate of recurrence. The molecular mechanisms driving their proliferation remain poorly understood. Although metabolic reprogramming is a hallmark of cancer, neither lipid metabolism nor its regulatory pathways have been systematically investigated in SCAs. METHODS: We performed integrative analyses of bulk and single-cell RNA sequencing datasets from SCAs, functioning corticotroph adenomas (FCAs), and normal pituitary tissue t

Partial or full preservation of the pituitary stalk in craniopharyngioma surgery: improving endocrinological outcomes without an increased risk of recurrence.

OBJECTIVE: The risk-benefit balance of pituitary stalk (PS) preservation during craniopharyngioma (CP) surgery remains insufficiently investigated. Although preservation requires meticulous microsurgical dissection, frequent invasion of the PS by the CP also raises the question of the feasibility of preservation. Herein, the authors assessed the impact of preserving the PS in a large consecutive series of adult patients who underwent endoscopic endonasal surgery for CP. METHODS: The authors performed a retrospective single-center study of adult, surgery-naive patients with supradiaphragmatic
