Chronic hyperprolactinemia is associated with enhanced high-fat diet binge eating in female mice.

Prolactin is fundamental for pregnancy and lactation, and also has numerous metabolic roles including the stimulation of appetite. Mice with chronic hyperprolactinemia display increased food intake, enhanced expression of orexigenic genes in hypothalamic brain centers, and also develop obesity. Here, we tested whether the consumption of a rewarding high-fat diet (HFD) is altered in a mouse model of chronic hyperprolactinemia: LacDrd2KO mice, which lack dopamine Drd2 receptors in lactotrophs, preventing dopamine inhibition of prolactin secretion. We exposed control and LacDrd2KO mice to a binge

Restoration of ovarian endocrine function with encapsulated immune-isolated human ovarian xenograft in ovariectomized mice.

Anticancer treatments may cause depletion of the nonrenewable ovarian reserve of follicles, the source of key hormones, leading to premature ovarian insufficiency (POI) and substantial endocrine, musculoskeletal, and neurological complications. Hormone replacement therapy, the only approved treatment for POI, poorly mimics ovarian physiology. Here, we examined whether human ovarian tissue implanted in an immune-isolating hydrogel-based capsule restored ovarian endocrine function in ovariectomized immunodeficient mice. Daily vaginal cytology, hormone measurements, and histological analysis demo

Consensus on the Diagnosis of Cushing's Disease: A Collaborative Statement from the Korean Endocrine Society and Japan Endocrine Society.

Cushing's disease (CD) is a rare but serious endocrine disorder caused by excessive cortisol secretion due to adrenocorticotropic hormone-secreting pituitary tumors. Despite recent developments in diagnostic criteria and treatment options, CD remains associated with substantial comorbidities and mortality. Early and accurate diagnosis is thus essential. Both the Korean Endocrine Society (KES) and Japan Endocrine Society (JES) guidelines are intended to standardize diagnostic approaches to CD, and they share common principles; however, notable differences exist, particularly in biochemical test

New Insights from the Expression of the Mismatch Repair System in Pituitary Neuroendocrine Tumors.

Although defects in the mismatch repair (MMR) system have been occasionally reported in aggressive/metastatic pituitary neuroendocrine tumors (PitNETs), the potential role of MMR dysregulation in pituitary tumorigenesis is largely unknown. This study aimed to evaluate the expression of the four key MMR components in a large series of PitNETs. MMR gene expression was studied by RT-qPCR in 127 tumors (54 PIT1, 51 SF1, 22 TPIT), and semi-quantitative immunohistochemistry (score 0-12) in selected cases (n=46). MSH2/6 and MLH1 promoters methylation was studied in 96 tumors. Except for MLH1, tumor l

Malignant craniopharyngiomas: Institutional experience and literature review.

Malignant craniopharyngiomas, de novo or via malignant transformation, are exceedingly rare with a dismal prognosis and unclear treatment standards. Little is known about the factors involved in their pathogenesis. A natural language search, performed in our institutional CoPath system, identified 65 adamantinomatous craniopharyngiomas from 56 patients (25 males, 31 females; median age at initial diagnosis = 22 years). Among those, a unique case of malignant craniopharyngioma was identified in a 36-year-old male initially diagnosed with a benign adamantinomatous craniopharyngioma 16 years prio

Diagnosis and management of pituitary neuroendocrine tumors: recent advances.

Pituitary neuroendocrine tumors (PitNETs) are a heterogeneous group of neoplasms originating from anterior pituitary cells, encompassing a spectrum from indolent microadenomas to highly invasive, aggressive tumors. Their clinical presentation varies from classic endocrine hyper- or hyposecretory syndromes to mass effect symptoms or incidental imaging findings. The 2022 World Health Organization classification has redefined PitNETs, emphasizing their neuroendocrine nature and the central role of transcription factor-based immunohistochemistry. Recent years have witnessed rapid progress in molec

Splicing factor FUS facilitates the progression of PIT1-lineage PitNETs by upregulating MDM2.

Background: Splicing factors play pivotal roles in mRNA processing and are implicated in tumor progression. The aberrant expression of splicing factors is closely associated with the invasiveness and secretion profiles of pituitary neuroendocrine tumors (PitNETs). In this study, we explored the involvement of splicing factors in PIT1-lineage PitNET progression and assessed the feasibility of targeting the splicing process as a therapeutic approach. Methods: Statistical data on PitNET subtypes were obtained from the National Brain Tumor Registry of China (NBTRC), and gene expression analysis wa

Cytokine associated neuroinflammation in Parkinson's disease: Molecular pathways, therapeutic targets, and translational insights.

Parkinson's disease (PD) is a progressive neurodegenerative disorder in which neuroinflammation plays a key role. An imbalance between pro- and anti-inflammatory cytokines has been observed in both experimental models and PD patients. The inflammatory mediators activate signaling pathways that lead to oxidative stress, excitotoxicity, blood-brain barrier (BBB) disruption, gut dysbiosis, and hypothalamic-pituitary-adrenal axis (HPA-axis) dysregulation. Increased levels of pro-inflammatory cytokines such as tumor necrosis factor-α (TNF-α), Interleukin-1β (IL-1β), Interleukin-6 (IL-6), and others

Unlocking surgical precision: intraoperative MRI in endoscopic skull base surgery, a meta-analysis.

OBJECTIVE: Intraoperative magnetic resonance imaging (iMRI) is increasingly used in purely endoscopic endonasal skull base surgery (EESBS) to improve surgical accuracy. Given its high cost and logistical demands, quantifying its clinical utility is essential for evidence-based implementation. This meta-analysis was conducted to evaluate the clinical efficacy of iMRI in purely endoscopic endonasal skull base surgery. METHODS: PubMed, Scopus, and Web of Science databases were searched in accordance with PRISMA guidelines. The last search was conducted on November 7, 2024. Eligible studies were o

Long-term cabergoline use does not predict degree of prolactinoma fibrosis nor significantly impact surgical outcomes.

PURPOSE: Prolactinomas are often treated initially with dopamine agonists (DA). For patients subsequently treated with surgery, the effect of cabergoline (CAB) on tumor fibrosis and its potential impact on surgical outcomes is largely unexplored. METHODS: Records of patients with prolactinoma treated by a single surgeon between 2006 and 2024 were examined. Analyses considered relationships among duration and cumulative dose of presurgical DA (DA + vs. DA-), extent of fibrosis measured quantitatively by collagen volume fraction (CVF) and qualitatively by surgeon assessment, and remission statu

Current Approaches and Therapeutic Strategies for Hypothalamic Syndrome in Patients with Childhood-onset Craniopharyngioma.

Patients diagnosed with craniopharyngioma often experience rapid and pronounced weight gain that can progress to severe hypothalamic obesity. This phenomenon is predominantly attributed to disruption of critical hypothalamic regulatory circuits, caused either by direct tumor infiltration or by treatment-related injury. Hypothalamic obesity is best conceptualized within the broader framework of hypothalamic syndrome, a complex clinical disorder encompassing multiple neuroendocrine deficits, impairments in circadian homeostasis, dysregulation of hunger, satiety, and thirst mechanisms, disturbanc

Hsp90α as a promising therapeutic target for suppressing tumor progression in Lactotroph PitNETs.

BACKGROUND: Aggressive Lactotroph Pituitary Neuroendocrine Tumors (Lactotroph PitNETs) usually exhibit invasive growth behavior and resistance to dopamine agonists, showing difficulty in radical treatment and a high recurrence rate. Heat shock protein 90α(Hsp90α),a pivotal isoform of the heat shock protein 90(Hsp90) family, acts as a central chaperone that stabilizes numerous oncoproteins driving tumor progression, but its role in Lactotroph PitNETs remains unclear. OBJECTIVE: To study the effect of Hsp90α knockdown on the proliferation and invasiveness of Lactotroph PitNETs cells (MMQ). MET

Change in salivary cortisol levels in children (aged 9-12 years) with obesity and respiratory diseases during a 28-day spa treatment: a pilot prospective study.

UNLABELLED: Evidence on the neuroendocrine effects of pediatric spa treatment remains limited. Salivary cortisol is a validated, non-invasive biomarker of hypothalamic-pituitary-adrenal (HPA) axis activity, which may serve as an indicator of physiological adaptation to structured environmental and behavioral interventions. To determine whether a standardized 28-day spa treatment is associated with changes in morning salivary cortisol in children aged 9-12 years with obesity or chronic respiratory diseases, and to compare cortisol trajectories in these diagnostic groups with those of healthy co

Preoperative medical therapy for acromegaly: current knowledge and clinical significance.

Acromegaly is a chronic endocrine disorder characterized by excessive secretion of growth hormone (GH), predominantly caused by pituitary adenomas. Despite advancements in neurosurgical techniques, the surgical remission rates for invasive macroadenomas or giant adenomas remain unsatisfactory. Therefore, multimodal treatment strategies, including preoperative medical therapy (POMT), have been implemented to improve patient outcomes. Among these, first-generation somatostatin receptor ligands (fg-SRLs) have been the most extensively studied preoperative agents; however, their clinical efficacy
