Bilateral cranial polyneuropathy associated with an intracranial mass in a cat with concurrent tonsillar lymphoma.

A 14-year-old spayed female Siamese cat was presented with signs consistent with a rapidly progressive cranial polyneuropathy. Magnetic resonance imaging of the brain revealed an extra-axial mass at the calvarial floor, extending caudally to involve bilateral trigeminal nerves and rostrally to involve the pituitary gland and optic chiasm. Additional findings included contrast-enhancing content within the bilateral tympanic bullae, mild mandibular and medial retropharyngeal lymphadenopathy, and a left tonsillar mass. Cytologic and histopathologic findings confirmed tonsillar lymphoma. The concu

Activator of Sirt6 exerts antitumor activity in pituitary neuroendocrine tumors.

CONTEXT: Pituitary neuroendocrine tumor (PitNET) is one of the most common intracranial neuroendocrine tumors. Surgical resection is the first choice for most subtypes of PitNETs. However, radical removal of invasive tumors remains challenging. A novel pharmacological therapy is objectively needed. Sirtuin 6 (Sirt6) is a member of the NAD+-dependent class III deacetylase sirtuin family, which plays a key role in tumorigenesis in many tumors. However, the role of Sirt6 and its activator in PitNETs is still unclear. OBJECTIVE: In the study, we investigated the expression of Sirt6 and the antitu

Early life stress caused glucose homeostasis impairment and anxiety-like behavior along with oxidative stress in adolescent male rat offspring: Protective effect of saffron extract.

Early life stress (ELS) significantly contributes to metabolic and psychological disorders by triggering oxidative stress and disrupting the hypothalamic-pituitary-adrenal (HPA) axis. This research examined ELS effects on redox status, along with anxiety-like behavior, in adolescent male Wistar rats, also assessing saffron (Crocus sativus L.) extract's potential benefits. Rats were exposed to foot shock stress from postnatal days 15-19, with treatment groups receiving saline or saffron extract (10 mg/kg) via intraperitoneal injection. Control groups were maintained without stress exposure. Ele

A post-operative fluid restriction protocol reduces the incidence of delayed hyponatremia following trans-sphenoidal pituitary surgery.

PURPOSE: The study assessed the effect of a postoperative fluid restriction on the incidence of delayed hyponatremia (DH) following transsphenoidal pituitary surgery (TSS). The incidence of DH in the prospective cohort was compared to that of a published retrospective cohort from our centre managed according to the standard postoperative protocol without fluid restriction. METHODS: This prospective interventional study included patients aged ≥18 years who underwent TSS for pituitary neuroendocrine tumors (PitNETs). A structured postoperative fluid restriction of ≤1.5 L/day was implemented fro

Dopamine agonist-associated fibrosis in prolactinomas: clinical challenges, impacts on the tumor microenvironment, and future directions.

Prolactinomas account for approximately 30% to 50% of pituitary adenomas. Dopamine agonists (DAs) are the first-line treatment and effectively induce tumor regression while normalizing prolactin levels. However, DA therapy not only inhibits tumor growth but also is associated with fibrosis within the tumor. This fibrotic transformation remodels the tumor microenvironment (TME), altering cellular interactions, vascular structures, and extracellular matrix (ECM) composition. We hypothesize that this fibrotic response may not be a mere side effect; emerging, though largely indirect, evidence sugg

Proopiomelanocortin (POMC)-β-Endorphin System in the Rat Colon: Expression, Up Regulation by Stress, and Correlation With Stress-Induced Visceral Analgesia.

BACKGROUND: The pituitary proopiomelanocortin (POMC)-β-endorphin (β-END) system is activated by hypothalamic corticotropin-releasing factor (CRF) signaling and stress. We investigated whether this pathway is also expressed in the rat colon, regulated by peripheral CRF and stress, and correlated with stress-induced visceral analgesia. METHODS: POMC and the prohormone convertases PC1/3 and PC2 transcripts were assessed by RT-PCR, and POMC and β-END localization was examined by immunohistochemistry in the proximal and distal colon of naïve adult male Sprague-Dawley rats. Proximal colonic POMC mR

Prepregnancy Dopamine Agonist Treatment Duration as a Predictor of Postpartum Prolactinoma Remission.

OBJECTIVES: Prolactinoma is a major cause of infertility in women of reproductive age. Although pregnancy has been proposed to induce prolactinoma remission, evidence remains inconsistent because of the lack of well-matched control groups. This study evaluated the impact of pregnancy on prolactinoma outcomes and identified predictors of sustained postpartum remission. METHODS: We retrospectively analyzed 225 women with prolactinoma treated between 2003 and 2024. Seventy-five patients who became pregnant during dopamine agonist (DA) therapy were matched 1:2 with 150 nonpregnant controls using

Epigenetics of Chronic Stress: Mechanisms and Health Impacts.

Chronic stress is a major public health concern, contributing to a spectrum of physiological and psychological disorders, including anxiety, depression, cardiovascular diseases, and metabolic syndromes. This review examines the epigenetic mechanisms mediating the impact of chronic stress on human health, focusing on DNA methylation, histone modifications, and non-coding RNAs. These epigenetic alterations act as dynamic interfaces between environmental stressors and genomic responses, modifying gene expression in critical stress-related pathways, such as the hypothalamic-pituitary-adrenal (HPA)

Machine Learning-Driven PCDI Classifier for Invasive PitNETs.

INTRODUCTION: Aggressive Pituitary Neuroendocrine Tumors (PitNETs) pose significant therapeutic challenges due to their invasive behavior and resistance to conventional therapies. Current prognostic markers lack the ability to capture molecular heterogeneity, necessitating novel biomarkers. Dysregulated Programmed Cell Death (PCD) pathways are implicated in tumorigenesis, but their prognostic relevance in invasive PitNETs remains unexplored. METHODS: GEO datasets (GSE51618, GSE169498, GSE260487) were analyzed to identify differential gene expression between noninvasive and invasive PitNETs. A

The Incidence of Calcified Pituitary Macroadenomas: Challenging the Craniopharyngioma Assumption.

BACKGROUND AND PURPOSE: Large sellar/suprasellar masses containing calcifications are classically associated with craniopharyngiomas. In our practice, however, we have noticed that some giant pituitary macroadenomas have calcifications. The purpose of our study was to evaluate the incidence and characteristics of calcifications in pituitary macroadenomas. MATERIALS AND METHODS: We reviewed an internal database for pathology-proven giant macroadenomas (≥4 cm). Basic patient demographics were collected, including age and sex. Adenoma subtype and maximum adenoma dimension were recorded. Preopera

Clinical and Pathological Spectrum of Acromegaly: Distinguishing GH PitNETs, Mammosomatotrophs, and Mixed Tumors.

INTRODUCTION: Acromegaly is a rare disease usually caused by a pituitary neuroendocrine tumor (PitNET) that produces GH PitNET. PitNETs secreting GH and prolactin (GH&PRL PitNETs) contribute up to 30% to the spectrum of acromegaly and have been attributed a more aggressive behavior. GH&PRL PitNETs can be classified into 2 predominant phenotypes: mammosomatotroph arising from a single-cell population of Pit-1 lineage and mixed somatotroph-lactotroph PitNETs (mixed SL PitNETs). PURPOSE: To evaluate the clinical and molecular differences between GH PitNETs, mammosomatotroph, and mixed SL PitNETs
