A spatial transcriptomics comparison of the adult versus metamorphosed axolotl brain.

The Mexican axolotl (Ambystoma mexicanum) is an established model for studying tetrapod regeneration and development. Notably, axolotls exhibit remarkable brain regeneration as adults, a trait rarely observed in other adult vertebrates. Adult axolotls can undergo metamorphosis, a process that induces dramatic remodeling of multiple organs and is accompanied by a gradual decline in regenerative capacity and lifespan. However, systematic studies on whole-brain cellular dynamics and molecular mechanisms in both adult and metamorphosed individuals remain lacking. Here, we profiled five representat

Class III Obesity as a Presenting Syndrome in Prolactinoma Patients: Insights Into Obesity and Metabolic Dysfunction in This Unique Group.

The occurrence of patients with lactotroph adenoma presenting with class III obesity is extremely rare. This study reports long-term outcomes of such patients treated with transsphenoidal surgery (TSS) or dopamine agonists (DAs) and integrates findings with existing literature to explore prolactin's (PRL) role in obesity pathophysiology. We analysed BMI and metabolic changes and compared findings with published case series. Four patients presented with class III obesity (BMI range, 40.0-43.1 kg/m2). The diagnostic work-up revealed hyperprolactinemia (PRL range, 56.1-4389 μg/L) and hypogonadism

Consensus on the diagnosis of Cushing's disease: a collaborative statement from the Korean Endocrine Society and Japan Endocrine Society.

Cushing's disease (CD) is a rare but serious endocrine disorder caused by excessive cortisol secretion due to adrenocorticotropic hormone-secreting pituitary tumors. Despite recent developments in diagnostic criteria and treatment options, CD remains associated with substantial comorbidities and mortality. Early and accurate diagnosis is thus essential. Both the Korean Endocrine Society (KES) and Japan Endocrine Society (JES) guidelines are intended to standardize diagnostic approaches to CD, and they share common principles; however, notable differences exist, particularly in biochemical test

Pituitary Neuroendocrine Tumors Extending Primarily Below the Sella and into the Clivus: A Distinct Growth Pattern with Specific Challenges.

Evaluation of pituitary neuroendocrine tumors remains complex depending on the exact growth pattern, involvement of critical neurovascular structures, pituitary function and endocrinological activity of the tumor. A predominant growth into the sphenoid sinus and clivus poses specific challenges. We reviewed 557 surgeries for pituitary neuroendocrine tumors in an endonasal endoscopic technique performed between 1 January 2015 and 31 August 2025 to identify 13 cases (2.3%). Clinical, radiological and surgical data were selected by chart review. Thirteen cases aged from 31 to 68 years with almost

Multi-class classification of brain tumors using optimized CNN and transfer learning techniques.

Brain tumors are abnormal cell growths within the brain or central nervous system that disrupt normal brain function and can be life-threatening. Early detection and precise classification of brain tumors are critical for effective treatment and improved patient prognosis. In this research, a Convolutional Neural Networks model with four convolution blocks is proposed for automated brain tumor detection and classification. The proposed scheme is applied to four types of MRI images, i.e., glioma tumor, meningioma tumor, no tumor, and pituitary tumor. The proposed architecture is optimized using

Mitochondrial Dysfunction: The Cellular Bridge from Emotional Stress to Disease Onset: A Narrative Review.

Severe emotional stress constitutes a significant public-health concern associated with negative health outcomes. Although the clinical effects are well acknowledged, the specific biological mechanisms that translate emotional suffering into systemic disease remain incompletely understood. Psychological stress activates the sympathetic nervous system and hypothalamic-pituitary-adrenal axis, which directly target mitochondria and alter their bioenergetic and redox capacity. For this reason, this narrative review proposes that mitochondria serve as the primary subcellular link in the mind-body c

Age-Dependent Effects of Heavy Metals on the Hypothalamic-Pituitary-Testicular Axis-Related Hormones in Men.

The effect of heavy metals on male hormonal regulation-particularly the hypothalamic-pituitary-testicular (HPT) axis-remains poorly characterized. We aim to investigate associations between heavy metal exposure and HPT axis-related hormones. We analyzed data, including male participants aged 3-80 years, from a nationally representative survey. Five metals and twelve sex hormones were measured. We used multivariate linear regression and restricted cubic splines to assess associations and dose-response relationships. Mixture effects were quantified using quantile-based g computation. The modifyi

Critical views for safe surgical phase progression in endoscopic endonasal transsphenoidal pituitary adenoma resection: modified Delphi consensus.

PURPOSE: Endonasal transsphenoidal surgery is the gold-standard for pituitary adenoma resection, yet no intraoperative framework exists to confirm safe phase progression. Inspired by the Critical View of Safety in laparoscopic cholecystectomy and engineering “phase-gate” process, we propose the Critical Views for Phase Progression (CVPPs) – a set of visual cues confirming phase objectives and safe phase progression. Designed to be clinically relevant and machine-readable, CVPPs aim to support training and future AI-driven guidance systems. METHODS: A three-round modified Delphi process was con

Mixed gangliocytoma-pituitary neuroendocrine tumour: clinical, immunohistochemical, and molecular genetic profiles in a series of four patients.

The vast majority of tumours in the sellar region are pituitary neuroendocrine tumours, also called pituitary adenomas. Sellar gangliocytomas (GCs), benign tumours that originate from neuronal ganglionic cells, account for less than 1% of sellar tumours. Even rarer are mixed gangliocytoma-pituitary neuroendocrine tumours (GC-PitNET). These tumours are often associated with hormone hypersecretion, most commonly resulting in acromegaly. The histogenesis of mixed GC-PitNET is currently unclear. In this paper, we present comprehensive clinical, immunohistochemical, targeting enrichment next genera

Incidental 18 F-Flortaucipir Uptake in Pituitary Macroadenoma.

A 73-year-old woman with over 2 years of progressive memory decline underwent amyloid PET/CT ( 18 F-florbetaben) and tau PET/CT ( 18 F-flortaucipir) imaging for suspected Alzheimer disease (AD). This series of examinations confirmed the diagnosis of AD. Unexpectedly, a pituitary macroadenoma with significant 18 F-flortaucipir uptake was revealed, which might represent another instance of off-target binding.

Growth Hormone, Not Simply Just a Hormone for Growth.

Since its discovery almost 100 years ago, growth hormone (GH) has been extensively studied to elucidate its structural characteristics, receptor interactions and its physiologic and non-physiologic effects. These actions include but are not limited to its effects on somatic growth, substrate metabolism, body composition, bone mineral density, cardiovascular system, and cognitive function. Contextually, recombinant human GH was approved for growth promotion in children and to enhance metabolic health in adult patients with GH deficiency (GHD), along with other clinical indications. Studies invo

Radiotherapy regimens and concurrent Cabergoline use for non-functioning pituitary neuroendocrine tumors: a large, single-center cohort.

BACKGROUND: The standard treatment for non-functioning pituitary neuroendocrine tumors (PitNET) consists in transsphenoidal surgical resection, however when progression or recurrence happens, additional treatments like stereotactic radiotherapy (SRT) and dopaminergic agonists can be used. In this article we aim to describe and quantify the impact of SRT regimen and dose, as well as the use of concurrent cabergoline, on local tumor control and toxicities. METHODS: This is a retrospective cohort of patients with non-functioning PitNET treated between 2008 and 2023 with LINAC-based SRT on differe

Preoperative Metabolic Predictors of Granulation Subtypes in Somatotroph Tumors: A Multicenter Retrospective Cohort Study.

PURPOSE: Differentiating between sparsely granulated and densely granulated somatotroph tumors (SGSTs and DGSTs) currently relies on postoperative immunohistochemistry. This study aimed to evaluate whether triglyceride (TG), uric acid (UA), and their composite TG-UA index [ln(TG × 1000/UA)] could serve as preoperative indicators for distinguishing granulation subtypes of somatotroph tumors. METHODS: In this multicenter retrospective cohort study, 230 patients with somatotroph tumors were analyzed. Logistic regression and generalized additive models assessed associations and potential nonlinea

BRISC: Annotated Dataset for Brain Tumor Segmentation and Classification.

Accurate segmentation and classification of brain tumors from Magnetic Resonance Imaging (MRI) remain key challenges in medical image analysis, primarily due to the lack of high-quality, balanced, and diverse datasets with expert annotations. In this work, we address this gap by introducing BRISC, a dataset designed for brain tumor segmentation and classification tasks, featuring high-resolution segmentation masks. The dataset comprises 6,000 contrast-enhanced T1-weighted MRI scans, which were collated from multiple public datasets that lacked segmentation labels. Our primary contribution is t
