Persistent androgen receptor (AR) activation is an important contributor to polycystic ovary syndrome (PCOS) and is affected by transcriptional regulation via histone acetylation; however, the underlying mechanisms are partially understood. This study demonstrated that AR activation in ovarian granulosa cells (GCs) of both dehydroepiandrosterone (DHEA) and high-fat diet-induced PCOS mouse models correlated with a significant increase in the histone acetyltransferase p300 and histone acetylation. Conversely, GC-specific p300 knockout or pharmacological inhibition with C646/A-485 effectively red

The quality-of-life (QoL) impacts of benign gynecological conditions, such as polycystic ovarian syndrome (PCOS), endometriosis, and uterine fibroids on patients' lives are underestimated. Upon analysis of the 36-Item Short Form Health Survey questionnaire (SF-36), which assesses physical functioning, role physical, bodily pain, general health, vitality, social functioning, role emotional, and mental health, benign gynecological conditions reveal comparable or worse QoL scores compared to QoL scores in other chronic conditions such as diabetes mellitus, breast cancer, and heart disease. Infertility is defined as the inability to conceive spontaneously after 12 months of routinely unprotected intercourse in patients <35 years of age, and after 6 months in patients ≥35 years of age. Infertility also refers to the inability to achieve a successful pregnancy based on the medical, sexual, and reproductive history; age, physical findings; diagnostic testing; the need for medical intervention; or a combination of these variables for a patient. In the United States, 13% of women 15 to 49 years of age have accessed assisted fertility services and it has been suggested that the rate of

A prevalent endocrinopathy affecting reproductive-age women is polycystic ovarian syndrome (PCOS), which affects around 5-20 % of these individuals. PCOS is closely linked to the following causes: immunological, endocrine, and reactive oxygen species (ROS). Plant extracts are used in the synthesis of silver nanoparticles (AgNPs), which are employed for PCOS treatment. Ag NPs can regulate the hormonal imbalance, reducing pro-inflammatory cytokines and ovarian cysts in PCOS. Conventional medicines, such as metformin, clomiphene citrate, letrozole, flutamide, and rosiglitazone, can regulate menst

BACKGROUND: Lifestyle interventions have been proposed to be the first-line treatment for polycystic ovarian syndrome (PCOS), but our knowledge of PCOS-related lifestyle factors is still limited. This study aimed to explore PCOS-related lifestyle factors in reproductive-age women in China. METHODS: A case-control study was carried out in Shandong, China, from June to October 2014. 350 PCOS-related infertile women were recruited as cases, while 308 women who were seeking the treatment of fallopian tube obstruction or assisted reproduction due to male infertility were recruited as controls. Capitalizing latent unobserved structures to improve the accuracy of predictive models has become an active avenue for deep learning research. Most approaches cluster original features to infer latent structures. However, the information gained during the process can often be implicitly derived using sufficiently complex models. Thus, these approaches often provide minimal benefits. We propose a Shapley Additive exPlanations-based supervised deep learning framework called FORCE (Feature Oriented Representation with Clustering and Explanation), which relies on two-stage usage of SHAP values in

Polycystic ovarian syndrome (PCOS) is a female endocrine and reproductive disease. The exact etiology remains unknown. There are several associated factors involved in the pathogenesis of PCOS, such as hyperandrogenism, insulin resistance, obesity, inflammation, oxidative stress, genetic, epigenetic, and environmental factors. However, the main diagnostic criteria involve at least two out of three symptoms, which are hyperandrogenism, polycystic ovaries, and menstrual cycle dysregulation. The treatment of the multiple symptoms of PCOS is still a medical challenge. Different molecular pathways

INTRODUCTION: Abundant evidence suggests that women with polycystic ovary syndrome (PCOS) have increased metabolic aberrations and cardiovascular risks and present with signs of endothelial cell (EC) dysfunction. However, whether and how androgen is involved in the pathogenesis of EC dysfunction in PCOS remains unclear.
METHODS: In this study, induced pluripotent stem cells (iPSCs) were established from three phenotype A PCOS patients (presenting with oligomenorrhea, hyperandrogenism (HA), and polycystic ovarian morphology (PCOM)) and three control participants. These iPSCs were differentiate

BACKGROUND: Polycystic Ovarian Syndrome (PCOS) is a hormonal disorder affecting women of reproductive age. It frequently causes hormonal imbalance, irregular menstrual cycle, and in some cases, infertility. For centuries, traditional herbs like Shatavari (Asparagus racemosus Willd.) have been used to support women's reproductive health, and some studies suggest it may help with PCOS symptoms. Thus, this study aimed to evaluate the safety and efficacy of standardized Shatavari root extract in women with PCOS.

BACKGROUND: The global obesity surge has precipitated a parallel rise in diabetes, cardiovascular, kidney, and reproductive-metabolic diseases like polycystic ovarian syndrome (PCOS), creating an unsustainable healthcare burden fragmented by organ-specific nomenclature. Legacy terms like type 2 diabetes and non-alcoholic fatty liver disease (NAFLD) focus on end-stage manifestations, obscuring shared pathogenic roots. This review aimed to propose and define the Fuel Toxicity-Associated Spectrum (FTAS) as a unifying nomenclature and staging framework for adiposity-related metabolic disorders. Th
