Network Pharmacology Integrated Molecular Docking Analysis Identifies Potential Bioactive Compounds in Nigella sativa Against Polycystic Ovary Syndrome.

Polycystic ovarian syndrome (PCOS) is a complex endocrine disorder affecting 8%-13% of women of reproductive age. Although Nigella sativa (NS) has been traditionally used for PCOS management, its molecular mechanisms are not fully characterized. This study employed network pharmacology, molecular docking, and molecular dynamics (MD) simulations to identify the key targets and pathways associated with NS bioactives. Venn analysis identified 53 common genes shared between NS-derived phytochemicals and PCOS-associated targets. KEGG and Reactome enrichment revealed 15 significantly enriched pathwa

Phytochemical-rich extract of Psidium guajava ameliorates letrozole-induced PCOS in female rats by modulating the NF-κB inflammatory pathway.

Polycystic ovarian syndrome is a multidisciplinary endocrinopathy with complex symptomatology. This study aimed to explore the pharmacological potential of the ethanolic extract of Psidium guajava (EEPG) in PCOS with a mechanistic approach and its effects on ovarian histopathology. EEPG was characterized by using HPLC. Thirty female adult rats were divided into six groups (n = 5): the normal control, the disease-induced, metformin, and EEPG (200, 400, and 600 mg/kg). PCOS was induced by administering letrozole (1 mg/kg). Biochemical, molecular, and histopathological evaluations were performed.

Emerging and Off-Label Uses Of Glucagon-Like Peptide-1 Receptor Agonists (GLP1-RA) and Dual GIP/GLP1-RAs.

BACKGROUND: Glucagon-like peptide-1 receptor agonists (GLP1-RAs) and the glucagon-dependent insulinotropic polypeptide (GIP)/GLP1-RA are approved for type 2 diabetes (T2D) and obesity given their profound impact on glycemic weight management. Additional indications include reducing cardiovascular disease risk and progression of chronic kidney disease (CKD) in T2D as well as obstructive sleep apnea in patients with obesity. These enhanced effects are likely due to their pleiotropic effects, leading to decreased inflammation and other benefits. This review explored emerging evidence for uses of

Fracture incidence in women with polycystic ovarian syndrome: A retrospective cohort analysis.

BACKGROUND: Polycystic Ovarian Syndrome (PCOS) impacts reproductive-age women through metabolic disturbances and hormone imbalances. These changes have been linked to bone health, but the association between PCOS and incidence of fractures remains unclear. Clarifying the link between PCOS and fractures is important to guide clinical care and develop prevention strategies. METHODS: A retrospective cohort analysis using the TriNetX research platform evaluated fracture incidence in female patients with and without PCOS. The study assessed the occurrence of distal radius, femoral neck, ankle, and

Metabolic-endocrine correlates of gut-brain axis mediators in polycystic ovary syndrome: A case-control study.

Polycystic ovary syndrome (PCOS) is increasingly recognized as a neuro-metabolic disorder involving disrupted signaling between the brain-gut axis and reproductive system. Peptides such as kisspeptin, ghrelin, serotonin, and peptide tyrosine tyrosine (PYY) are emerging as integrators of metabolic and endocrine function, yet their coordinated alterations in PCOS and relevance to metabolic risk remain underexplored. Therefore, we investigated the combined pattern of serum kisspeptin, ghrelin, serotonin, and PYY in PCOS, and their associations with metabolic and hormonal indices. In this case-con

Preoperative Neutrophil-to-Lymphocyte Ratio is Associated with Early Postoperative Pain and Opioid Consumption after Total Knee Arthroplasty: A Retrospective Study.

BACKGROUND: Improving quality and effectiveness of health care is one of the priorities of health policies. Hospital or physician volume represents a measurable variable with a relevant impact on effectiveness of health care. A recent Italian law, the "spending review", calls for the definition of "qualitative, structural, technological and quantitative standards of hospital care". There is a need for an accurate evaluation of the available scientific evidence in order to identify these standards, including the volume of care above or below which the public and private hospitals may be accredi

Thyrotoxicosis and Moyamoya Syndrome: A Fatal Intersection of Rare Disorders in a Young Patient With a Background of Hypothyroidism.

Thyroid storm, also known as thyrotoxic crisis, is an uncommon and potentially life-threatening condition that can cause metabolic, cardiovascular and neurological complications. Thyrotoxic crisis often develops in the context of untreated or poorly controlled hyperthyroidism and is typically precipitated by acute stressors such as infection, surgery, or trauma. While the progression from hypothyroidism to hyperthyroidism is itself an uncommon clinical occurrence, the emergence of thyroid storm in this setting is sporadic. Such a case highlights the importance of being aware of atypical presen

Quercetin-3-Glucoside from Ocimum basilicum ameliorates polycystic ovary syndrome by targeting the IL-6/JAK-STAT3 signaling axis.

Chronic inflammation is a key contributor to the pathophysiology of polycystic ovary syndrome (PCOS), disrupting hormonal signaling and ovarian function. This study investigated the therapeutic potential of Quercetin-3-Glucoside (Q3G), a bioactive flavonoid from Ocimum basilicum (OB), against PCOS-associated inflammation. An integrated approach of network pharmacology and molecular docking predicted Q3G interaction with key inflammatory targets IL6, STAT3, and BCL2, predominantly through the JAK-STAT signaling pathway. These predictions were validated in a letrozole-induced PCOS mouse model. M

Approach to the diagnosis and treatment of polycystic ovarian syndrome in adolescent patients.

Polycystic ovary syndrome (PCOS) is one of the most common endocrine disorders affecting women of reproductive age. Its diagnosis during adolescence is particularly challenging, as physiological pubertal changes overlap with key features of PCOS; nevertheless, this developmental stage often represents the earliest window for clinical manifestation. Accordingly, a definitive diagnosis should be established only in the presence of clear and persistent evidence of both hyperandrogenism and menstrual irregularity. Adolescents presenting with a single feature should be considered at increased risk
