Neisseria meningitidis Serogroup Y Sequence Type 1466 and Urogenital Infections.

Neisseria meningitidis is a common commensal bacterium of the nasopharynx that can cause invasive meningococcal disease (IMD). In comparison, N. gonorrhoeae is always a pathogen usually limited to mucosal sites. However, increased evidence for overlapping clinical syndromes is emerging. We compared N. meningitidis samples from a urogenital outbreak in Australia with sequences from the United States and other countries. We conducted phylogenetic analyses to assess relatedness and examine for genomic changes associated with meningococcal adaptation; we collated a total of 255 serogroup Y (MenY),

A systematic review to identify research gaps in studies modeling MenB vaccinations against Neisseria infections.

The genus Neisseria includes two major human pathogens: N. meningitidis causing bacterial meningitis/septicemia and N. gonorrhoeae causing gonorrhoea. Mathematical models have been used to simulate their transmission and control strategies, and the recent observation of a meningococcal B (MenB) vaccine being partially effective against gonorrhoea has led to an increased modeling interest. Here we conducted a systematic review of the literature, focusing on studies that model vaccination strategies with MenB vaccines against Neisseria incidence and antimicrobial resistance. Using journal, prepr

Serum bactericidal activity against meningococcus in patients with systemic lupus erythematosus.

BACKGROUND: Patients with systemic lupus erythematosus (SLE) are susceptible to infectious diseases owing to various immunosuppressive treatments and disease characteristics. Meningococcal infections progress rapidly with a high incidence of severe complications and mortality; therefore, meningococcal vaccination is needed. However, there is limited evidence regarding the immunity and immunogenicity of patients with SLE. PURPOSE: This study aimed to analyze the serum bactericidal activity against meningococci in patients with SLE in 2 domestic institutions in Korea. METHODS: Serum samples we

Adjuvant-dependent impacts on vaccine-induced humoral responses and protection in preclinical models of nasal and genital colonization by pathogenic Neisseria.

Neisseria gonorrhoeae, which causes the sexually transmitted infection gonorrhea and Neisseria meningitidis, a leading cause of bacterial meningitis and septicemia, are closely related human-restricted pathogens that inhabit distinct primary mucosal niches. While successful vaccines against invasive meningococcal disease have been available for decades, the rapid rise in antibiotic resistance has led to an urgent need to develop an effective gonococcal vaccine. Several surface antigens are shared among these two pathogens, making cross-species protection an exciting prospect. However, the type

Amino acid substitutions in PBP3 in Haemophilus influenzae strains, their phenotypic detection and impact on resistance to β-lactams.

BACKGROUND: Data from surveillance on antibiotic resistance have shown an increasing prevalence of non-enzymatic resistance (β-lactamase-negative ampicillin-resistant) to β-lactam antibiotics among H. influenzae strains in the Czech Republic. Aminopenicillins are recommended agents for non-invasive Haemophilus influenzae infections. The phenomenon of non-enzymatic resistance to β-lactams is complicated by the fact that the phenotypic detection of PBP3 with specific amino acid substitutions (rPBP3) is challenging, since rPBP3 isolates have repeatedly been demonstrated to be split by the epidemi

Fully automated identification of Neisseria meningitidis using the BD MAX system in a clinical laboratory setting: A preliminary study.

The rapid and accurate identification of Neisseria meningitidis, a pathogen that causes invasive infections and meningitis, is crucial for its effective clinical management and infection control. However, identification using matrix-assisted laser desorption/ionization time-of-flight mass spectrometry may misidentify other Neisseria species as N. meningitidis, thus necessitating confirmatory tests based on biochemical properties. These tests require high bacterial concentrations that are achieved through subculturing, which can increase biosafety risks in laboratories. In this study, we develo

Nanoparticles displaying fHbp elicit an enhanced antibody response against meningococcal B isolates compared to low valency fHbp antigens.

Serogroup B meningococcus (MenB) is one of the leading causes of invasive meningococcal disease (IMD) in Western countries. While outbreaks of IMD are rare, this disease can lead to long-term disabilities and even death. These outbreaks typically occur in infants, children, and young adults. There are currently two licensed MenB vaccines: 4CMenB (Bexsero®; GSK Vaccines, Srl, Italy) and MenB-FHbp (Trumenba®, bivalent rLP2086; Pfizer Inc., Collegeville, PA). The effectiveness of these vaccines is dependent upon their ability to elicit a protective antibody response against the various disease-ca

Comparison of Streptococcus pneumoniae isolates occurring in optochin-susceptible and optochin-resistant variants by analyzing whole-genome sequencing data.

UNLABELLED: The paper presents the study of a set of isolates of Streptococcus pneumoniae, which comprised two heterogeneous subpopulations, one of which was susceptible and the other resistant to optochin. The aim of the study was to compare the results of serotyping, multilocus sequence typing (MLST), ribosomal multilocus sequence typing (rMLST), and variation analysis of these subpopulations and to investigate the genetic probable causes of optochin resistance. The strains studied were cultured from samples taken from patients with invasive pneumococcal disease in the Czech Republic in 2019

Identification of immunogenic outer membrane vesicle vaccine antigen components using a meningococcal protein microarray.

Outer membrane vesicle (OMV)-based vaccines have been employed worldwide in response to epidemic meningococcal disease outbreaks caused by Neisseria meningitidis. The complex composition of OMVs raises challenges in the identification of antigens which contribute to a protective immune response. Here, we measured total IgG antibody binding profiles to a dedicated antigen microarray using human sera from an open-label Phase II trial (NCT00962624) of 4CMenB (Bexsero), a licensed vaccine containing an OMV component. Significant IgG responses were observed against specific Outer Membrane Proteins

Two New Kindreds with Complete Factor D Deficiency.

Inborn deficiencies of the alternative pathway (AP) of the complement system have been associated with life-threatening infections, mainly by encapsulated bacteria. Complete factor D (FD) deficiencies have been reported in only seven families in the literature. We report two new cases of biochemically and genetically confirmed complete FD deficiency, including the first in a Down syndrome patient. The index cases respectively suffered from severe H. influenza and N. meningitidis infections. Their FD activity was undetectable but was restored by adding recombinant human FD. FD levels were undet

Emergence of high-level penicillin-resistant Neisseria meningitidis harboring ROB-1-type β-lactamase gene in Japan.

While antimicrobial resistance in Neisseria meningitidis, the causative agent for invasive meningococcal disease (IMD), is rare, some meningococcal isolates in Japan were resistant to ciprofloxacin and penicillin (PCG). Among 290 meningococci isolated from 2003 to 2020 in Japan, four PCG resistant (PCGR) meningococci were found but showed the minimum inhibitory concentration (MIC) with 0.5 μg/mL at a maximum due to mutation in penA gene that encodes penicillin-binding protein 2. However, we herein report that PCGR meningococci harboring the ROB-1-type β-lactamase gene (blaROB-1) with PCG MIC o
