Differential microRNA Expression Profiles in Girls with Idiopathic Central Precocious Puberty and Premature Thelarche.

Idiopathic central precocious puberty (CPP) is increasingly observed in girls. Premature thelarche (PT) and exaggerated thelarche (ET) are early pubertal variants that can be challenging to distinguish from CPP in clinical practice. Exosomal microRNAs are stable biomarkers capable of crossing the blood-brain barrier. Although miR-30b-5p has been reported to increase in pubertal boys and girls, human studies investigating microRNAs in CPP and puberty remain limited. To investigate exosomal microRNA expression profiles and associated pathways in early pubertal development, we conducted a cross-s

Optimization and verification of high-fat diet formulation for establishing a rat model of obesity-related precocious puberty.

Childhood obesity is closely linked to the rising incidence of precocious puberty (PP), yet valid preclinical models of obesity-related PP remain lacking. High-fat diets (HFD) are widely used for obesity models, but the suitability of different HFD formulations for inducing PP is unknown. Female Sprague-Dawley (SD) rats were randomized to control, 45% HFD, or 60% HFD groups at postnatal day (PND) 21. Body weight and daily caloric intake were monitored; vaginal opening (VO) was recorded from PND 35. At study termination, serum levels of estradiol (E2), luteinizing hormone (LH), follicle-stimula

Bone and body composition by DXA in girls with precocious puberty, premature thelarche, and prepubertal controls.

CONTEXT: Lumbar spine bone mineral density (BMD) rises sharply during puberty; earlier onset, as in central precocious puberty (CPP), may accelerate skeletal maturation and modify bone accrual. OBJECTIVE: To assess bone and body composition in girls with CPP/early puberty (EP), premature thelarche (PT), and prepubertal controls (PC). PATIENTS AND METHODS: We analyzed 184 girls aged 5-9 years with suspected CPP/EP (108 CPP/EP, 76 PT) and 47 PC. DXA assessed L1-L4 and total body less head (TBLH) BMD, bone mineral content (BMC), and body composition. Derived measures included bone mineral appar

Final adult height in male patients with central precocious puberty after gonadotropin-releasing hormone agonist treatment.

PURPOSE: We aimed to compare the final adult height (FAH) of male patients with central precocious puberty (CPP) after treatment with a gonadotropin-releasing hormone agonist (GnRHa). Specifically, we compared FAH with the target height (TH) and the predicted adult height (PAH) before and after GnRHa treatment to quantify height gain and identify predictive factors. METHODS: We retrospectively reviewed the medical records of 92 male patients with CPP and known FAH after GnRHa treatment at the Department of Pediatrics of Severance Children's Hospital between January 2000 and June 2024. Development and evaluation of a predictive model for central precocious puberty in girls.

BACKGROUND: Early puberty, particularly central precocious puberty (CPP), is an increasingly common pediatric endocrine disorder affecting young girls. Existing CPP predictive models either rely on post-gonadotropin-releasing hormone (GnRH) stimulation data or lack comprehensive integration of clinical, laboratory, and imaging indices, limiting their utility for early screening. This study developed a single blood draw-based predictive model for CPP screening or diagnosis. METHODS: This retrospective study was conducted at a single center (The Second Affiliated Hospital of Shantou University

Solution structure of the novel CH-domain zinc finger from the puberty regulator Makorin-3.

The makorin (MKRN) family of ubiquitin ligases regulates diverse biological processes, including reproduction, neurogenesis, and immune function. The first identified member, MKRN3, is an inhibitor of sexual development that is the site of inherited mutations linked to central precocious puberty (CPP). All makorin proteins share a distinctive cysteine/histidine-rich (CH) domain. In MKRN3, the CH domain lies between the second C3H zinc finger and the RING domain. Using CD and NMR spectroscopy, we show that the CH domain folds upon coordination of a single Zn2+ ion with picomolar affinity. Spect

Fixed Dosing of Leuprolide Acetate, a GnRH Agonist, in Children with Central Precocious Puberty: A Population Pharmacokinetic Justification.

BACKGROUND: Central precocious puberty is the early onset of puberty due to premature activation of the hypothalamic-pituitary-gonadal axis, which can reduce adult height. Leuprolide, a gonadotropin-releasing hormone agonist, reduces gonadotropin secretion and is the standard treatment for central precocious puberty. OBJECTIVE: This study aimed to build a population model to describe the pharmacokinetics of a 3-month leuprolide acetate depot formulation in pediatric patients with central precocious puberty, evaluate covariate effects (age and weight) on leuprolide pharmacokinetics, and assess

Precocious puberty: An overview of pathogenesis, clinical presentation, and management.

Precocious puberty, defined as the onset of secondary sexual characteristics at 2.5 SD below the mean age of onset of puberty (8 years in girls and 9 years in boys) is a complex clinical condition that has considerable physiological and psychological consequences on children and their parents. There are two types - central (gonadotrophin-dependent) and peripheral (gonadotrophin independent). Among the causes of central precocious puberty are various genetic mutations, syndromes and central nervous system disorders that prematurely activate the hypothalamic pituitary gonadal axis leading to the

The complex interaction between obesity and puberty and its implications for pubertal timing: a review of recent evidence.

The childhood and adolescent obesity epidemic remains one of the most important and challenging health issues of the 21st century. Excess body weight affects the immediate health of children and adolescents, being associated with a higher and earlier risk of several non-communicable diseases (NCDs), such as type 2 diabetes and cardiovascular diseases, in addition to adverse psychosocial consequences and impacts on pubertal timing. Evidence dating back to the past century points to a secular trend of earlier puberty in boys and girls in various parts of the world, and this earlier onset has bee

Central Precocious Puberty and Optic Pathway Glioma in Children with Neurofibromatosis 1: Associations with Tumor Location, Vision, and Treatment.

Neurofibromatosis 1-associated optic pathway gliomas (OPGs) may co-occur with central precocious puberty (CPP). Using large language model-based data analysis from 2 NF centers, we compared OPGs in children with NF1, with and without CPP. CPP was associated with hypothalamic involvement, poorer vision, and a higher likelihood of OPG treatment.

Bone age advancement in non-obese children with premature adrenarche: relationship to growth acceleration and predicted adult height outcomes.

OBJECTIVES: Premature adrenarche (PA) is commonly associated with bone age (BA) acceleration, but it is difficult to assess the independent contribution of adrenal androgens to skeletal maturation in studies that include obese children. This study examined BA advancement in non-obese children with PA and evaluated predicted adult height (PAH) estimates in this population. METHODS: The medical records of children diagnosed with PA between April 2022 and April 2023 were reviewed retrospectively. Children who were obese, had congenital adrenal hyperplasia, central precocious puberty, genetic or

A Review of Pituitary Duplication and First Report of Associated Precocious Puberty in a Boy.

INTRODUCTION: Pituitary duplication is a rare congenital malformation, with fewer than 80 cases reported in the literature. It is often associated with midline malformations but can also occur in isolation. Central precocious puberty (CPP) is the most common endocrinological manifestation, but to date this has only ever been reported in female patients. CASE REPORT: A 9-year-old boy presented with precocious puberty. His medical history was notable for a ventricular septal defect. Physical examination showed Tanner stage P3 G2, a growth rate of 10 cm/year, and a bone age of 12.5 years. A GnRH
