Primary Amenorrhea Revealing Adamantinomatous Craniopharyngioma Associated With Central Hypogonadotropic Hypogonadism in an Adolescent Girl: A Case Report.

Adamantinomatous craniopharyngioma is an uncommon benign epithelial tumor arising from remnants of Rathke's pouch within the sellar-suprasellar region. Although patients typically present with headache, visual impairment, growth retardation, or multiple endocrine abnormalities, primary amenorrhea as the initial clinical presentation is unusual, particularly in the absence of neurological symptoms. We report the case of a 15-year-old adolescent girl who presented with primary amenorrhea and delayed pubertal development without headache, visual complaints, or other focal neurological deficits. Recurring and isolated intronic FGFR1 variants as an underdiagnosed cause of congenital hypogonadotropic hypogonadism.

BACKGROUND: Congenital hypogonadotropic hypogonadism (CHH) is a genetically heterogeneous disorder characterized by absent or delayed puberty and infertility due to gonadotropin-releasing hormone deficiency. While loss-of-function variants in FGFR1 are the most commonly known genetic cause of CHH, diagnostic screening typically focuses on coding exons, potentially overlooking pathogenic variants in noncoding regions. METHODS: We investigated 4 unrelated families with CHH. Family 1 included a mother and 3 daughters presenting with variable reproductive phenotypes and incomplete penetrance of a

Hydrocephalus-related amenorrhoea: A scoping review.

STUDY OBJECTIVE: Hydrocephalus is a rare, potentially reversible cause of gonadotropic dysfunction, amenorrhoea, and delayed puberty; fewer than 60 cases have been reported. This scoping review aimed to characterize hydrocephalus-associated amenorrhoea, assess time to postoperative menstrual (re)occurrence, and identify factors influencing recovery. METHODS: This scoping review followed Joanna Briggs Institute methodology, with a systematic search of PubMed, Embase, Google Scholar, and Web of Science for cases of amenorrhoea associated with hydrocephalus. Two reviewers independently screened

Endocrine dysfunctions in patients with beta thalassemia major: a single-center retrospective study from Basrah.

BACKGROUND: The management of patients with beta thalassemia major (BTH) involves regular blood transfusions which are associated with iron overload. This may result in various endocrine dysfunctions (EDs). This study aimed to assess the obesrved frequency of different EDs in patients with transfusion-dependent BTH. METHOD: A retrospective study that included 496 patients with BTH who were referred from the Basra Center for hereditary blood diseases to Faiha Specialized Diabetes, Endocrine and Metabolsim Center for endocrine-related complaints and underwent detailed endocrine assessments from

Disappearance of Posterior Pituitary Hyperintensity in Arginine Vasopressin Resistance: A Case Report.

The evaluation of arginine vasopressin resistance (AVP-R) and arginine vasopressin deficiency (AVP-D), respectively, previously referred to as nephrogenic and central diabetes insipidus, often includes brain magnetic resonance imaging (MRI). Its purpose is to assess pituitary structural changes and identify the physiological T1 hyperintensity of the posterior pituitary, present in most individuals. This hyperintensity reflects vasopressin storage, which shortens the T1 relaxation time. Its absence supports a diagnosis of AVP-D over AVP-R. A 13-year-old boy was referred to endocrinology in his

Multisystem developmental abnormalities and insulin resistance in ADTKD-HNF1B with the p.E105K variant: A clinical conundrum.

AIMS: HNF1B variants cause autosomal dominant tubulointerstitial kidney disease (ADTKD-HNF1B), but the phenotypic spectrum of variants of uncertain significance (VUS) remains incompletely defined. We aimed to characterise the multisystem phenotype associated with the HNF1B p.E105K variant and to examine the mechanism underlying insulin resistance in this setting. METHODS: A 15-year-old male proband with chronic kidney disease and multisystem developmental abnormalities underwent clinical, biochemical and genetic evaluation, including trio-based whole-exome sequencing. The identified variant w

An Evaluation of Language Preference between Spanish and English with Picture Exchange Users.

BACKGROUND: Puberty is a crucial developmental milestone that involves significant physiological, emotional, and behavioral changes. Early puberty onset, influenced by both biological and social factors, is associated with an increased risk of engaging in substance use, such as tobacco and marijuana. While high family income is generally linked to delayed puberty onset and lower behavioral risks, these benefits may not be equally protective for Black youth due to the phenomenon of Minorities' Diminished Returns (MDRs). MDRs suggest that higher family income does not offer the same protective e

Artesunate promotes liver regeneration by modulating mitophagy via the AMPKα1/2/mTOR signaling pathway.

BACKGROUND: Occupational, inflammatory skin diseases are a common work-related disease. Often, these diseases are irritative and/or allergic hand eczema. Affected groups include, for example, nursing staff and metal workers. Individual outpatient and inpatient prevention programs aim at improving self-management of the disease (i.e., improving skin protection behavior) and preventing affected workers from job loss due to their illness. The common-sense model is a widely used framework to explain the patients' illness perceptions of their disease. Qualitative research methods are appropriate me

Reversal of Congenital Hypogonadotropic Hypogonadism.

CONTEXT: Congenital genetic disorders have been traditionally considered to be lifelong. An exception to this long-held view is the reversal of congenital hypogonadotropic hypogonadism (CHH). Approximately 10% of male individuals with CHH undergo reversal with sustained hypothalamic-pituitary-gonadal (HPG) axis activation and/or fertility after discontinuing hormonal treatment. EVIDENCE ACQUISITION: We conducted a structured, systematic literature search to identify relevant articles published on reversal of CHH in males (up to 2025). This mini-review provides a concise overview and synthesiz

Haploinsufficiency of Sox2 causes fewer GnRH neurons and delayed puberty in mice.

Mutations in the SOX2 gene have been previously linked to a syndromic form of isolated hypogonadotropic hypogonadism, with additional ocular and neurodevelopmental phenotypes. Recently, we reported a functional role for SOX2 in hypothalamic kisspeptin-expressing neurons and established a mechanistic relationship between SOX2 heterozygous variants and isolated hypogonadotropic hypogonadism. To further test the role of Sox2 in the hypothalamic-pituitary-gonadal axis, we generated mice with a whole-body heterozygous knockout of Sox2 (Sox2WT/KO). We found that heterozygous loss of Sox2 significant

Transcriptomics identified crucial candidate genes and pathways regulating early testicular development of drakes in the hypothalamic-pituitary-gonadal/thyroid axes.

BACKGROUND: The crosstalk between the hypothalamic-pituitary-gonadal (HPG) and hypothalamic-pituitary-thyroid (HPT) axes plays an important role in gonadal development. However, the mechanism by which these 2 axes regulate early testicular development in drakes remains largely unknown. Therefore, the aim of the present study was to compare the morphology, histology, and transcriptomics between the precocious puberty (PP) and delayed puberty (DP) Longyan Shan-ma drakes (Anas platyrhynchos) to preliminarily reveal the mechanisms regulating early testicular development. RESULT: The histomorpholo

Functional rescue and AI analysis of a human inactivating GPCR mutation using a small molecule.

G protein-coupled receptors (GPCRs) carry out the majority of cellular transmembrane signaling. Many pathologies have underlying GPCR mutations, most of which cause misfolding and GPCR cell surface trafficking failure. Large libraries of existing small molecule GPCR ligands could be repurposed as pharmacological chaperones (PCs) which restore mutant GPCR folding and function, presenting an exciting alternative to complex gene repair, yet such in vivo studies are limited. Therefore, as proof-of-concept, we use one such known ligand/PC, Org42599/Org43553, to show functional rescue in mice bearin

Delayed puberty and early-onset type 2 diabetes risk: a nationwide cohort study of male adolescents in Israel.

BACKGROUND: Delayed puberty has been associated with adverse metabolic outcomes, yet longitudinal evidence on its relation to type 2 diabetes risk is scarce. We aimed to investigate the association between delayed puberty during adolescence and early-adult-onset type 2 diabetes in male adolescents. METHODS: This nationwide, population-based, retrospective cohort study included Israeli male adolescents aged 16-19 years who were examined before military recruitment during 1992-2015 and followed up until Dec 31, 2019. Exclusion criteria were diabetes at the baseline medical assessment, hypogonad

Mauriac Syndrome: Growth and Clinical Outcomes After 2.5 Years of Automated Insulin Delivery Treatment.

Mauriac syndrome is a rare complication of type 1 diabetes mellitus (T1D) with chronically elevated hemoglobin A1C (HbA1c) that is characterized by short stature, delayed puberty, cushingoid features, and hepatic glycogenosis. We report a 14-year-old male patient with T1D managed with multiple daily insulin injections who presented with growth failure and delayed puberty in the setting of several years of HbA1c > 12% (SI: > 108 mmol/mol) (reference range, < 5.7% [SI: < 39 mmol/mol]). Paternal obesity by lactation overnutrition induces sex-related metabolic and reproductive changes in their offspring.

The prenatal period, childhood, and adolescence are critical periods of development characterized by high plasticity. As an extension of the Developmental Origins of Health and Disease (DOHaD) paradigm, known as Origins of Paternal Health and Disease (POHaD), recent studies in rodents provide evidence that paternal obesity is associated not only with infertility but also with an increased risk of metabolic disorders in the offspring. In rodents, litter size reduction is used to induce lactational overfeeding by increasing the amount of breast milk to pups, which causes metabolic and reproducti

Clinical Presentation and Management of Swyer Syndrome: A Case Report.

A pure 46, XY gonadal dysgenesis, or Swyer syndrome, is an extremely rare condition causing primary amenorrhea. It is distinguished by the existence of a female phenotype with a 46, XY karyotype. Reports of new cases are essential to improve early diagnosis and management. We present a case of a 16-year-old Moroccan girl with pubertal delay. The patient exhibited a female, partially developed morphology with Tanner stage 2 secondary sexual characteristics, and a hormonal profile consistent with hypergonadotropic hypogonadism. Morphological evaluation revealed the presence of a uterus and vagin

Nanoplastics Impair GnRH Neuron Migration and Neuroendocrine Function: Emerging Players in the Pathogenesis of Reproductive Disorders.

Nanoplastics (NPs) pose an emerging threat to environmental and human health. Still, the impacts of NPs on the endocrine control of reproduction remain poorly understood, despite increasing trends of infertility worldwide. In mammals, reproductive function relies on the hypothalamus-pituitary-gonadal axis, centrally regulated by gonadotropin-releasing hormone (GnRH) neurons. Disruption in GnRH neuron development or function leads to GnRH deficiency (GD), a genetic condition presenting delayed puberty and infertility. Yet, genetic causes explain only ∼50% of GD cases, suggesting a role for envi

Human Sexual Development in the Somatic and Psychosexual Context.

Human sexual development begins in the prenatal period and continues throughout life, shaped by both biological and psychosocial factors. Somatic development leads to reproductive maturity through several stages regulated by the hypothalamic-pituitary-gonadal axis. Psychosexual development, described in classical theories such as Freud's and in contemporary models, emphasizes the development of gender identity and sexual behaviors from infancy through early and middle childhood into adolescence, a period characterized by the integration of sexual and emotional components. This developmental tr
