SOX10 Mutation of Waardenburg Syndrome With Hypogonadism: A Report of 2 Cases.

Waardenburg syndrome (WS) is a complex genetic disorder primarily characterized by auditory and pigmentary abnormalities, resulting from neural crest cell migration disorders. We reported 2 genetically confirmed SOX10-mutant WS cases illustrating critical management principles. Both patients underwent early auditory intervention (case 1: cochlear implantation at age 3; case 2: hearing aids from age 2), resulting in preserved age-appropriate language acquisition. Each case manifested delayed puberty with biochemical evidence of hypogonadotropic hypogonadism, necessitating gonadotropin therapy t

Compound Heterozygous Hemoglobin E-Beta (HbE-β)-Thalassemia Presenting With Chipmunk or Rodent Facies, and a Severe Thalassemia Major Phenotype.

Hemoglobin E-beta (HbE-β)-thalassemia is a common compound hemoglobinopathy in South and Southeast Asia and is marked by considerable clinical heterogeneity, ranging from mild anemia to a severe, transfusion-dependent condition resembling β-thalassemia major (β-TM). The severity of disease is largely determined by the nature of the associated β-thalassemia mutation, modifiers of fetal hemoglobin, and access to early diagnosis and regular treatment. Severe skeletal deformities are now uncommon in the era of structured transfusion programs, but continue to be encountered in regions with delayed

Familial WT1-associated nephropathy - 46, XY Frasier syndrome and 46, XX steroid-resistant nephrotic syndrome in female siblings: A case report and review of literature.

BACKGROUND: The Wilms tumor 1 (WT1) mutation is significantly associated with steroid-resistant nephrotic syndrome (SRNS) in children. It causes a spectrum of disorders, including Frasier and Denys-Drash syndromes. The WT1 gene encodes a transcription factor essential for renal, gonadal, and mesothelial development. The prevalence of Frasier syndrome is 1 in 1000000. Frasier syndrome presents with childhood onset of SRNS, progressing to end-stage kidney disease, has an XY female phenotype, delayed puberty, and gonadal tumor risk. Two siblings with identical WT1 mutations but differing phenotyp

Cadmium-Induced Neuroendocrine Alterations: Gene Expression of the Kisspeptin-GnRH Axis and Delayed Puberty in Male Rats.

Puberty is a neuroendocrine process required for sexual maturity; it is regulated by the hypothalamic-hypophysis-gonadal (HHG) axis. Kisspeptin (KISS1) plays a vital role in activating this axis by stimulating the secretion of gonadotropin-releasing hormone (GnRH). Cadmium (Cd) exposure disrupts KISS1 signaling in female rodents; its effects on hypothalamic gene expression during male puberty remain poorly understood. This study investigated the effects of Cd exposure on hypothalamic Kiss1, Kiss1r, and Gnrh1 expression, preputial separation (PS) as a marker of pubertal onset, testosterone leve

Multinucleation and cytotoxicity induced by multikinase inhibition in highly proliferative cells.

Phenotypic screening using motor neurons derived from the induced pluripotent stem (iPS) cell of a patient with amyotrophic lateral sclerosis led to the identification of Compound X, a novel agent with potent neuroprotective activity that is hypothesized to act by inhibiting hematopoietic progenitor kinase/germinal center kinase-like kinase (HGK). Unlike known HGK inhibitors, including Prostetin and GNE-495, Compound X and related Compounds Y and Z exhibited broader multi-kinase-inhibition profiles, including the strong inhibition of Aurora B kinase and strong-to-moderate inhibition of Src-fam

Growth Disturbances in Children With Rheumatologic Diseases.

Growth impairment is a well-recognized complication in pediatric rheumatologic diseases, particularly juvenile idiopathic arthritis, systemic lupus erythematosus, and systemic sclerosis. These conditions interfere with normal childhood development through the combined influences of chronic inflammation, altered endocrine signaling, malnutrition, delayed puberty, reduced physical activity, and medication toxicity-especially glucocorticoids. Although the advent of biologic therapy has dramatically improved disease control and enhanced growth potential, many children continue to exhibit significa

Puberty and Growth in Cystic Fibrosis: A Review.

Cystic fibrosis (CF) is a chronic disease process that has historically been associated with delayed puberty and growth. Mechanisms for delayed growth and puberty in chronic disease include malnutrition, inflammation, alterations of the growth hormone axis, chronic corticosteroid use, and changes to gonadotropin signaling or release. Early evidence suggested that people with CF have delayed puberty, decreased peak height velocity, and decreased final adult height compared to the general population. Management of delayed puberty in CF typically involves addressing the underlying cause of delay,

Cystic Fibrosis-Related Bone Disease in Youth: High-Risk Population With Low Screening Adherence.

OBJECTIVES: Cystic fibrosis (CF)-related bone disease (CFBD) is characterized by low bone mineral density and increased fracture risk. The U.S. CF Foundation recommends using dual-energy x-ray absorptiometry (DXA) to screen all adults and at-risk children 8 to <18 years. We aimed to establish the proportions of children who meet CFBD screening criteria, are ordered a DXA, and complete DXA screening. METHODS: The medical charts of children seen in our pediatric CF clinic in 2022 were reviewed to identify who met criteria for CFBD screening by DXA per CF Foundation guidelines: prior fracture, d

Relationship between kisspeptin-10, neurokinin B and dynorphin A in the course of normal and delayed puberty in ewes.

INTRODUCTION: Kisspeptin (KiSS), neurokinin B (NKB) and dynorphin A (Dyn A) participate in the neuroendocrine regulation of reproductive system development and functioning; however, their roles in the onset of sexual maturity and pathomechanism of delayed puberty have not yet been fully elucidated. The aim of the study was to determine changes in the kisspeptin-10 (KiSS-10), NKB and Dyn A concentration in blood plasma collected from ewe lambs during puberty in relation to the first ovulation time, and to examine the relationship between these neuropeptides. MATERIAL AND METHODS: Twenty-four e

Caring for Gender Diverse Youth With Duchenne Muscular Dystrophy: A Multisite Case Series.

BACKGROUND: Duchenne muscular dystrophy (DMD) is a progressive neuromuscular condition requiring long-term multidisciplinary care. Endocrine management often includes pubertal induction with testosterone for delayed puberty, which commonly occurs in individuals receiving chronic glucocorticoid therapy. Although gender diversity is increasingly recognized among adolescents, gender identity and gender-affirming care have not been well described among individuals with DMD. METHODS: We conducted a multi-institutional, retrospective case series across pediatric neuromuscular centers in the United

Severe enduring anorexia nervosa with significant height increase in adulthood: A case report.

BACKGROUND: Anorexia nervosa (AN) is a severe psychiatric disorder characterized by restrictive eating, low body weight, and distorted body image, and is often complicated by endocrine dysfunction. While hypogonadism and delayed puberty are well documented in female patients, reports in males remain scarce because of the lower prevalence of AN in this group. Hypogonadism may result in impaired pubertal development, growth arrest, and reduced testicular volume in male patients. Although weight restoration can sometimes enable catch-up growth when AN begins before puberty, the long-term potentia
