Aspartame consumption linked to delayed puberty and mitochondrial Dysfunction: Evidence from human and animal studies.

This study investigates the impact of aspartame consumption on pubertal timing in females. The research employs both human and rat models to explore underlying mechanisms. In the Taiwan Pubertal Longitudinal Study (2018-2022), 858 girls aged 6-12 were assessed for aspartame intake and puberty outcomes. Concurrently, female Sprague Dawley rats were exposed to low (30 mg/kg) or high (60 mg/kg) doses of aspartame from prenatal to postnatal stages. Results demonstrate that aspartame disrupts the hypothalamic-pituitary-gonadal (HPG) axis in rats, causing mitochondrial dysfunction and reduced ovaria

Taking care of patients with recessive dystrophic epidermolysis bullosa from birth to adulthood: a multidisciplinary Italian Delphi consensus.

BACKGROUND: Recessive dystrophic epidermolysis bullosa (RDEB) is a rare and severe mucocutaneous fragility disorder due to mutations in the COL7A1 gene encoding collagen VII, the major constituent of anchoring fibrils essential for epithelial adhesion. RDEB is characterized by unremitting blistering, chronic painful wounds and fibrotic scarring that results in hand and foot pseudosyndactyly, microstomia, and esophageal strictures. RDEB complications include nutritional compromise, chronic anemia, failure to thrive, delayed puberty, osteoporosis, and renal involvement. In addition, early onset

Transcriptomic profiling of murine GnRH neurons reveals developmental trajectories linked to human reproduction and infertility.

Rationale: Neurons producing Gonadotropin-Releasing Hormone (GnRH) are essential for human reproduction and have to migrate from nose to brain during prenatal life. Impaired GnRH neuron biology results in alterations of the reproductive axis, including delayed puberty and infertility, with considerable effects on quality of life and metabolic health. Although various genes have been implicated, the molecular causes of these conditions remain elusive, with most patients lacking a genetic diagnosis. Methods: GnRH neurons and non-GnRH cells were FACS-isolated from mouse embryo microdissections to

A Leptin Receptor Mutation Which Impairs Fertility in Ewes Causes Delayed Puberty in Male and Female Mice.

Reproductive function is tightly linked to nutritional status due to its high energetic demands. Leptin, a key adipose tissue-derived hormone signalling energy reserves to the brain, integrates metabolic status with the hypothalamic-pituitary-gonadal axis to ensure reproductive function is maintained or suppressed appropriately. Mutations in leptin or its receptor (LepR) are known to cause infertility and obesity in mice. In Davisdale ewes, 2 naturally occurring LepR mutations (R62C and P1019S) were associated with delayed puberty and subfertility, but their effects in males or in other specie

Influence of neurokinin B, dynorphin A and kisspeptin-10 on in vitro gonadotropin secretion by anterior pituitary cells isolated from pubescent ewes.

INTRODUCTION: The hypothalamic neuropeptides kisspeptin-10 (KiSS-10), neurokinin B (NKB), and dynorphin A (Dyn A) play roles in the endocrine regulation of the hypothalamic-pituitary-ovarian (HPO) axis in puberty. Livestock's timely attainment of sexual maturity increases reproductive efficiency and raises profitability. The pituitary relationship between these neuropeptides and gonadotropins in puberty in ewes being undercharacterised. The aim of the study was to analyse their direct effect on gonadotropin secretion by pituitary cells isolated from pubescent ewes. MATERIAL AND METHODS: Cells

Turner syndrome: the promise of fertility via stem cell technology.

Turner syndrome (TS) is the most common female sex chromosome disorder, occurring in one out of every 2500 to 3000 live female births. It is caused by the partial or complete loss of one X chromosome. TS is associated with certain physical and medical features, including short stature, estrogen deficiency, delayed puberty, hypothyroidism, and congenital heart defects. The majority of women with TS are infertile as a result of gonadal dysgenesis and primary ovarian insufficiency causing hypergonadotropic hypogonadism. Several reproductive options are available for TS patients. The recent use of

Central lipid sensing pathways contribute to the control of puberty and its alterations in conditions of obesity.

Childhood obesity, especially in girls, often correlates with advanced puberty and long-term comorbidities. Among the central circuits controlling energy homeostasis, hypothalamic lipid sensing pathways, involving free fatty-acid receptors (FFARs), peroxisome proliferator-activated receptors (PPARs), and the bile-acid (BA) receptor, Takeda G protein-coupled receptor 5 (TGR5), have been recognized as major players, with putative pathogenic roles in obesity and its complications. However, their contribution to pubertal regulation and obesity-induced pubertal alterations remains largely unexplore

Deleterious variants in intolerant genes reveal new candidates for self-limited delayed puberty.

OBJECTIVE: Self-limited delayed puberty (SLDP) is the most common cause of delayed puberty and exhibits high heritability, although few causal genes have been identified. This study aims to identify potential candidate genes associated with SLDP. METHODS: Whole-exome sequencing was conducted in 71 children with SLDP, most of whom presented with short stature. Rare coding variants were prioritized through comprehensive bioinformatics analyses and classified as high-impact or moderate-impact based on predicted functional effects. Candidate genes were selected based on the absence of human pheno

Challenges in Gaucher disease: Perspectives from an expert panel.

This focused review concentrates on eight topics of high importance for Gaucher disease (GD) clinicians and researchers: 1) The consideration of GD as distinct types rather than a spectrum. A review of the literature clearly supports the view that there are distinct types of GD. Type 1 is characterized by the absence of primary neuronopathic involvement, while types 2 and 3 are characterized by progressive primary neuronopathic disease. 2) Neurologic and neuronopathic manifestations. A growing body of evidence indicates that the peripheral nervous system may be involved in GD type 1 and that t

Corticosteroid-binding Globulin Dysfunction Due to Homozygous SERPINA6 Lyon Variant in a Pediatric Patient.

Corticosteroid binding globulin (CBG) deficiency is a rare disorder with poorly understood pathophysiology and variable presentations. We report on a pediatric patient presenting with poor growth and delayed puberty who was diagnosed initially with primary adrenal insufficiency following 2 failed ATCH stimulation tests with normal CBG and low stimulated salivary (free) cortisol. Genetic testing demonstrated a homozygous SERPINA6 variant known as CBG Lyon. The patient's 2 siblings were later diagnosed with the same homozygous variant but were asymptomatic with normal stimulated salivary cortiso
