BACKGROUND: Mucoepidermoid carcinoma (MEC) is a malignant tumor primarily composed of epidermoid, mucous, and intermediate cells. While it predominantly occurs in the salivary glands, MEC can also develop in the lungs, esophagus, breast, and thymus. Thymic MEC is rare, accounting for 2-3% of primary thymic malignancies. The primary clinical characteristic is incidental discovery in asymptomatic individuals. Due to the rarity of thymic MEC, much of the literature consists of case reports, and no standardized treatment protocol exists. Surgical resection is the primary treatment for MEC.

Good's syndrome (GS) is a rare immunodeficiency disorder associated with thymoma, characterized by hypogammaglobulinemia and recurrent infections; however, its clinical significance in thymoma-associated myasthenia gravis (TAMG) remains unclear. We retrospectively reviewed 30 patients with TAMG admitted to our center between January 2010 and March 2022. We defined GS-like immunodeficiency as serum IgG below the institutional cutoff of 861 mg/dL and a history of two or more infections requiring antimicrobial treatment; 11 patients (36.7%) met this definition. Compared with the remaining patient

BACKGROUND: The necessity and optimal duration of post-thymectomy immunoregulatory medication (IM) in myasthenia gravis (MG) remain unclear.
OBJECTIVES: This study aimed to evaluate whether IM is required after thymectomy in MG patients and to determine the appropriate duration of IM in patients without postoperative deterioration by comparing long-term clinical outcomes.
DESIGN: This triple-center, retrospective study included 1248 MG patients who underwent thymectomy. Among these patients, we compared outcomes between 483 patients without post-thymectomy IM and 765 patients receiving IM.

BACKGROUND: Thymic epithelial tumors presented a variable 5-year survival rate, histological subtype dependent. Efforts have been made to identify new prognostic markers. Neutrophil-to-lymphocyte ratio (NLR), platelet-to-lymphocyte ratio (PLR), and Systemic Inflammatory Index (SII) proved effective in predicting survival in other solid tumors. We evaluated the prognostic significance of NLR, PLR, and SII on disease-free, overall, and tumor-related survivals in patients undergoing surgery with radical intent for thymic epithelial neoplasms.
METHODS: We conducted a retrospective analysis in pat

Myasthenia gravis (MG) is an autoimmune disorder characterized by impaired neuromuscular transmission and motor symptoms. Its genetic background remains unclear, particularly beyond specific subtypes reported in European populations. Here, we perform a genome-wide association study (GWAS) of 1,434 MG cases covering all disease subtypes and 42,913 controls of Japanese, which newly identify the TERT locus (odds ratio [OR] = 1.31, P = 1.7×10-10). Subtype-stratified GWASs show stronger signals for generalized MG (gMG; OR = 1.38, P = 1.6×10-12), anti AChR antibody-positive gMG (g-AChR-Ab(+)MG; OR=

Central to the linkage of pain circuitry with the limbic system is its initial NAα2-mediated antinociceptive effect in acute pain models, followed by contradictory pronociceptive activation by the locus coeruleus seen in chronic pain models. Rats with a stable, long-term (>10 weeks) inflammatory compression of the trigeminal infraorbital nerve (FRICT-ION) preclinical model were given daily doxazosin, a slow-release NAα1 receptor antagonist, in weeks 8-10. Facial hypersensitivity was reversed back to baseline in male and female rats, but anxiety was only reduced in male animals. Doxazosin-decre

Thymic epithelial tumors (TETs) sit at a unique intersection of cancer, central tolerance, and autoimmunity. These tumors arise in the thymus, an immune-rich organ responsible for T-cell education, and despite their low tumor mutational burden (TMB) and microsatellite stability, they display paradoxical behavior under immune checkpoint inhibition. This review synthesizes data on thymic biology, paraneoplastic autoimmunity, molecular profiling, and clinical trials of immune checkpoint inhibitors (ICIs) in thymoma and thymic carcinoma. We describe how disruption of autoimmune regulator (AIRE)/Fe

The thymus is essential for establishing T cell diversity early in life, but undergoes profound involution with age and has therefore traditionally been regarded as largely nonfunctional in adults1,2. Here we propose that preserving thymic functionality is integral to adult health and longevity. We developed a deep learning framework to quantify thymic health from routine radiographic images and evaluated its association with longevity and risk of major age-associated diseases in two large prospective cohorts of asymptomatic adults: the National Lung Screening Trial (n = 25,031) and the Framin

Although immunotherapy has revolutionized cancer treatment, many patients still experience limited benefit, highlighting the urgent need for improved biomarkers1. Although immunotherapy is founded on unleashing T cells2, most existing biomarkers remain tumour-centric and mainly overlook host immune competence. The thymus is a key immune organ that is crucial for T cell maturation, and we hypothesized that thymic functionality is associated with immunotherapy outcomes3. Here we show that thymic health, a radiographic measure of thymic functionality, is strongly associated with immunotherapy out

Morvan syndrome (MoS) is a rare paraneoplastic syndrome that is predominantly characterised by autonomic dysfunction, peripheral nerve hyperexcitability and altered mental status. Cases in literature often describe pre-existing autoimmune diagnoses in patients with MoS, alluding to its association with systemic immune dysregulation. We report a woman presenting with hypersomnolence and fluctuating mental status consistent with MoS who had been previously diagnosed with metastatic thymoma, acetylcholine receptor-positive myasthenia gravis and multiple other chronic medical conditions. The report

INTRODUCTION/AIMS: Thymectomy is associated with positive effects on myasthenia gravis (MG), but there is conflicting evidence regarding potential deleterious long-term outcomes, such as increased cancer and autoimmune disease risk. We aimed to assess these outcomes in thymectomized versus non-thymectomized MG patients.
METHODS: We retrospectively reviewed the medical records of patients with acquired, autoimmune MG followed at the Prosserman Neuromuscular Clinic, University Health Network (UHN), between January 2000 and July 2025. We excluded patients with malignancy or other autoimmune dise

INTRODUCTION: Trimethyltin chloride (TMT) is a widespread environmental contaminant with significant biotoxicity. Selenium is an essential trace element, and its deficiency can exacerbate pollutant toxicity, with ferroptosis and mitochondrial stress as key events.
OBJECTIVES: This research aimed to explore the influence of combined selenium deficiency (SeD) and TMT exposure on poultry thymus toxicity and the underlying molecular mechanisms.
METHODS: Broiler chicken and MDCC-MSB-1 cell models were established with individual or combined SeD and TMT treatment. Mechanisms of thymic injury were

The thymus plays a critical role in sustaining T-cell immunity, although its function is highly vulnerable to acute injury and physiologically declines with age, resulting in compromised immune responses. Impaired thymic function represents a major clinical challenge, particularly in settings of immunosuppression associated with cancer therapy and aging. Yet, effective strategies to rejuvenate the thymus remain limited. To explore novel regenerative approaches, we focused on FOXN1, a master regulator of thymic epithelial cell (TEC) development and function. By developing a custom screening pla

A growing body of research indicates that systemic inflammation contributes substantially to the progression of Parkinson's disease (PD). Foundational studies propose that targeting inflammatory pathways may offer therapeutic benefits for PD and other neurodegenerative conditions. Our previous work demonstrated that a novel zinc aspartate compound enriched with the light isotope 64Zn (64Zn-asp) can counteract inflammatory and cognitive impairments triggered by intra-hippocampal Aβ1-40 in rats, and can also mitigate neuroinflammation while promoting neuronal survival in a PD model. BACKGROUND: Atopic dermatitis (AD) is an increasingly common skin disease that can lead to serious morbidity in a subset of more severely affected infants. Thymus and activation-regulated chemokine (TARC) and squamous cell carcinoma antigen 2 (SCCA2) are biomarkers. This study examined how severity, defined by Patient-Oriented Eczema Measure (POEM) score, correlates with TARC, SCCA2, total immunoglobulin E (IgE), and eosinophil counts in infantile AD (< 2 years), comparing their diagnostic performance and examining each biomarker's association with individual POEM items.

BACKGROUND: Traditionally, immunology has assumed the dogma that immunological memory is exclusive to cells of the adaptive immune system. However, over the past decade, evidence has challenged this paradigm by demonstrating that innate immune cells can exhibit adaptive characteristics, resulting in long-term functional changes. After exposure to infections or sterile inflammation, innate immune cells can exhibit either an enhanced or suppressed inflammatory response upon secondary challenge. This de facto immune memory has been termed trained immunity or immune tolerance, respectively.

AIMS: To synthesize evidence on the demographic and prognostic profile of surgically treated intrathyroidal thymic carcinoma (ITC).
METHODS: A PRISMA-compliant systematic review and meta-analysis was conducted. Due to the rarity of the disease, all patients with surgically treated ITC were combined from individual case reports to create a cohort of patients which was subsequently pooled with other eligible case series. Single-arm meta-analysis was used to synthesize a demographic profile and Kaplan-Meier statistics were used for survival analyses.
RESULTS: Analysis of 55 articles (154 patien

The division of the mediastinum into specific compartments is essential for identifying, characterizing, and managing mediastinal abnormalities. The current international standard is the three-compartment model proposed by the International Thymic Malignancy Interest Group (ITMIG), which divides the mediastinum into prevascular (anterior), visceral (middle), and paravertebral (posterior) compartments. This model has replaced the traditional four-compartment system, which included the superior mediastinum, due to its simplicity, familiarity, and clearly defined boundaries. However, given the su

BACKGROUND AND OBJECTIVE: According to the National Comprehensive Cancer Network (NCCN) and other international guidelines, thymectomy is the standard surgical procedure for early-stage thymic epithelial tumors (TETs). However, the optimal extent of resection remains controversial. Recent evidence suggests that partial thymectomy, which preserves part of the thymus, may achieve outcomes comparable to thymectomy in the short- and long-term. This review aims to compare perioperative and long-term oncologic outcomes between complete and partial thymectomy in early-stage TETs.
