Cardiac adverse events following post-operative radiation therapy in patients with thymic malignancies.

BACKGROUND: Radiation therapy (RT) for thymic malignancies often exposes cardiac structures to ionizing radiation with associated cardiac adverse events risks. Given the curative intent and extended survival of patients with thymic malignancies, identifying modifiable predictors of cardiac risk is essential. While cardiac substructure dosimetry has been studied in other thoracic cancers, its role in thymic malignancies, where the target volume often abuts or overlaps the heart, is less well characterized. METHODS: We retrospectively identified patients treated with surgery and postoperative R

Induction of regulatory T cells by an aluminum-CpG combination adjuvant for therapeutic and preventive vaccination against pollen allergy.

Pollen allergy is a prevalent allergic disorder affecting a substantial proportion of the global population. Allergen-specific immunotherapy (ASIT) is the only disease-modifying treatment, acting by inducing regulatory T cells (Tregs) and Th1-based immunity. However, current ASIT approaches often elicit insufficient allergen-specific tolerance and fail to correct underlying immune dysregulation, thereby limiting clinical efficacy. Here, a combination adjuvant composed of aluminum oxyhydroxide and CpG oligodeoxynucleotides (Al-CpG) is developed and formulated with the T-cell epitope derived fro

Smartwatch-Derived Nocturnal Scratching Metrics Capture Disease Activity and Severity in Pediatric Atopic Dermatitis.

Background/Objectives: The itch-scratch cycle is a key driver of exacerbation in atopic dermatitis (AD) and requires objective monitoring, yet patient-reported itch scores are often unreliable in children. This study aimed to evaluate smartwatch-derived nocturnal scratching metrics as digital biomarkers of disease activity and treatment response in pediatric AD. Methods: In this prospective observational study, 50 children (median age 9 years) with physician-diagnosed AD wore an Apple Watch with the Itch Tracker application for 5-14 nights during initiation of topical therapy. Three scratch me

Comparison of anthracycline plus platinum versus other platinum-based regimens as first-line chemotherapy for untreated thymoma: A retrospective cohort study (NEJ023B).

BACKGROUND: Optimal management of advanced thymoma remains uncertain. We compared the efficacy of first-line platinum-anthracycline chemotherapy with other platinum-based regimens in a large Japanese cohort. METHODS: We retrospectively analyzed 157 patients with unresectable or recurrent thymoma treated at 39 institutions (2000-2020). Regimens were categorized as platinum-anthracycline (n = 104) or non-anthracycline platinum (n = 53). Propensity score matching (PSM) balanced baseline characteristics (46 pairs). The primary endpoint was overall response rate (ORR); secondary endpoints were rea

Graft-Versus-Host Disease-like Erythroderma in the Setting of Thymoma: A Rare Paraneoplastic Disorder.

We present a case of graft-versus-host disease (GVHD)-like erythroderma related to thymoma-associated multiorgan autoimmunity (TAMA), a rare paraneoplastic syndrome. A 45-year-old woman with a history of a recent thymoma resection was admitted to the intensive care unit for erythroderma. The erythroderma had progressed over 6 months from a scaling, pruritic rash on bilateral forearms despite treatment with topical corticosteroids. Dermatologic evaluation showed scaly, hyperkeratotic plaques covering greater than 90% of total body surface area. Biopsy taken during admission showed epidermal ver

Rapid 3D Immunolabeling and Light Sheet Microscopy for Quantitative Analysis of Intact Tissues.

Whole-organ 3-dimensional (3D) imaging of intact tissues provides high-resolution cellular and molecular insights into tissue and tumor microenvironments. However, immunolabeling and tissue clearing methods remain complex and time-consuming and often rely on toxic reagents and prolonged processing times. Here, we introduce a rapid 3D immunolabeling and light sheet microscopy platform for quantitative analysis of intact murine organs and human tissues within 2 to 2.5 d. This streamlined workflow integrates antigen retrieval, permeabilization, collagenase-based digestion, immunolabeling, dehydra

Postoperative radiotherapy improves overall survival and was not associated with a statistically significant increase in cardiac-specific mortality in thymoma patients: a propensity score-matched analysis of the SEER database.

BACKGROUND: Thymoma is an uncommon cancer of the thymus and postoperative radiotherapy (PORT) is often considered for selected patients with high-risk or locally advanced thymoma after resection. The effect of PORT on overall survival (OS) and cardiac-specific mortality in patients with thymoma is not well understood. The purpose of this study was to assess the impact of PORT on OS and cardiac-specific mortality in resected thymoma patients. METHODS: We analyzed the data from Surveillance, Epidemiology, and End Results (SEER) database to identify patients with thymoma who had a cancer-directe

The compound of microencapsulated essential oils improves productive parameters, antioxidant status, meat quality, and both humoral and cellular immune responses in broilers.

This experiment investigated the effect of the compound of microencapsulated essential oils (CMEOs) on productive parameters, antioxidant status, meat quality, and humoral and cellular immune responses in broilers. In a completely randomized design, a total of 640 (mixed-sex) one-day-old Ross 308 broiler chickens were divided into 4 treatments with 10 replicates (16 chicks/replicate) for 42 days. Treatments were basal diet supplemented with CMEOs at levels of 0 (control), 300 (CMEOs-300), 600 (CMEOs-600), and 900 (CMEOs-900) mg/kg of diet. The CMEOs contain equal weight ratios of eucalyptus, o

Thymic alveolar type 2 epithelial mimetic cells revealed by RUNX1 deficiency.

Autoimmune diseases can affect the lungs, and the mechanisms used to prevent autoimmune lung disease are incompletely understood. Recent studies in the thymus have identified unique populations of medullary thymic epithelial cells (mTECs) called mimetic cells that transcriptionally mimic peripheral epithelial populations. These mimetic cells have important functions in the thymus in immune tolerance. Here, we used a mouse line with thymic-specific deletion of Runt-related transcription factor 1 (Runx1) to identify a new mimetic cell akin to alveolar type 2 (AT2) lung epithelial cells. AT2 mTEC

Quercetin reverses DEHP-induced ZBP1-mediated PANoptosis in thymus by targeting NF-κB to regulate the TNF-α/ROS signaling pathway.

Quercetin (Que), a natural dietary flavonoid, possesses significant antioxidative and immunoregulatory properties. Di(2-ethylhexyl) phthalate (DEHP), a pervasive environmental endocrine-disrupting chemical, triggers oxidative stress (OS) and elicits immunotoxic effects in various species. However, whether Que can target NF-κB to alleviate DEHP-induced OS and PANoptosis in the chicken thymus remains unclear to date. To investigate the mechanism of thymus damage induced by DEHP and the intervention effect of Que, we established an in vivo chicken model treated with DEHP and/or Que. Based on the

The Heart of the Thymus: A Rare Tale of Nonischemic Cardiomyopathy in the Absence of Myasthenia Gravis.

Thymomas are rare epithelial tumors of the anterior mediastinum known for their association with autoimmune and paraneoplastic syndromes, most notably myasthenia gravis (MG). Cardiomyopathy is an exceptionally rare paraneoplastic manifestation. Emerging case reports suggest that thymoma may exert systemic effects independent of MG, including direct or immune-mediated myocardial injury. We report the case of a 54-year-old male with known nonischemic cardiomyopathy who presented with septic shock from pneumonia and cholecystitis, complicated by ventricular arrhythmias and decompensated heart fai

The multifaceted functions of STING in CD4+ T cell biology.

The stimulator of interferon genes (STING) protein has centered on its role as a transmembrane receptor in professional innate immune cells, mediating type I interferon (IFN-I) signaling. However, recent advancements have significantly revealed its marked expression and functional importance in adaptive immune cells, particularly CD4+ T lymphocytes. STING signaling intersects with crucial T cell pathways, influencing development, activation, survival, and differentiation. This review synthesizes current knowledge on the dynamic regulation and multifaceted roles of STING in CD4+ T cell biology.

Sodium-Glucose Linked Transporter-2 (SGLT-2) Inhibitor-Associated Unmasking of Myasthenia Gravis in a Patient With Undiagnosed Thymic Follicular Hyperplasia: The First Reported Case.

Myasthenia gravis is an autoimmune disease of the neuromuscular junction, primarily caused by antibodies directed against the acetylcholine receptor. Follicular hyperplasia of the thymus can be seen in patients with acetylcholine receptor-positive myasthenia gravis and may remain asymptomatic for many years until unmasked by a specific medication. Myasthenia gravis induced by drugs has been reported with D-penicillamine and immune checkpoint inhibitors, but never with the sodium-glucose linked transporter-2 inhibitors. We report a case of a 62-year-old man with type 2 diabetes presenting with

mTECs and B cells form a thymic microniche associated with B-cell licensing.

Thymic B cells rely on T cells and type-III IFN (IFN-λ) signaling for class switch recombination and effective licensing as APCs, a process essential for their role in establishing central T-cell tolerance. IFN-λ is produced exclusively by a subset of medullary thymic epithelial cells (mTECs); however, the spatial layout between B cells and IFN-λ-producing mTECs remain unclear. Here, we studied the distribution of thymic B cells and IFN-λ+ mTECs, alongside other mTEC populations, to better understand the niche required for thymic B-cell licensing. Using 26 target multiplex immunofluorescence i

Human interferon-ω: an underappreciated type I interferon.

Interferon-ω (IFN-ω) is a member of the human type I interferon family that has historically been overshadowed by IFN-α and IFN-β. Recent human "natural perturbations", most notably selective neutralization of IFN-ω by autoantibodies in life-threatening viral infections, have renewed interest in this comparatively understudied cytokine and indicate that its antiviral activity may not always be fully compensated in defined clinical settings. This renewed focus has prompted reassessment of its single-gene organization, distinct antigenicity, intermediate receptor-binding kinetics, cellular sourc

Ly6d expression delineates two putative postnatal thymus epithelial progenitor cells that are differentially affected by aging.

To date, the identity and maintenance of postnatal thymic epithelial progenitor cells (TEPCs) remain unclear, as does the persistence of bipotent TEPCs after birth or whether lineage-restricted progenitors independently maintain separate TEC compartments. Using an inducible lineage-tracing system based on expression of the thymoproteasomal protein β5t, which is expressed in embryonic and a subset of postnatal TEPCs, we explored the early dynamics of the relationships between thymic epithelial cell (TEC) progenitors and their progeny. Our results identified two potential lineage-biased progenit

TRECing down the source: naive and memory T cell generation in dirty mice and men.

Healthy aging relies on the maintenance of a diverse T cell pool. This diversity is ensured by balancing thymic output, differentiation of naive into memory T cells, T cell proliferation and cell death. For naive T cells, the balance of these processes differs between standard laboratory mice and humans. This may be a true species difference or, alternatively, result from the vastly different amounts of antigens to which standard laboratory mice and humans are exposed. Using wildlings, that is, laboratory mice born to wild mice, we studied the impact of antigen-exposure through a natural micro

Limited Clinical Associations of Age-Adjusted Serum Thymus and Activation-Regulated Chemokine in Kawasaki Disease.

BACKGROUND: Kawasaki disease is an acute systemic vasculitis, and the clinical significance of serum thymus and activation-regulated chemokine (TARC) remains incompletely understood. We examined whether pre-treatment age-adjusted serum TARC was associated with inflammation-based intravenous immunoglobulin (IVIG) resistance risk stratification. METHODS: We retrospectively studied 109 children with Kawasaki disease after excluding documented allergic comorbidities and age younger than 6 months. All included patients had serum TARC measured before initial treatment, and 18 of them also had paire

Cytokine concentrations in pediatric primary mediastinal large B cell lymphoma correlate with disease extent and outcome.

Primary mediastinal large B-cell lymphoma (PMBCL) predominantly affects female adolescents and young adults. It displays an immune-privileged phenotype and demonstrates the involvement of key cytokine signaling pathways, including increased expression of Thymus and activation-regulated chemokine (TARC/CCL17). Currently, there is a lack of established markers for stratification. We studied plasma concentrations of 24 cytokines at diagnosis in a large population-based pediatric cohort of 62 patients with PMBCL. Compared to a group of age-matched patients with other types of lymphoma in long-term
