Immune Checkpoint Therapy for Thymic Carcinoma.

Thymic carcinoma (TC) is a rare, aggressive cancer that originates from thymus's epithelial cells. It distinguishes itself from other thymic epithelial tumors with its unique pathological structure, clinical behavior, and immune characteristics. Immune checkpoint inhibitors (ICIs) targeting the Programmed cell death protein 1/Programmed cell death protein ligand 1 (PD-1/PD-L1) pathway have shown promise in advanced TC, potentially benefiting from frequent PD-L1 overexpression and abundant CD8+ tumor-infiltrating lymphocytes (TILs), despite typically low tumor mutational burden (TMB). While ICI

Thymus research in relation to myasthenia gravis: a new perspective on cell subpopulations and future directions.

The thymus generates T cells from immature thymocytes and prevents autoimmune diseases through negative selection and the generation of FOXP3+ regulatory T cells (Tregs). The thymic architecture is typically divided into two distinct microenvironments, the cortex and the medulla. These microenvironments are characterized by the presence of cortical thymic epithelial cells (cTECs) and medullary thymic epithelial cells (mTECs), respectively. Recent single-cell and spatial transcriptomic analyses have revealed the expanding diversity of TEC subpopulations in mice and humans. Myasthenia gravis (MG

Mesenchymal thymic niche cells enable regeneration of the adult thymus and T cell immunity.

Thymic atrophy and the progressive immune decline that accompanies it is a major health problem, chronically with age and acutely with immune injury. No definitive solution is available. Here we demonstrate that one of the three mesenchymal cell subsets identified by single-cell analysis of human and mouse thymic stroma is a critical niche component for T lymphopoiesis. The Postn+ subset is perivascular, and its depletion abrogates T cell progenitor recruitment, likely through production of the chemokine Ccl19. It markedly declines with age and in the acute setting of hematopoietic stem cell t

Battery pollutant leakage disrupts antioxidant ability and gut microbial homeostasis of chickens.

Over the past few decades, battery industry and electronic equipment have undergone explosive growth, but the heavy metal waste generated has led to significant global ecological and public health challenges. Currently, increasing evidences have confirmed the detrimental effects of heavy metal exposure on animal reproduction, immunity, and metabolism. However, research focused on the impacts of battery leakage on the gut microbiota remain scarce. Thus, this study aims to investigate the detrimental effects of battery on gut microbiota in chickens. Results revealed that battery exposure can lea

Significance of pathological changes and chemokine expression in various organs in a Graves' disease animal model.

Graves’ disease (GD) is the most common autoimmune disease characterized by the loss of immune tolerance induced by the activation of target antigens by the thyrotropin receptor (TSHR) autoantibody. Chemokines may be involved in the autoimmune inflammatory response in GD. In this study, we investigated the mechanism of chemokine expression in different tissues of GD hyperthyroid mice. Adenoviruses encoding the TSHR A subunit were employed to immunize female BALB/c mice to induce GD. HE-stained sections were used to observe the histopathological morphology, and RT-qPCR and immunohistochemistry

Lung-Targeting Cepharanthine Polymer Micelles Modified with Mannose: Effectiveness Against Acute Lung Injury Evaluated Using in vitro and in vivo Analyses.

<p>Introduction: The currently available therapies for acute lung injury (ALI), including gluco-corticoids, protease inhibitors, and heparin, have limited clinical efficacy and are often associated with significant side effects. Cepharanthine (CEP) has demonstrated effectiveness in treating pulmonary dis-eases, but its clinical application is restricted by low solubility and poor bioavailability. This study aimed to develop mannosylated cepharanthine-loaded polymeric micelles (MA-CEP-PMs) to improve CEP bio-availability and enhance lung-targeted delivery for the treatment of ALI. </p><p> Metho

Broadening the Phenotypic Spectrum of Forkhead Box N1 Gene Mutations.

A man in his 50s develops signs of immune dysfunction (recurrent chest infections and new-onset chronic diarrhoea), alongside a history of multiple distinct malignancies and nail dystrophy. Immunological testing reveals T-cell lymphopenia and hypogammaglobulinaemia. Whole genome sequencing reveals a heterozygous c.1465del mutation in the FOXN1 (forkhead box N1) gene. This case suggests that previously healthy carriers of heterozygous FOXN1 mutations may be at risk of developing immune dysfunction in later life. Potential reasons for the severe and delayed phenotype include a dominant-negative

Pathogenicity comparison of NADC34-like porcine reproductive and respiratory syndrome virus in 4-week-old weaned pigs versus 10-week-old growing pigs.

Porcine reproductive and respiratory syndrome virus (PRRSV) is a globally distributed and financially significant viral pathogen affecting the swine industry. Its extensive genotypic and pathotypic heterogeneity among strains makes disease control challenging. Since first reported in 2022, NADC34-like (lineage 1A, L1A) strains have become predominant in South Korea, attracting substantial attention in the domestic swine industry due to their high pathogenicity and considerable economic impact. This study aimed to explore age-dependent disease severity by comparing the pathogenicity of the Kore

Thymic Dendritic Cells Revisited.

Central tolerance in the thymus ensures that the developing T cell repertoire is safe yet effective against infections. This process relies greatly on antigen presentation by both stromal and hematopoietic antigen-presenting cells (APCs), with dendritic cells (DCs) playing a particularly critical role. Thymic DCs acquire a broad spectrum of self-antigens, including tissue-restricted antigens (TRAs), inflammation-associated antigens (ISAs), and peripheral antigens imported via circulation or immigrating DCs. These diverse inputs allow DCs to mediate clonal deletion, regulatory T cell (Treg) ind

Angiogenesis and thymic epithelial tumors: from preclinical insights to different therapeutic lines and combination strategies.

Thymic epithelial tumors (TETs) are rare thoracic malignancies for which surgical resection remains the mainstay treatment, even in advanced stages. For patients with metastatic disease, platinum-based chemotherapy represents the current standard of care in the first-line, with no standard treatment at progression. Much preclinical evidence supports a potential role of angiogenesis in the pathogenesis and progression of TETs. This biological rationale has led to assess the efficacy of several antiangiogenic agents, either in monotherapy or in combination in pre-treated metastatic TET, reportin

A Thymus-Independent Artificial Organoid System Supports Complete Thymopoiesis from Rhesus Macaque-Derived Hematopoietic Stem and Progenitor Cells.

Background: T cell regeneration in the thymus is intrinsically linked to the T cell-biased lineage differentiation of hematopoietic stem and progenitor cells (HSPCs). Although nonhuman primates (NHPs) serve as indispensable models for studying thymic output under physiological and pathological conditions, a non-animal technology facilitating efficient TCR-selected T cell development and evaluating T cell output from NHP-derived HSPCs has been lacking. To address this gap, we established a rhesus macaque-specific artificial thymic organoid (RhATO) modeling primary thymus-tissue-free thymopoiesi

Cellular immune endophenotypes separating early and late-onset myasthenia gravis.

The 2 main subgroups of autoimmune myasthenia gravis, a neuromuscular junction disorder associated with muscle weakness, are early- and late-onset forms, defined by onset before or after 50 years of age. Both carry acetylcholine-receptor autoantibodies but differ in sex ratios, genetics, and occurrence of disease-specific thymus inflammation. To distinguish the 2 forms by cellular immune phenotyping, we applied multimodal techniques, including deep spectral cytometric phenotyping and single-cell sequencing. Analysis of 2 independent cohorts identified immunological differences driven by 3 main

Long-Term Immune Alterations After Thymectomy in Early Childhood: A Case Series.

The thymus plays a central role in T-lymphocyte maturation and in maintaining immune defense against infections and autoimmunity. We describe three boys (mean age 7.3 years) who underwent total thymectomy during surgical correction of congenital heart disease, two in infancy and one in the neonatal period. Patient A presented (14 years old) with recurrent cervical lymphadenopathy, generalized Epstein-Barr virus (EBV) infection, and hepatic fibrosis. Patient B (two-year-old) developed splenomegaly and had a history of frequent lower respiratory tract infections and pseudohypoaldosteronism type

Genome-wide analysis of somatic noncoding mutation patterns and mitochondrial heteroplasmic shift in type B1 and B2 thymomas.

Type B1 and B2 thymomas are lymphocyte-rich malignant tumours with few somatic mutations in protein-coding regions of the nuclear genome; nonetheless, noncoding regions remain uncharacterized. Here, we developed a method to isolate pure thymoma cells from lymphocyte-rich tissues, and then performed genome-wide deep sequencing. The total number of somatic mutations was ~80 times higher in noncoding regions than in coding regions in type B12 thymomas (1,671.3 versus 21.1 per case). Coding mutations were identified in epigenetic regulators, DNA repair genes, and some other genes. Nevertheless, 40

Comprehensive proteome profiling of molecular endotypes in Japanese adults with moderate-to-severe atopic dermatitis.

INTRODUCTION: Atopic dermatitis (AD) is an inflammatory skin disease that is heterogeneous in clinical presentation and biological mechanisms. Several studies have suggested biomarker-defined molecular endotypes in AD. This study aimed to characterize potential endotypes in Japanese patients with moderate-to-severe AD and comprehensively evaluate their circulating protein profiles to better understand disease etiology. METHODS: Serum samples from Japanese patients with moderate-to-severe AD (n = 73) enrolled in a phase 3 study of baricitinib (BREEZE-AD2; NCT03334422) and samples from healthy

Genetic alterations and aberrant hormonal pathways in thymic epithelial tumors.

BACKGROUND: Patients with thymic epithelial tumors (TETs) frequently show coexistence of various paraneoplastic syndromes, which severely affect their survival. Moreover, there is a lack of effective clinical treatment strategies for patients with unresectable metastatic and recurrent TETs. METHODS: To explore the genetic alterations that play a key role in the pathogenesis of TETs, we analyzed the whole-exome sequencing data from 24 patients diagnosed to have TETs at the First Affiliated Hospital of Nanjing Medical University. RESULTS: Mutated genes in TETs were enriched in several hormone-as

Co-existence of Myasthenia Gravis and Addison's Disease Without Thymoma: A Case Report.

INTRODUCTION: Myasthenia Gravis (MG) and Addison's Disease (AD) co-existing is very rare. The estimated prevalence of this co-occurrence is less than 1 in 100 000, underscoring its clinical significance. We present a female case with coexisting AD and MG. A 45-year-old female arrived at our clinic complaining of breathing difficulties for a month, a drooping left eyelid, and swallowing difficulties for 10 days. She has been treated with 5 mg/day prednisone for AD for three years. Long-term corticosteroid therapy may modulate immune responses and influence the timing of MG onset. Neurological e
