PURPOSE: Glucose-dependent insulinotropic polypeptide (GIP) plays a crucial role in lipid metabolism. The effect of GIP on pancreatic lipid and whether it modulates risk of type 2 diabetes (T2D) associated with fatty pancreas remain unknown. The aim of this study was to investigate the interaction between genetically predicted GIP levels and fatty pancreas in the development of T2D.
METHODS: This is a large-scale cohort study using data from the UK Biobank. Participants of White ethnicity without diabetes at the imaging visit were included in the analysis. The loss-of-function GIPR variant E3

Adenomyosis is a common gynecological condition characterized by ectopic endometrial-like tissue deep within the myometrium, causing debilitating symptoms. Treatment options are limited to hormones that primarily focus on symptom management only or invasive hysterectomy. Despite its prevalence, the pathogenesis of adenomyosis remains poorly understood, considerably impeding the development of targeted, true disease modifying treatments. Here, through spatial transcriptomics analysis of full-thickness human uterine wall biopsies (n = 10), we demonstrate that adenomyosis lesions exhibit a distin

BACKGROUND: Ectopic adrenocorticotropic hormone (ACTH) syndrome is a rare form of ACTH-dependent Cushing syndrome, most commonly caused by neuroendocrine tumors. Pheochromocytoma is a rare source of ectopic ACTH secretion. Its clinical presentation is often heterogeneous and may lack the typical features of Cushing syndrome, which can lead to delayed diagnosis or misdiagnosis. Increased awareness of this rare entity is essential for early diagnosis and appropriate management.
CASE PRESENTATION: We report two cases of ectopic ACTH syndrome caused by pheochromocytoma. Both patients presented wi

Plant organ formation involves an initiation stage followed by growth, together determining organ shape. The plant hormone auxin regulates both initiation and growth. However, it is still not clear how auxin facilitates these two separate successive stages. Here, we show that in tomato compound-leaf development, auxin promotes leaflet initiation and growth via opposite effects on gibberellin (GA) metabolism. Short auxin treatment induced the expression of GA catabolism genes, while a longer increased auxin response induced the expression of GA biosynthesis genes. GA biosynthesis mutants suppre

Thediagnosis of Cushing syndrome can be challenging, particularly when bilateral inferior petrosal sinus sampling (BIPSS) is required to differentiate ectopic tumors from pituitary adrenocorticotropic hormone (ACTH)-releasing tumors. When corticotropin-releasing hormone (CRH) is used as a stimulus, a state of hypercortisolism is required to suppress CRH receptors in pituitary corticotrophs, preventing the release of ACTH by normal pituitary tissue. By contrast, desmopressin acts through vasopressin receptors that are preferentially expressed in ACTH-secreting pituitary tumors, suggesting poten

A 2-bp insertion in McLecrk1b encoding an L-type lectin receptor-like kinase (L-LecRLK), the candidate gene for the major QTL mfl5.1 for mature fruit length, results in a premature stop codon and causes short fruit in bitter gourd. Fruit length is a key determinant of yield and market class in bitter gourd, yet the causal genes controlling fruit elongation remain largely unknown. In our previous QTL analysis using two inbred accessions with extremely short (HNU004) and moderately long (HNU025) fruits, mfl5.1 showed the largest effect. Here, we applied marker-assisted backcrossing to minimize b

Calcitonin-secreting neuroendocrine neoplasms of the lung are extremely rare. Differentiating medullary thyroid carcinoma (MTC) from ectopic calcitonin secretion is challenging. Elevated calcitonin usually triggers a thyroid-focused workup, but ectopic secretion must be considered to avoid misdiagnosis and unnecessary thyroidectomy. We report a 54-year-old man presenting with weight loss and back pain. Serum calcitonin was markedly elevated at 1011 pg/mL (SI: 293 pmol/L) (reference range, <16 pg/mL [SI: <4.67 pmol/L] for basal levels in men). Thyroid ultrasonography showed a >2 cm nodule in th

The survival of ectopic endometrial lesions in endometriosis critically depends on their ability to evade immune recognition and clearance by the host, a process known as immune evasion. Recent studies suggest that this process is not driven by a single factor but rather results from a multidimensional, interactive regulatory network involving immune dysregulation, epigenetic remodeling, and metabolic reprogramming. The pathophysiology of endometriosis is characterized by reduced cytotoxic activity and functional exhaustion of effector immune cells, along with excessive activation and expansio

In acute myeloid leukemia (AML), therapeutic resistance is intimately linked to vascular microenvironment remodeling; however, the initiating molecular determinants remain elusive. Here, we identified exosomal pyruvate kinase M2 (PKM2) as a pivotal mediator of this pathogenic process. PKM2 was abundantly present in AML-derived exosomes and transferred to human umbilical vein endothelial cells (HUVECs), eliciting metabolic reprogramming characterized by enhanced glycolysis and angiogenic activation. Mechanistically, PKM2 knockdown in AML cells substantially attenuated exosome-induced endothelia

Primary hyperparathyroidism (PHPT) is characterized by autonomous secretion of parathyroid hormone, resulting in hypercalcemia and systemic complications involving kidney, bone, and neurological functions. It is usually caused by a single benign parathyroid adenoma, with the majority of adenomas located in the cervical region. Ectopic parathyroid adenomas account for up to 5% of cases, most frequently within the mediastinum, posing diagnostic and surgical challenges. We report the case of a 55-year-old male patient who presented with a three-month history of persistent lumbar pain, nephrolithi

Bronchial carcinoid tumors are malignant neuroendocrine tumors that typically arise from enterochromaffin cells found in the epithelial lining of the lungs. We report a case of a 62-year-old female who was diagnosed with an atypical bronchial carcinoid tumor and subsequently underwent left pneumonectomy in December 2002. A year later, the patient developed metastases to the liver, bone, and pancreas. Over the subsequent decade, she developed clinical features of acromegaly and had persistently elevated insulin-like growth factor 1 (IGF-1) levels. Further investigation of the metastatic lesions

Endometriosis, defined as the ectopic growth of endometrial tissue outside of the uterine cavity, is an inflammatory and hormone-dependent disease that causes excruciating pelvic pain, infertility, and significantly decreases quality of life in affected patients. The JUN N-terminal kinases (JNKs) are a leading class of nonhormonal therapeutic targets that have been validated in preclinical models of endometriosis and in a Phase 1/2 clinical trial. Despite their therapeutic potential, JNK inhibitors with increased potency and specificity are needed to address the inflammatory pathology of endom

Precise regulation of organ size is essential for proper function, yet the underlying logic remains unclear. Here, we identify a Hippo-IGF2 signaling axis as a regulator of organ growth. During mouse liver development, Igf2 is highly expressed in fetal and neonatal hepatocytes to fuel rapid growth but is directly silenced by the Hippo signaling pathway at the postnatal stage, enforcing growth arrest and determining liver size. In contrast, chronic liver injury inactivates Hippo signaling and induces Igf2 expression, which is essential for regeneration. Notably, this regenerative response is de

Endometriosis (EMs) is a common gynecological disorder in women of reproductive age, often presenting with pelvic pain and infertility. Its progression is closely linked to estrogen levels. While hormonal therapies are effective, long-term use can disrupt systemic hormone balance. To address this, we developed M1 macrophage membrane-derived nanovesicles (M1NVs) that leverage the natural CCL2-CCR2 targeting axis to selectively deliver drugs to ectopic endometrial lesions. The estrogen receptor antagonist bazedoxifene (BAZ) was encapsulated into M1NVs, enabling targeted delivery to ectopic tissu

A 53-year-old man underwent resection of a nonfunctional 9 cm oncocytic adrenal cortical carcinoma (ACC) with focal high-grade features. Two years later he presented again, with gynecomastia and elevated levels of estradiol, 11-deoxycortisol, and serum calcium (10.5 mg/dL, SI: 2.62 mmol/L) (reference range 8.4-10.2 mg/dL [SI: 2.10-2.55 mmol/L]). Imaging demonstrated locally recurrent ACC, which was resected. Steroid excess resolved and he started mitotane. Months after, his calcium was 11.9 mg/dL (SI: 2.97 mmol/L), parathyroid hormone (PTH) 209 pg/mL (SI: 22.0 pmol/L) (reference range 15-65 pg

Diffuse idiopathic pulmonary neuroendocrine cell hyperplasia (DIPNECH) is a rare, preinvasive lung disorder characterized by multifocal neuroendocrine cell proliferation. Because it presents with chronic cough and wheezing, it is frequently misdiagnosed as asthma. Ectopic adrenocorticotropic hormone (ACTH) syndrome (EAS) arising from DIPNECH is exceptionally rare. We present a rare case of a 62-year-old woman with a long-standing history of presumed asthma who developed ACTH-dependent Cushing syndrome. Bilateral inferior petrosal sinus sampling confirmed ectopic ACTH secretion. Subsequently, c

This report describes a rare instance of an adrenocorticotropic hormone (ACTH)-secreting metastatic hepatic neuroendocrine tumour (MHNET) exhibiting as an ectopic Cushing's syndrome. A 41-year-old woman with a history of supraglottic neuroendocrine tumour presented with rapid weight gain, severe hypokalemia, and hypercortisolism. Pituitary imaging was unremarkable. However, the DOTATATE PET/CT showed a large liver lesion. Initiating etomidate infusion caused a rapid drop in cortisol, leading to severe vasoplegic and septic shock, which delayed the surgery. The patient required intensive care s

Aging drives physiological decline and predisposes individuals to multiple age-related pathologies, constituting a major global health challenge. Growth hormone receptor (GHR), a critical regulator of growth, development, and metabolism, has emerged as a potential therapeutic target. However, its precise role in aging and age-related diseases remains incompletely defined. Here, we demonstrate that hepatocyte-specific GHR knockout mice display accelerated aging-related phenotypes, characterized by shortened lifespan, enhanced cellular senescence, reduced metabolic stress resilience, cognitive d
