Expanded prevalence of group 4 capsule expression and its genetic requirements in a broad array of Shigella strains.

UNLABELLED: The expression of a group 4 capsule (G4C) has been identified in Shigella sonnei and Shigella flexneri serotype 6 and demonstrated to play a significant role in pathogenic processes in S. sonnei. However, it has not been examined phenotypically in S. flexneri 6. Despite the impact of G4C, no systematic analysis of its expression or the presence of the genomic determinants has been performed across a large collection of S. sonnei and S. flexneri serotypes 2a and 6 or any of the other >46 Shigella strains and serotypes that cause disease. Our examination of the genomes of 1,219 geogr

Development of a Novel Shigella Quadrivalent Conjugate Vaccine Using Shigella O-Polysaccharide and IpaB Carrier Protein.

Background/Objectives: Shigella is the leading bacterial cause of diarrheal disease worldwide. Although multiple vaccine candidates are under development and in clinical trials, no Shigella vaccine is currently available on the market. Shigella comprises four species: S. dysenteriae, S. flexneri, S. boydii, and S. sonnei. S. flexneri has been recognized as the most prevalent species, particularly in low- and middle-income countries (LMICs), and the top serotypes are S. flexneri 2a, 3a and 6. Conversely, S. sonnei has a single serotype and predominates in high-income countries (HICs). Invasion

Isolation and characterization of lytic Shigella bacteriophages with rapid in vitro and ex vivo bactericidal activity.

INTRODUCTION: The emergence of shigellosis caused by Shigella spp. poses a significant threat to global health, and the prevalence of multidrug-resistant (MDR) strains necessitates the development of effective antimicrobial therapies. Bacteriophages present a promising approach for both prevention and treatment of MDR infections. METHODS: Ten lytic bacteriophages targeting Shigella flexneri 2457T were isolated from wastewater using the double-layer agar method. Phage morphology, lytic activity spectrum, latent period, burst size, bacterial killing efficiency, and efficacy under ex vivo microb

Genomic epidemiology and antimicrobial resistance profile of Shigella isolated from diarrhoea diseases in under-five children in Blantyre, Malawi.

Antimicrobial resistance (AMR) in Shigella is rising globally, complicating shigellosis management. Whole-genome sequence analysis (WGSA) has advanced our understanding of AMR and transmission dynamics, yet contemporary whole-genome sequencing data from Shigella in sub-Saharan Africa remain scarce. In this study, we applied WGSA to 27 Shigella isolates collected from children presenting with diarrhoea at Ndirande Health Centre in Malawi (2022-2023), as part of the Enterics for Global Health Shigella surveillance study. Serotyping, AMR profiling and phylogenetic analysis revealed Shigella sonne

Shigella O-antigen diversity and evolution: Comprehensive analysis and development of ShigO3D, a three-dimensional structural repository.

The genus Shigella comprises four species: Shigella dysenteriae (serogroup A), Shigella flexneri (serogroup B), Shigella boydii (serogroup C), and Shigella sonnei (serogroup D), which cause shigellosis. The emergence and global spread of multidrug resistance make these species a severe threat to public health, prompting the World Health Organization (WHO) to designate fluoroquinolone-resistant Shigella as a high-priority pathogen. Shigella employs a repertoire of virulence factors to evade the host immune response, among which the lipopolysaccharide (LPS), a primary endotoxin, plays a key role

Shigellosis.

Shigella is a Gram-negative, facultative intracellular, gastric acid-resistant bacterium of the Enterobacteriaceae family, which includes four serogroups: Shigella dysenteriae, Shigella sonnei, Shigella flexneri, and Shigella boydii. Globally, shigellosis is the most common cause of invasive bloody diarrhoea in children younger than 5 years. Humans are the only natural reservoir and an inoculum of only 10-100 organisms is required for infection. Rising antibiotic resistance rates increasingly reduce the ability to adequately treat severe disease. The prevention of infection with vaccination an

Genomic and epidemiological characteristics of Shigella boydii in Australia, 1991-2022.

Shigellosis is a leading cause of diarrheal mortality worldwide. Shigella boydii is one of four Shigella species that contributes to this burden, however studies on S. boydii are limited. Here we combined epidemiological and genomic data to better understand S. boydii circulating both in Australia and globally. Between 1991 and 2019, there were 294 cases of S. boydii infections notified to the National Notifiable Diseases Surveillance System by Australian states and territories, with an increasing trend in notifications observed from 2013. Of cases whose place of acquisition was known, 54% (11

Evaluating the ciprofloxacin potentiation efficacy of repurposed drug flupentixol through efflux pump inhibition of pathogenic bacteria.

Antimicrobial resistance concerning burdens is increasing day by day. Efflux pump upregulation is one of the mechanisms through which bacteria develop resistance to antibiotics. To overcome this escalating problem in healthcare, repurposing existing drugs as efflux pump inhibitors in combination with antibiotics can help mitigate the growing resistance issue worldwide. In our study, we investigated the efflux pump inhibiting capability of the repurposed drug flupentixol in combination with different classes of antibiotics. In vitro combination susceptibility testing (disk diffusion and minimum

Immunoinformatics-based design of a next generation multi-epitope vaccine candidate against Shigella boydii using a hierarchical subtractive proteomics approach.

Shigella boydii (strain CDC 3083-94 / BS512) is a Gram-negative facultative anaerobic bacterium that plays a significant role as a pathogen causing bacillary dysentery, especially in developing areas where hygiene is poor. Although less frequently reported compared to other Shigella species, S. boydii still contributes substantially to global shigellosis cases, particularly in low-income endemic regions. Despite its clinical relevance, there has been limited research dedicated on identifying strain-specific antigenic targets for vaccine development. The current study utilized the subtractive p

Modulating the antibacterial effect of the existing antibiotics along with repurposing drug metformin.

Owing to the extensive prevalence of resistant bacteria to numerous antibiotic classes, antimicrobial resistance (AMR) poses a well-known hazard to world health. As an alternate approach in the field of antimicrobial drug discovery, repurposing the available medications which are also called antibiotic resistance breakers has been pursued for the treatment of infections with antimicrobial resistance pathogens. In this study, we used Haloperidol, Metformin and Hydroxychloroquine as repurposing drugs in in vitro (Antibacterial Antibiotic Sensitivity Test and Minimum Inhibitory Concentration-MIC)

Genome-Wide Investigation Reveals Potential Therapeutic Targets in Shigella spp.

Shigella stands as a major contributor to bacterial dysentery worldwide scale, particularly in developing countries with inadequate sanitation and hygiene. The emergence of multidrug-resistant strains exacerbates the challenge of treating Shigella infections, particularly in regions where access to healthcare and alternative antibiotics is limited. Therefore, investigations on how bacteria evade antibiotics and eventually develop resistance could open new avenues for research to develop novel therapeutics. The aim of this study was to analyze whole genome sequence (WGS) of human pathogenic Shi
