Ocular Clues to Liver Disease: A Strategic Diagnostic Lens.

BACKGROUND/OBJECTIVES: Hepatic diseases frequently present with ocular manifestations that aid diagnosis, provide prognostic data, and guide therapy. Despite the clear utility of the liver-eye axis, the literature lacks reviews that categorize these manifestations by etiology. This review evaluates current evidence to identify ocular findings that serve as clinical tools for diagnosis, prognosis, and therapeutic monitoring of hepatic pathologies. METHODS: A narrative review was conducted using PubMed and Google Scholar to identify English-language articles addressing ocular manifestations ass

Transthyretin and Vitamin A Metabolism: A Review for the Cardiac Amyloidosis Specialist.

Transthyretin (TTR) amyloidosis is a systemic, progressive, and fatal disease. TTR is integral in vitamin A (retinol) transport via its binding to retinol binding protein 4 (RBP4). Current and emerging therapies for TTR amyloid cardiomyopathy (ATTR-CM), including RNAi therapies and potentially CRISPR-based therapies, reduce hepatic transthyretin production and hence decrease serum RBP4, which decreases circulating vitamin A levels. However, despite these reductions in circulating vitamin A, hepatic reserves and alternative delivery mechanisms may prevent clinical manifestations of vitamin A de

A rapid LC-MS/MS method with 2-nitrosopyridine derivatization to determine fat-soluble vitamins, revealing a biphasic bile acids association in clinical cohorts.

Fat-soluble vitamins (A, D, and E) are essential micronutrients whose absorption critically depends on bile acid-mediated solubilization. However, studies associating bile acids and these vitamins remain limited, partly due to methodological challenges. Therefore, we developed a novel and rapid LC-MS/MS method for the simultaneous quantification of these vitamins in serum, based on 2-nitrosopyridine (PyrNo) click derivatization. This strategy employs Diels-Alder cycloaddition for vitamin A/D and acid-catalyzed oxidative dehydrogenation for vitamin E, enabling one-step protein precipitation fol

The role of specific nutrients in preventing immune system and blood cell disorders: an umbrella review.

INTRODUCTION: Nutritional deficiencies affecting the immune and haematopoietic systems represent a well-known global public health challenge: only the iron deficiency anaemia affects 1.62 billion individuals, especially in vulnerable populations. However, the protective effect that nutrition might give on disorders of these systems is still poorly understood. This umbrella review aims to synthesise the available evidence on the effectiveness of nutritional interventions in the primary prevention of blood and immune disorders, with a focus on the role of essential micronutrients and bioactive c

Maternal vitamin A deficiency programs offspring visceral hypersensitivity through RARβ-COX-2/PGE2 signaling.

Visceral hypersensitivity (VH) is regarded as a core pathophysiological mechanism in irritable bowel syndrome (IBS). Although maternal vitamin A deficiency (VAD) during pregnancy has been linked to gastrointestinal dysfunction in offspring, its specific role in VH and the underlying mechanisms are not yet fully understood. This study aimed to explore the effects of maternal VAD on the development of VH in offspring and to assess the potential therapeutic role of postnatal vitamin A supplementation (VAS). Using a maternal VAD rat model, we observed that maternal VAD was associated with the deve

Motor and cognitive development is associated with anthropometry and severe zinc deficiency in infants, in rural Madagascar.

Globally, more than 250 million children under five fail to reach their developmental potential, with malnutrition representing a major contributing factor. In Madagascar, as in many low- and middle-income countries, malnutrition remains a serious public health concern. Although the high prevalence of micronutrient deficiencies, stunting, and wasting is well documented, their consequences for early motor and cognitive development, which can be assessed reliably by six months, are less studied. In 2019, a baseline data collection prior to intervention as part of a randomized controlled trial co

Blocking hepatocyte retinoic acid signaling increases hepatic retinol-RBP4 secretion and disrupts vitamin A homeostasis in mice.

Vitamin A is an essential micronutrient with broad physiological roles. The liver is of central importance in whole-body vitamin A homeostasis, with hepatocytes distributing retinol to the rest of the body by secreting it in complex with Retinol-binding protein 4 (RBP4). The goal of this study was to elucidate homeostatic mechanisms regulating hepatic vitamin A metabolism by specifically blocking retinoic acid (RA) signaling in hepatocytes. Herein, we used Albumin-Cre (AlbCre) mice to conditionally express a dominant negative retinoic acid receptor (Rardn) in hepatocytes, generating AlbCre:Rar

Maternal nutrition determines the variable expressivity of STRA6-associated eye malformations.

Mutations in STRA6, the high-affinity receptor for retinol-binding protein, cause Matthew-Wood syndrome, a congenital disorder characterized by striking phenotypic variability ranging from isolated ocular anomalies to severe multisystem malformations. The mechanisms underlying this variable expressivity and incomplete penetrance remain poorly understood. Here, we used mice to determine how maternal vitamin A status and embryonic Stra6 genotype interact to shape ocular development. Using Stra6-deficient mice bred under defined dietary conditions, we demonstrate that maternal vitamin A availabil

Multiplexed CRISPR/Cas9 mediated knockdown of BCH gene in potato enhances beta-carotene to combat vitamin A deficiency.

The inadequate amounts of provitamin A carotenoids in crops contribute to the widespread vitamin A deficiency, leading to malnutrition and blindness in humans. Suppression of the β-carotene hydroxylase (BCH) increases β-carotene levels. In the current study, we utilized the multiplexed CRISPR/Cas9 approach by designing three targets against the BCH gene in a local potato cultivar. Transformation efficiency was recorded as 15%, the successful integration of the CRISPR/Cas9-BCH multiplex construct in potatoes was confirmed through PCR. When analysed using TIDE software, Sanger sequencing reveale

Spray dried β-carotene: Encapsulation, stabilization, and controlled-release.

Vitamin A deficiency remains a major public health challenge worldwide, and β-carotene, as a provitamin A carotenoid, is widely used in food fortification to improve vitamin A intake. However, its application in foods is constrained by poor water solubility, limited bioaccessibility, and high susceptibility to heat, light, and oxygen. This review evaluates spray drying as an encapsulation strategy for β-carotene, focusing on the effects of wall materials and operating conditions on powder formation, encapsulation efficiency, stability, and release behavior. It also summarizes the chemical stru

Organic-Microbial Strategies for Iron and Zinc Biofortification in Orange-Fleshed Sweet Potato [Ipomoea batatas (L.) Lam.].

Orange-fleshed sweet potato (OFSP) is widely promoted in sub-Saharan Africa, including Senegal, for its contribution to vitamin A deficiency mitigation. However, its potential to improve mineral nutrition remains limited by low iron (Fe) and zinc (Zn) concentrations and their restricted bioavailability, which may contribute to persistent micronutrient deficiencies affecting 42% of children under 5 years and 37% of women of reproductive age in Senegal. In this context, organic waste products (OWPs) and microbial inocula (MIs) represent promising agroecological alternatives to conventional ferti
