BACKGROUND: Sepsis is a complex and life-threatening disease process related to a systemic response to severe infection. Due to the challenges of treating patients with sepsis, new therapies are being investigated, including cell-based approaches. Trophoblast stem cells (TSCs) are immune privileged cells with immunomodulatory properties. Thus, we proposed that TSCs may be beneficial in experimental models of sepsis to regulate the immune response and curtail organ injury.
METHODS: Sepsis was induced by experimental models in mice; cecal ligation and puncture (CLP) and lung infection with Stre

Streptococcus pneumoniae is a gram-positive bacterium that causes a variety of diseases, including otitis media, sinusitis, pneumonia, bacteremia, and meningitis. While pneumococcal conjugate vaccines have reduced disease burden globally, serotype replacement affects the effectiveness of the vaccine, and S. pneumoniae has become resistant to a variety of antibiotics. Infections caused by antibiotic-resistant strains can interfere with the treatment of associated diseases. Monoclonal antibody therapy has emerged as a promising solution to the clinical problems caused by refractory S. pneumoniae

INTRODUCTION: Streptococcus pneumoniae is a human pathobiont that asymptomatically colonizes the upper respiratory tract but can cause severe diseases such as pneumonia, sepsis, and meningitis, as well as non-invasive infections like otitis media and sinusitis. It thrives in the nutrient-limited environment of the nasopharynx and has evolved mechanisms to manage host-induced stress and regulate protein levels accordingly.
METHODS: To investigate the molecular biology of S. pneumoniae under in vitro and infection-relevant conditions, a suitable cultivation medium is essential for reproducible

Streptococcus pneumoniae (pneumococcus), a major human pathogen, causes pneumonia, bacteraemia and meningitis. Unlike free-living bacteria such as Escherichia coli, pathogenic bacteria like S. pneumoniae possess incomplete metabolic pathways and depend on host-derived nutrients. While nucleotide metabolism is essential for bacterial proliferation, how S. pneumoniae takes up uracil is still not well understood. Identifying uracil transporters is important, as they may serve as targets for novel antimicrobial strategies. In this study, we functionally characterized a uracil transporter using the

Purpura fulminans is a rare but severe complication of septic shock, often associated with Streptococcus pneumoniae infection in asplenic individuals, with high mortality and limb amputation risk. Although pneumococcal vaccines are available, infections caused by non-vaccine serotypes remain a major concern. A 39-year-old man with a history of splenectomy 18 years prior presented with dyspnea, malaise, and gastrointestinal symptoms for two days. He had never received pneumococcal vaccination following his splenectomy. On arrival, he was hypotensive, tachycardic, and exhibited signs of multi-or

OBJECTIVES: To evaluate whether simultaneous vaccination of the 13-valent pneumococcal conjugate vaccine (PCV13) and the 23-valent polysaccharide vaccine (PPSV23) elicits higher antigen-specific memory B cell responses compared to sequential (PCV13 followed by PPSV23 after 6 months) or single PPSV23 vaccination in elderly.
METHODS: In this monocentric, randomized trial, vaccine-naïve adults aged ≥60 years were assigned 1:1:1 to a simultaneous, sequential or single vaccination group. The primary outcome was the change in memory B cells specific for four vaccine-serotypes (ST3, ST14, ST19A, and

Influenza virus and Streptococcus pneumoniae are major respiratory pathogens responsible for significant global mortality. While influenza causes seasonal flu, pneumococcus is associated with pneumonia, meningitis, sepsis, and otitis, often worsening influenza cases through secondary infections. Aiming to develop a bivalent vaccine against these two pathogens, we used reverse genetics to construct a recombinant influenza virus that carries the gene of the pneumococcal PspA protein (Flu-PspA). This study assessed the safety and efficacy of a heterologous prime-boost vaccine protocol consisting

Introduction. Infection is a leading cause of mortality during the first year of life, with the neonatal period being particularly high risk. It is critical to identify non-antibiotic approaches to improve neonatal infection outcomes due to the non-specific clinical signs of neonatal infection and negative consequences of early-life antibiotic exposure. Lactoferrin is a protein found in all mammalian milk that has a variety of antimicrobial properties. Clinical trials have shown that lactoferrin supplementation during the neonatal period decreases rates of sepsis.Knowledge Gap. It remains uncl

BACKGROUND: Bacterial pneumonia remains a leading cause of morbidity and mortality worldwide despite the widespread availability of antibiotics. Novel pneumonia therapies and biomarkers are urgently needed to improve outcomes and advance personalized therapy. Using an established baboon model of S. pneumoniae pneumonia, we sought to characterize the temporal dynamics of pneumonia host responses to identify novel potential diagnostic and therapeutic molecular targets.
METHODS: We performed whole blood transcriptomics, unbiased proteomics, and peripheral cytokine measurements serially in baboon

BACKGROUND: Patients with sepsis are prone to complications such as cardiac damage and arrhythmias, including atrial fibrillation (AF). Previous studies have suggested an association between certain pathogens and new-onset atrial fibrillation (NOAF) in sepsis, but a detailed and comprehensive analysis is lacking.
OBJECTIVE: This study aimed to detect and analyze pathogens in patients with sepsis complicated by NOAF to provide a scientific basis for its prevention.
METHODS: In this retrospective observational study, we selected 123 septic patients with NOAF as the case group and 2000 septic p

The rise of multidrug-resistant Streptococcus pneumoniae poses a major public health threat, necessitating novel therapeutic targets. Pneumococcal Surface Adhesin A (PsaA), a conserved surface lipoprotein, plays a key role in manganese acquisition, colonization and virulence. Immunization with PsaA elicits protective immunity, while fragment-based drug design has identified inhibitors disrupting its function. PsaA emerges as a promising molecular target for innovative therapeutic strategies against pneumococcal diseases. Antimicrobial peptides (AMPs) are crucial components of the innate immune

Patients are known to be at an increased risk of sepsis post-splenectomy due to encapsulated organisms, such as Streptococcus pneumoniae. This is clinically important, as overwhelming post-splenectomy infection (OPSI) is very serious. Prevention, including vaccination, is recommended. Herein, we describe the case of a patient who survived a second attack of OPSI due to S. pneumoniae, serotype 34, that occurred 8 years after acute pericarditis related to S. pneumoniae-associated OPSI. The patient underwent a splenectomy due to malignant lymphoma and had been vaccinated with PPSV 23, but had not

Campylobacter spp. bacteremia can be seen in immunocompromised patients. Nephrotic syndrome (NS) predisposes patients to serious bacterial infections (SBI). The most common SBIs in NS are spontaneous bacterial peritonitis (SBP), sepsis, and bacteremia due to Streptococcus pneumoniae or Escherichia coli. To date, despite several case reports in adults, there has been no report of Campylobacter bacteremia in the context of pediatric NS. A seven-year-old boy with NS receiving steroid treatment was admitted to our hospital, presenting with fever, diarrhea, and abdominal pain. Ceftriaxone was initi

UNLABELLED: Streptococcus pneumoniae is a leading causative pathogen of community-acquired pneumonia and sepsis. The increasing prevalence of antibiotic resistance among S. pneumoniae strains poses a major challenge to conventional antibiotic therapies. This study aimed to systematically evaluate the antibacterial, anti-biofilm, and in vivo therapeutic effects of montelukast against S. pneumoniae and to explore its potential for drug repurposing as an anti-infective agent. In vitro susceptibility testing revealed that montelukast exhibited inhibitory activity against both standard reference st

Streptococcus pneumoniae is responsible for causing a range of diseases, from self-limiting otitis media to more severe disease such as pneumonia, sepsis, and meningitis. Vaccines and antibiotics have successfully decreased this disease burden; however, serotype replacement due to vaccines and antibiotic resistance remain issues pointing to the need for further research into alternative methods of treatment. S. pneumoniae continues to pose a global threat to young children and the elderly. Endolysins, which function as peptidoglycan hydrolases, are an attractive alternative treatment given the
