Ethnic differences in thiamine status persist after adjusting for diet; findings from the UK National Diet and Nutrition Survey.

BACKGROUND/OBJECTIVES: Thiamine (vitamin B1) status might play a role in cognition and mental health. Recent research suggests that there are ethnic differences in thiamine status. We aimed to test the hypothesis that black individuals have a higher risk of thiamine deficiency than white individuals, based on the erythrocyte transketolase activity coefficient (ETKAC), and assess differences in thiamine intake, using nutritional survey data. METHODS: We used the published data of the UK's National Diet and Nutrition Survey Rolling programme (2008-2019) and examined differences in the ETKAC bet

Loss of Mtarc1 Protects Against Steatotic Liver Disease in Mice.

BACKGROUND & AIMS: Metabolic dysfunction-associated steatotic liver disease (MASLD) spans from simple steatosis to metabolic dysfunction-associated steatohepatitis (MASH) and can progress to cirrhosis or hepatocellular carcinoma. Despite its prevalence, effective therapies are lacking. Recent genome-wide association studies identified a common missense variant (rs2642438) in the Mitochondrial Amidoxime Reducing Component 1 (MTARC1) gene that protects against liver cirrhosis without increasing cardiovascular disease risk. Biochemical and disease risk signatures associated with carriers of this

Dietary thiaminase alters morphology and decreases swimming performance of lake trout (Salvelinus namaycush).

The consumption of thiaminase can cause thiamine deficiency, which has been hypothesized to impede reintroduction efforts of lake trout (Salvelinus namaycush). In fishes, consumption of thiaminase is hypothesized to affect swimming performance and morphology because thiamine deficiency manifests as cardiac and neurological impairments. However, how those effects may differ among populations with different historical exposures to thiaminase remain understudied, despite the importance of these traits to survival and fitness. Here, juvenile lake trout from strains that originated from Seneca Lake

Vitamin B6 deficiency anemia resembles iron-deficiency anemia but does not activate intestinal HIF2⍺.

Microcytic hypochromic anemias, including iron-deficiency anemia (IDA) and vitamin B6-deficiency anemia (B6DA), share similar hematologic features but differ fundamentally in etiology and therapeutic responsiveness. IDA results from insufficient dietary iron, activating adaptive intestinal iron absorption via hypoxia-inducible factor 2⍺ (HIF2⍺), whereas B6DA impairs heme biosynthesis without systemic iron depletion, rendering it refractory to iron therapy. Here, we investigated whether intestinal HIF2⍺ activation distinguishes these anemias. Using mouse models, we established that severe dieta

Vitamin B12 Supports Skeletal Muscle Oxidative Phosphorylation Capacity in Male Mice.

BACKGROUND: Vitamin B-12 is a cofactor in folate-mediated 1-carbon metabolism, which generates nucleotides {thymidylate [deoxythymidine monophosphate (dTMP)] and purines} and methionine. Depressed de novo thymidylate (dTMP) synthesis leads to uracil accumulation in DNA. OBJECTIVES: This study aimed to determine how B-12 availability affects mitochondrial DNA (mtDNA) integrity and mitochondrial function in skeletal muscle. B-12 deficiency was modeled in young-adult mice. Intramuscular B-12 injection in aged mice assessed the role of B-12 supplementation in age-related changes in skeletal muscl

Efficacy of L-methylfolate and methylcobalamin in treating resistant hypertension associated with elevated serum homocysteine in hemodialysis patients.

BACKGROUND: End-stage renal disease (ESRD) patients receiving hemodialysis are experiencing a considerable increase in the burden of cardiovascular diseases (CVDs). In this patient population, hypertension is a prevalent modifiable cardiovascular risk factor that is associated with poor prognosis. Resistant hypertension in dialysis patients is challenging to manage since some individuals do not respond to antihypertensive medications or volume control. Hyperhomocysteinemia is common among ESRD patients. "H-type hypertension" or hyperhomocysteinemia-associated hypertension refers to resistant h

Preventable cerebral venous sinus thrombosis from diet- and alcohol-induced hyperhomocysteinemia: illustrative case.

BACKGROUND: Cerebral venous sinus thrombosis (CVST) is a rare stroke subtype (0.5%-3%) that frequently presents with nonspecific symptoms, delaying diagnosis. Although hyperhomocysteinemia is a known risk factor, acquired cases resulting from severe vitamin B12 and folate deficiencies can be easily overlooked by clinicians in any setting, making this a critical yet preventable cause of life-threatening thrombosis. OBSERVATIONS: A 44-year-old man with obesity, who adopted a carbohydrate-heavy diet and consumed excessive alcohol (70-140 g/day) following a major life event 3 years prior, present

Clinician awareness and implementation of vitamin B12 monitoring guidance in metformin users: a primary care survey.

BACKGROUND: Vitamin B12 deficiency is a recognised adverse effect of long-term metformin use. In 2022, the United Kingdom (UK) Medicines and Healthcare products Regulatory Agency (MHRA) issued a Drug Safety Update advising clinicians to consider periodic vitamin B12 monitoring in at-risk patients. However, no specific testing interval or operational guidance has been published, and uptake within primary care remains uncertain. This study aimed to assess clinician awareness of metformin-associated vitamin B12 deficiency, evaluate the impact of the MHRA alert, and identify perceived barriers and

Folic acid fortification of iodized salt improves the folate status of nonpregnant adult Ethiopian females and does not impair their iodine status: a community-based, household-randomized, dose-response trial.

BACKGROUND: Folic acid (FA) fortification of cereal grains reduces the prevalence of folate insufficiency among adult females and related neural tube defects (NTDs) in their offspring. However, fortifiable cereal grains have limited reach in many lower-income countries, so additional fortification options need to be developed and evaluated. OBJECTIVES: This study assessed the effects of FA fortification of iodized salt on women's discretionary salt intakes, biomarkers of folate and iodine status, and the occurrence of adverse events. METHODS: We conducted a community-based, 3-arm, household-

High-fat and choline-deficient western diets disrupt the ovarian microenvironment in mice†.

Obesity and metabolic dysfunction induced by high-fat and Western-style diets are key contributors to reproductive aging. However, the specific contributions of diet-induced obesity and liver damage to ovarian aging and follicle depletion remain unclear. This study investigated the impact of a high-fat diet (HFD) and a choline-deficient Western diet (CDWD) on ovarian aging in two mouse strains. Three-month-old female mice (C57BL/6 and Swiss) were assigned to a control standard diet (CSD), choline-deficient control diet (CDC), HFD, or CDWD for 10 weeks. C57BL/6 mice fed an HFD exhibited signifi

Combined B-vitamin supplementation on homocysteine and vascular outcomes in coronary heart disease: a meta-analysis.

OBJECTIVE: Hyperhomocysteinemia (Hcy) independently predicts coronary heart disease (CHD) and adverse cardiovascular events. Although folic acid plays a key role in Hcy metabolism, the effect of combined B-vitamin supplementation (folic acid, VB6, and VB12) on clinical outcomes in CHD remains uncertain. METHODS: A systematic search of PubMed, Embase, and the Cochrane Library was conducted from inception through April 2025 using MeSH terms including "folic acid," "vitamin B6," "vitamin B12," "coronary heart disease," and "homocysteine." A random-effects model was used for meta-analysis. RESUL

A Meta-Analysis of the Effects of Homocysteine-Lowering Therapy on Chronic Kidney Disease.

Hyperhomocysteinemia is common in chronic kidney disease; however, whether homocysteine-lowering therapy slows chronic kidney disease progression remains uncertain. Following the Preferred Reporting Items for Systematic reviews and Meta-Analyses guideline, we systematically searched PubMed and Web of Science (inception to January 2025) for randomized controlled trials evaluating folic acid alone or combined with vitamin B versus placebo/usual care/low-dose regimens in populations across the chronic kidney disease spectrum. The primary end point was composite kidney outcomes (all-cause mortalit
