Placental Autophagy: Causes and Consequences.

Suboptimal intake of dietary vitamin C (ascorbate) increases the risk of several chronic diseases but the exact metabolic pathways affected are still unknown. In this study, we examined the metabolic profile of mice lacking the enzyme gulonolactone oxidase (Gulo) required for the biosynthesis of ascorbate. Gulo-/- mice were supplemented with 0%, 0.01%, and 0.4% ascorbate (w/v) in drinking water and serum was collected for metabolite measurements by targeted mass spectrometry. We also quantified 42 serum cytokines and examined the levels of different stress markers in liver. The metabolic profi

Multi-factorial Hyperlactatemia Due to Linezolid and Metformin Treatment in the Setting of Vitamin C and B1 Deficiency.

BACKGROUND: Lactic acidosis can be a sign of poor tissue perfusion but may also be caused by some commonly used drugs, namely metformin and linezolid. PATIENTS AND METHODS: A 70-year-old overweight female presented to the emergency department with abdominal pain. Although there was concern for biliary sepsis, imaging and lab work were incompatible. RESULTS: Her home medications included linezolid for osteomyelitis and metformin for type II diabetes mellitus. Once these were stopped, the lactate level normalized within 36 h. In addition, she was malnourished and found to be vitamin C and B1 d

Shining light on vitamin C deficiency and scurvy in Canada: A scoping review protocol of risk profiles, health outcomes, and interventions.

Scurvy, resulting from vitamin C deficiency, is linked to serious health outcomes, including impaired collagen synthesis, anemia, and delayed wound healing. Once considered largely eradicated in high-income countries, scurvy has re-emerged among specific Canadian populations, driven by factors such as inadequate dietary intake, socioeconomic disparities, and limited access to nutritious foods. Despite growing awareness, evidence regarding its prevalence, risk factors, and public health responses in Canada remains sparse and fragmented. To address this knowledge gap, this study protocol outline

Development and validation of an LC-MS/MS method for the analysis of total ascorbic acid capacity in plasma.

A robust and fast LC-MS/MS method for quantifying total ascorbic acid (vitamin C) in plasma was developed and validated. Ascorbic acid is vital for antioxidant defense and enzymatic processes. Deficiency and hypovitaminosis remain prevalent in specific patient groups and can result in overlooked or misattributed clinical symptoms. To preserve ascorbic acid after venapuncture, EDTA blood samples were kept on ice and processed within two hours. Dehydroascorbic acid was reduced to ascorbic acid using tris(2-carboxyethyl)phosphine (TCEP). Ascorbic acid was separated by reversed-phase chromatograph

Vitamin C-deficient gulo-/- mice exhibit increased susceptibility to Helicobacter pylori colonization and gastric pathology.

Low dietary intake of vitamin C (ascorbic acid) has been associated with unfavorable clinical outcomes of Helicobacter pylori infection in human studies. However, experimental evidence regarding the influence of host vitamin C status on H. pylori colonization remains inconsistent across animal models. In this study, we generated vitamin C-deficient L-gulono-γ-lactone oxidase-deficient (gulo-/-) mice on an FVB genetic background to evaluate host susceptibility to H. pylori infection under defined vitamin C supplementation conditions. FVB gulo-/- mice were orally infected with H. pylori and prov

Vitamins and oral mucosal diseases: From bench to the bedside.

Vitamins are essential biocatalysts that have wide-ranging functions in supporting cellular metabolism. Vitamin deficiencies can significantly impact oral health, manifesting in oral mucosal and periodontal conditions. Deficiencies in the vitamin B complex are associated with glossitis, angular cheilitis, and recurrent aphthous stomatitis. Vitamin C deficiency leads to scurvy-related periodontal issues, whereas vitamin A insufficiency predisposes to oral epithelial keratinization abnormalities such as dry mouth and susceptibility to Candida. A reduction in vitamin D levels is linked to the ris

Unexplained bleeding tendency and scleroderma-like skin changes: a diagnostic journey to vitamin C deficiency.

BACKGROUND: A subset of patients with a bleeding tendency undergoing hemostatic evaluation may be misdiagnosed as bleeding disorder of unknown cause (BDUC). KEY CLINICAL QUESTION: Should vitamin C deficiency be considered in patients with bleeding symptoms but normal hemostatic test results suggestive of BDUC? CLINICAL APPROACH: We report on a patient with bleeding symptoms despite normal hemostatic examination, in whom vitamin C deficiency was ultimately identified as the cause of the bleeding disorder. The patient, a 26-year-old man, presented with extensive ecchymoses, anemia, and sclerod

Maternal Dyslipidaemia Aggravates Offspring Metabolic Dysfunction-Associated Steatotic Liver Disease.

Paediatric metabolic dysfunction-associated steatotic liver disease (MASLD) and steatohepatitis (MASH) display a distinct and severe histopathology. Maternal metabolic status may program offspring susceptibility to MASLD. This study investigated if maternal dyslipidaemia and low vitamin C (VitC) status influence offspring MASLD using a validated guinea pig model. Thirty female guinea pigs received a low-fat (LF) diet until pregnancy was confirmed, then remained on LF or switched to a high-fat (HF) diet with or without reduced VitC. Offspring were evaluated at birth and after 10 weeks on postwe

Atypical Presentation of Scurvy in the Oral Cavity.

Vitamin C is crucial for normal physiology and function. Severely insufficient levels of vitamin C can disrupt these processes, resulting in scurvy, a condition characterized by compromised collagen production and tissue degradation. The purpose of this report is to present a complex case of a pediatric patient with autism spectrum disorder exhibiting nonspecific oral lesions. Histopathological examination of the lesions revealed granulomatous inflammation, suggesting an initial diagnosis of orofacial granulomatosis, with a recommendation to rule out Crohn's disease. Further investigation conf

Milk and honey: how skin changes reveal the deeper dangers of nutritional deficiencies.

INTRODUCTION: The wide-reaching consequences of a restrictive diet are often underestimated or poorly recognized both by the public and many physicians. Deficiency symptoms may go unnoticed leading to unnecessary investigations and ineffective treatments, and a delay in a simple and directed treatment: suppletion of the missing nutrients. CASE PRESENTATION: This case-report describes a 68-year-old man in a high-income country who presented with a 3-week history of painful, indurated edema of the lower legs, immobility due to pain, and dyspnea. Treatment with antibiotics and LMWH was ineffecti

Erbin deficiency exacerbates sepsis‑induced liver injury through mitochondrial ROS/NLRP3‑mediated inflammatory responses.

BACKGROUND: Idiopathic pulmonary fibrosis (IPF) is a rare interstitial lung disease with a poor prognosis that currently lacks effective treatment methods. Preventing the acute exacerbation of IPF, identifying the molecular subtypes of patients, providing personalized treatment, and developing individualized drugs are guidelines for predictive, preventive, and personalized medicine (PPPM / 3PM) to promote the development of IPF. Oxidative stress (OS) is an important pathological process of IPF. However, the relationship between the expression levels of oxidative stress-related genes (OSRGs) an
