Genetic vitamin E deficiency does not affect MPTP susceptibility in the mouse brain.

Oxidative stress is involved in the degeneration of the nigrostriatal dopaminergic system in Parkinson's disease (PD). Vitamin E (alpha-tocopherol) is a potent antioxidant in the cell membrane that can trap free radicals and prohibit lipid peroxidation. The retention and secretion of vitamin E are regulated by alpha-tocopherol transfer protein (TTP) in the brain and liver. Dysfunction of TTP results in systemic deficiency of vitamin E in humans and mice, and increased oxidative stress in mouse brain. In this study, we investigated the effect of vitamin E deficiency in PD development by generat

Cortical gene expression in the vitamin E-deficient rat: possible mechanisms for the electrophysiological abnormalities of visual and neural function.

In mammals, severe and chronic deficiency of vitamin E (alpha-tocopherol) is associated with a characteristic neurological syndrome. Previously, we have shown that this syndrome is accompanied by electrophysiological abnormalities of neural and visual function. To investigate the molecular basis of the observed abnormalities, we used microarrays to monitor the expression of approximately 14,000 genes in the cerebral cortex from rats which had received diets containing 0, 1.25 and 5.0 mg/kg diet of all-rac-alpha-tocopheryl acetate for 14 months. Compared to the groups receiving 1.25 and 5.0 mg/

Cerebellar ataxia due to isolated vitamin E deficiency.

Ataxia is a common and important neurological finding in medical practice. Severe deficiency of Vitamin E can profoundly affect the central nervous system and can cause ataxia and peripheral neuropathy resembling Friedreich's ataxia. Vitamin E deficiency can occur with abetalipoproteinemia, cholestatic liver disease or fat malabsorption. Ataxia with isolated Vit E deficiency (AVED) is an Autosomal Recessive genetic disorder with a mutation in the alpha tocopherol transfer protein gene (TTPA). This condition responds to high dose of Vit E and is one of the important causes of treatable ataxia.

Phospholipid transfer protein deficiency impairs apolipoprotein-B secretion from hepatocytes by stimulating a proteolytic pathway through a relative deficiency of vitamin E and an increase in intracellular oxidants.

Genetic deficiency of the plasma phospholipid transfer protein (PLTP) in mice unexpectedly causes a substantial impairment in liver secretion of apolipoprotein-B (apoB), the major protein of atherogenic lipoproteins. To explore the mechanism, we examined the three known pathways for hepatic apoB secretory control, namely endoplasmic reticulum (ER)/proteasome-associated degradation (ERAD), post-ER pre-secretory proteolysis (PERPP), and receptor-mediated degradation, also known as re-uptake. First, we found that ERAD and cell surface re-uptake were not active in PLTP-null hepatocytes. Moreover,

Vitamin E prevents cell death induced by mild oxidative stress in chicken skeletal muscle cells.

Apoptosis and necrosis are two forms of cell death that can occur in response to various agents and oxidative damage. In addition to necrosis, apoptosis contributes to muscle fiber loss in various muscular dystrophies as well participates in the exudative diathesis in chicken, pathology caused by dietary deficiency of vitamin E and selenium, which affects muscle tissue. We have used chicken skeletal muscle cells and bovine fibroblasts to study molecular events involved in the cell death induced by oxidative stress and apoptotic agents. The effect of vitamin E on cell death induced by oxidants

Effect of dietary vitamin E and selenium on DNA damage in fresh and frozen tissues.

To determine whether deficiencies of dietary vitamin E and Se can elevate background DNA damage, rats were fed diets deficient in or supplemented with vitamin E (30 and 200 mg/kg diet) and Se (0.2 mg/kg diet) for 8 weeks. DNA damage was measured using the Comet (single-cell electrophoresis) assay and 8-oxodeoxyguanosine (8-oxo-dG) in liver, kidneys, and lymphocytes. We found that a deficiency of vitamin E and/or Se for 8 weeks did not significantly increase DNA damage in freshly isolated liver, kidneys, or lymphocytes. However, deficiency of vitamin E and/or Se for 8 weeks markedly increased D

Brown bowel syndrome: case report and review.

Brown bowel syndrome is characterized by deposits of lipofuscin in the tunica muscularis of the small intestine. Its etiology is associated with chronic malabsorption resulting in a deficiency of vitamin E. This hypovitaminosis is believed to cause a mitochondrial myopathy secondary to loss of the antioxidant properties of vitamin E, which further worsens the malabsorption and leads to atonic, dilated segments of bowel. Current treatment options involve nutritional supplementation, surgical resection of the affected segments, and intestinal transplantation.

Association of serum antioxidants and risk of coronary heart disease in South Indian population.

BACKGROUND AND AIM: Higher prevalence of coronary heart disease (CHD) has been reported in south Indian population, which cannot be accounted for by the traditional risk factors like hyperlipidemia. Identification of new risk factors may help in treatment and prevention of CHD in this part of the world. In an attempt to investigate the causes of increased incidence of CHD in this part of the world, we intended to look for oxidative stress in our patients as a possible risk factor. As an initial step in this perspective, a case- control study was conducted to find out the serum antioxidant leve

Dietary factors in lipofuscinogenesis and ceroidogenesis.

The presence of ceroid pigments in human and animal tissues is associated with numerous pathological conditions in which the main pathogenic factor is the primary or secondary deficiency of vitamin E or imbalances between anti- and pro-oxidants. That oxidative stress, particularly through its consequent lipid peroxidation, plays a capital role in the genesis of ceroid pigments, is supported by numerous in vitro and in vivo studies. Discussed in this presentation are two examples of oxidative stress on ceroidogenesis, namely the in vivo rat model of dietary hepatic necrosis, and the in vitro fo

[Substantiation of the need of fortification with vitamins of the diet of children with gastrointestinal diseases].

Surveys of 28 children have shown that 64 per cent of the children suffering from ulcerous disease of the stomach and the duodemun or chronic duodenitis have a combined deficiency of vitamin E, vitamins of the B group and beta-carotene. Diet No. 1 and No. 5 could not provide daily needs for vitamins. The patients were found to have a lower antioxidant activity of the blood serum, whereas the contents of hydroperoxides, double bonds and malonic dialdehydes were higher than in healthy children. The POL index in children with high beta-carotene deficiency was significantly higher than that in ail

Decreased messenger RNA expression of key markers of the nigrostriatal dopaminergic system following vitamin E deficiency in the rat.

We have evaluated the effect of a vitamin E-deficient diet on the rat nigrostriatal dopaminergic system. After 15 days of deficient diet, the amount and activity of striatal and nigral tyrosine hydroxylase increased, which contrasted with a decreased messenger RNA expression for tyrosine hydroxylase and the dopamine transporter in the ventral mesencephalon. When we prolonged the deficiency of vitamin E for 30 days, dopamine levels did not differ in both areas. In contrast, messenger RNA levels for tyrosine hydroxylase and the dopamine transporter were markedly reduced in 30-day deficient rats.

Effect of different vitamin E levels on lipid peroxidation in streptozotocin-diabetic rats.

Administration of streptozotocin is used to induce diabetes in experimental models, causing a selective destruction of pancreatic beta islet cells associated with generation of free radicals. Supplementation with antioxidant vitamins such as vitamin E is a protective factor against free radicals. The objective of this study was to determine the effect of administration of a diet supplemented with, or deficient in vitamin E to streptozotocin diabetic rats, controlled or not with insulin, on plasma glucose, hepatic vitamin E and hepatic thiobarbituric acid reactive substance (TBARS) levels befor

Vitamin E status of infants at birth.

Preterm infants may be susceptible to chronic lung disease and retinopathy of prematurity because of deficient antioxidant mechanisms including deficiency of vitamin E. The aim of this study was to evaluate the status of the antioxidant vitamin E among preterm and term livebirths. Umbilical cord blood samples collected from 40 preterm and 180 term babies were analyzed for vitamin E levels using high performance liquid chromatography. Linear regression analysis was used to examine the relationship of vitamin E with gestational age, birth weight and appropriateness of weight for gestational age.

Retinal abnormalities in experimental vitamin E deficiency.

Physiological and biochemical studies have been carried out longitudinally over a period of 12 months in vitamin E deficient and control rats to gain an understanding of the mechanism whereby vitamin E conserves normal retinal function. Electroretinographic studies indicated that the primary effect of vitamin E deficiency was on the photoreceptors. Ultrastructural studies, however, did not show any morphological changes to the photoreceptors which could explain receptor dysfunction. A 30-40% loss of vitamin A (retinol) was found to be associated with vitamin E deficiency. This could be correct

Interactions between vitamin C and vitamin E are observed in tissues of inherently scorbutic rats.

To investigate in vivo interactions between antioxidant vitamins C and E, sparing effects of vitamin C on vitamin E as well as those of vitamin E on vitamin C were evaluated using inherently scorbutic [Osteogenic Disorder Shionogi (ODS)] rats. Rats were divided into four groups (control, vitamin E-deficient, vitamin C-deficient and simultaneously vitamins C and E-deficient). The levels of vitamins C and E in tissues were determined at 0, 14 and 21 d of deficiency. On d 14, the vitamin E concentration in plasma, liver, brain and lung of the vitamin C-deficient group was significantly lower than
