Use and safety of elevated dosages of vitamin E in adults.

Subjects with a variety of enteropathies, hemolytic anemias, acute respiratory distress syndrome, hepatitis, Gaucher's disease as well as those on TPN and hemodialysis, often have low ("deficient") blood levels of vitamin E. A deficiency of vitamin E can be manifested by accelerated red blood cell destruction and neuromuscular deficit. Supplementation of these patients may be advisable. Neurological dysfunction has been observed in adults with prolonged vitamin E deficiency resulting from lipid malabsorption. Long-term treatment with high doses of vitamin E results in improvement. Brown bowel syndrome with manifestation in the gastrointestinal tract and thyroid gland.

Brown bowel syndrome (BBS) is a rare disease accompanied by deposits of lipofuscin predominantly in smooth muscle cells of the gastrointestinal tract. To determine whether cells other than smooth muscle cells show pigment deposition we studied biopsies of the stomach, small and large intestine, and thyroid gland of a 52-year-old male with malabsorption syndrome. Light and electron microscopy found lipofuscin pigment in vascular endothelial cells and in smooth muscle cells of the muscularis mucosae, arterioles and venules of the gastrointestinal tract and thyroid gland. All other cells of bowel

Nonheme iron in sickle erythrocyte membranes: association with phospholipids and potential role in lipid peroxidation.

Previous studies documented the abnormal association of heme and heme proteins with the sickle RBC membrane. We have now examined RBC ghosts and inside-out membranes (IOM) for the presence of nonheme iron as detected by its formation of a colored complex with ferrozine. Sickle ghosts have 33.8 +/- 18.2 nmol nonheme iron/mg membrane protein, and sickle IOM have 4.3 +/- 3.0 nmol/mg. In contrast, normal RBC ghosts and IOM have no detectable nonheme iron. The combination of heme and nonheme iron in sickle IOM averages nine times the amount of membrane-associated iron in normal IOM. Kinetics of the

The alpha-tocopherol and phospholipid fatty acid content of rat liver subcellular membranes in vitamin E and selenium deficiency.

(1) Rats were given a diet deficient in vitamin E and selenium, or diets supplemented with either or both of these nutrients. The livers were subfractionated by standard procedures, and the purity of the fractions was assessed by marker enzyme techniques. alpha-Tocopherol was measured and profiles of phospholipid fatty acids were determined. (2) All the organelles studied were severely depleted of alpha-tocopherol in the rats deprived of vitamin E: no organelle was particularly severely depleted. There was a large rise in the alpha-tocopherol content in organelles of rats deprived of selenium

The effect of vitamin E deficiency on the induction of age pigment in various tissues of the mouse.

The effect of a dietary deficiency of vitamin E on the concentrations of lipofuscin in the hippocampus and the supraoptic nucleus (SON) of the hypothalamus, and in adrenal cortical cells was assessed in male mice. The animals were fed either a control diet or the vitamin E deficient diet after 2 months of age for a period of 6 months. There was no significant difference in the growth curves of the 2 groups of animals over the period studied. Fluorescence microscopy and transmission electron microscopy were used to assess the effect of the diet on lipofuscin in the different tissues. Quantitati

Changes in vitamin E concentration in red blood cells and plasma of patients with olivopontocerebellar ataxia within the Schut-Swier kindred.

Many reports have documented the importance of vitamin E for the function of the nervous system, especially of the cerebellum. Therefore, we studied the concentrations of vitamin E in the blood plasma and red blood cells of patients with a hereditary form of olivopontocerebellar ataxia. The concentrations of alpha tocopherol (the principal biologically-active form of vitamin E) in the plasma and red cells of the ataxic subjects were significantly lower than those of unaffected, close relatives as well as unrelated control subjects. Vitamin E deficiency and its clinical significance in adults with primary biliary cirrhosis and other forms of chronic liver disease.

The vitamin E status of 146 adults with chronic liver disease was assessed by estimating both their serum vitamin E concentration and the ratio of serum vitamin E to serum cholesterol concentration. Low levels of vitamin E occurred most frequently in patients with primary biliary cirrhosis and other forms of chronic cholestatic liver disease. When a serum vitamin E concentration of 12.3 mumol/l (mean-2 SD of a control population) was taken as the lower limit of normal, 44% of patients with primary biliary cirrhosis and 32% with other chronic cholestatic liver disease had a reduced concentratio

Studies on the neurobiology of vitamin E (alpha-tocopherol) and some other antioxidant systems in the rat.

To understand why nervous tissue should be particularly affected by severe deficiency of vitamin E, the distribution of vitamin E (alpha-tocopherol) and some other antioxidant systems was investigated. The concentration of vitamin E and the activities of glutathione peroxidase and superoxide dismutase were determined in different regions of the nervous system in male Wistar rats. The cerebral cortex had the highest, and the cerebellum the lowest concentration of alpha-tocopherol (P less than 0.02). Activity of glutathione peroxidase tended to show an inverse relationship to the alpha-tocophero

Vitamin E.

Vitamin E activity was first identified as a dietary factor essential for reproduction in the rat. It is now known that this vitamin has a far wider range of functions in the body than its role in fertility. It interacts with selenium-containing glutathione peroxidase to prevent the oxidative breakdown of tissue membranes associated with the hydroperoxides of polyunsaturated fatty acids. Relationships with other factors such as stress and vitamin C, have been proposed. The symptoms of deficiency of vitamin E vary according to species. With so many variables it is difficult to estimate the opti

Deficiency of vitamin E in the alveolar fluid of cigarette smokers. Influence on alveolar macrophage cytotoxicity.

Cigarette smoking produces oxidant-mediated changes in the lung important to the pathogenesis of emphysema. Since vitamin E can neutralize reactive oxygen species and prevent peroxidation of unsaturated lipids, it may constitute an important component of the lung's defense against oxidant injury. To better characterize the antioxidant protective role of vitamin E, young asymptomatic smokers and nonsmokers were evaluated by bronchoalveolar lavage before and immediately after a 3-wk course of oral vitamin E (2,400 IU/d). Smoker alveolar fluid at baseline was relatively deficient in vitamin E com

[Effect of vitamin E from by-products of cotton oil processing on the glutathione peroxidase system in rats].

The activities of glucose-6-phosphate dehydrogenase, glutathione reductase and glutathione peroxidase from liver, skeletal muscles and erythrocytes of rats fed a vitamin E-deficient, or supplemented, diet were studied. Vitamin E was added in the diet either as a pure pharmacy form of alpha-tocopherol or as a tocopherol mixture derived from oil wastes. The deficiency of vitamin E caused an increase in the activity of the above mentioned enzymes. Both alpha-tocopherol and the tocopherol mixture were found to influence the glutathione peroxidase system. The dose-dependent response of the glutathi

Effects of vitamin E and selenium deficiencies on rat immune function.

The effects of dietary restriction of vitamin E and selenium were studied in male Long-Evans hooded rats. Weanling animals were maintained for 5-6 weeks on torula yeast-based diets, with or without the addition of vitamin E (150 IU/kg) or selenium (0.5 mg/kg), to form the following dietary groups: +E, +Se; +E, -Se; -E, +Se; -E, -Se, and a fifth group pair-fed with the -E, -Se group. This latter group exhibited a decreased rate of growth similar to the -E, -Se group. Lymphocyte blastogenesis in response to mitogens was decreased in animals fed the diets deficient in either vitamin E or selenium

Measurement of vitamin E in serum and plasma by high performance liquid chromatography with electrochemical detection.

A deficiency of vitamin E has been associated with a wide variety of pathological conditions, many of them involving neonates. Previously published methods for the quantitation of vitamin E require large sample volumes as well as extensive extraction and concentration procedures. The present work describes a method for the quantitation of vitamin E in small volumes (less than or equal to 50 microliters) of serum or plasma without the need for extraction or concentration of the sample. With further optimization, even smaller volumes (less than or equal to 10 microliters) can be employed. Sample

Effect of dietary vitamin E on nitrite-treated rats.

The effect of dietary vitamin E on cellular responses to nitrite was studied in rats. One-month-old male Sprague-Dawley rats were fed a basal vitamin E-deficient diet with or without 100 ppm vitamin E and 1000 ppm sodium nitrite (NaNO2) for 9 weeks. In addition to a high mortality rate, nitrite-fed rats maintained on a vitamin E-deficient diet exhibited a marked increase in liver necrosis, tubular nephrosis and myodegeneration, as well as greater biochemical and hematological alterations when compared to the control animals. No animal mortality or histopathologic lesions in any tissues were ob

Influence of systemic factors on experimental epileptic brain injury. Structural changes accompanying bicuculline-induced seizures in rats following manipulations of tissue oxygenation or alpha-tocopherol levels.

A previous study from the laboratory showed that status epilepticus induced by bicuculline administration to ventilated rats produced astrocytic swelling and nerve cell changes ("type 1 and 2 injury") particularly in layers 3 and 5 of the neocortex (Söderfeldt et al. 1981). The type 1 injured neurons were characterized by condensation of cyto- and karyoplasm and the less common type 2 cells were characterized by swelling of endoplasmic reticulum including the nuclear envelope. In the present study we explored whether changes in cerebral oxygen availability altered the extent or character of th

Vitamin E deficiency during chronic childhood cholestasis: presence of sural nerve lesion prior to 2 1/2 years of age.

Vitamin E malabsorption and deficiency during chronic childhood cholestasis has been associated with a progressive ataxic neurologic syndrome. Hyporeflexia, the first sign of neurologic dysfunction, may begin prior to age 2 years, but severe symptoms do not develop until age 5 to 10 years. To establish the age of onset of neuropathologic lesions, we prospectively evaluated four young children with severe cholestasis. Malabsorption and deficiency of vitamin E were documented by low serum vitamin E concentrations, low serum vitamin E to total serum lipids ratios, elevated hydrogen peroxide hemol

Mechanism causing vitamin E deficiency during chronic childhood cholestasis.

In order to characterize the mechanism(s) causing vitamin E deficiency during chronic childhood cholestasis, we studied 6 vitamin E-deficient cholestatic children with clinical evidence of neurologic dysfunction (group A), 4 vitamin E-sufficient cholestatic children and young adults with normal neurologic status (group B), and 6 vitamin E-sufficient noncholestatic children (group C). Intestinal absorption of dl-alpha-tocopherol (assessed by an oral tolerance test) was markedly impaired (p less than 0.001) in group A compared with groups B and C, which did not differ from each other. Vitamin E deficiency due to increased consumption in beta-thalassemia and in Gaucher's disease.

Plasma vitamin E levels were found to be decreased (less than 0.5 mg) in thalassemia and in 17 out of 20 patients with Gaucher's disease, where the levels were two standard deviations below the normal mean value. In the latter, the decrease in vitamin E levels correlated with the severity of the clinical expression of the disease and correlated inversely with the degree of hepatosplenomegaly and serum tartrate-resistant acid phosphatase activity. In both diseases, there was no evidence for intestinal malabsorption of the lipid-soluble vitamin. In spite of the different etiology, pathophysiolog
