Dietary zinc deficiency regulates the ROS/TLR4/NF-κB pathway to induce pancreatic inflammation and cell death in mice.

Zinc (Zn) deficiency disrupts redox homeostasis in the body. The pancreas is a vital digestive and endocrine organ of the body, and its normal functional operation cannot proceed without the involvement of Zn. In this study, we established in vivo mouse models, including the normal Zn group (CG, 34 mg Zn/kg), Zn-deficient group (LZn, 2 mg Zn/kg), and Zn-supplemented group (HZn, 100 mg Zn/kg), as well as an in vitro Zn-deficient model of Mouse INsulinoma 6 (MIN6) cells. We systematically investigated the effects of Zn deficiency on pancreatic oxidative stress, inflammation, and cell death. The

A requirement for Acinetobacter baumannii purine biosynthesis during lung infection is exacerbated by host zinc deficiency.

Acinetobacter baumannii is a leading cause of hospital-acquired infections, including ventilator-associated pneumonia. Dietary zinc deficiency is a major risk factor for pneumonia, and hospitalized patients at risk for A. baumannii infection have increased rates of zinc deficiency. We previously showed that dietary zinc deficiency enhanced A. baumannii pneumonia pathogenesis via an IL-13-dependent mechanism in mice. Here, we identified A. baumannii genes required for proliferation in the lungs of zinc-deficient mice using a genome-wide transposon sequencing (Tn-seq) screen. In zinc-deficient m

Combined olfactory bulbectomy and zinc deficiency induce complex neurobiological alterations resistant to classical antidepressant treatments: Insights from proteomic and neurochemical analyses.

Major depression (MDD) is a complex and multifactorial mental illness. Despite advances in treatment, about 30% of patients still do not see improvement, underscoring the need for new insights into its pathophysiology. Clinical studies suggest that zinc deficiency may be an essential risk factor for the development of depressive disorder and may also contribute to reduced antidepressant efficacy. We developed a novel animal model combining olfactory bulbectomy (OB) with dietary zinc deficiency (ZnD; 3 mg Zn/kg) to further explore this relationship. After three weeks of OB + ZnD treatment, rats

Acute Dietary Zinc Deficiency Impairs Sperm Motility but Not Gonadal mRNA Methylation in Mice.

Zinc deficiency is associated with impaired male fertility and with reduced DNA and histone methylation in oocytes, but the temporal relationship between acute dietary zinc deficiency and sperm function is unclear. To fill in these gaps, the effects of acute dietary zinc deficiency on sperm function were tested. Five-week-old F1 (DBA × C57BL/6) male and female mice were fed a control diet (29 mg Zn/kg) or zinc-deficient diet (< 1 mg Zn/kg) for 6 days (n = 11/group), a duration selected to model acute zinc depletion based on previous studies demonstrating rapid reproductive defects within a sim

Effects of short term dietary zinc deficiency and zinc supplementation on nitro-oxidative stress in testes of Wistar rats.

Zinc is crucial for several cellular functions in the biological system. Zinc insufficiency is one of the most prevalent types of micronutrient malnutrition in the world. Present study was conducted to detect testicular free radical levels i.e. lipid peroxidation (LPO), hydroperoxides, hydroxyl radical (OH.), nitric oxide (NO) and peroxynitrite (ONOO-) after short term dietary zinc deficiency and zinc supplementation. Pre-pubertal rats (n = 144) were divided into two groups with 6 sub-groups viz. zinc control (ZC, 100 µg/g zinc diet), pair-fed (PF, 100 µg/g zinc diet), zinc deficient (ZD, < 1.

Severe Dietary Zinc Deficiency Does Not Significantly Alter Energy Balance in Adult Mice.

Zinc (Zn) is an essential dietary nutrient metal that functions as a cofactor for numerous enzymes involved in diverse cellular processes, including energy metabolism. In humans, Zn deficiency afflicts an estimated one-third of the global population and is a prominent risk factor for numerous diseases, including the development of obesity and diabetes. It is known that severe Zn deficiency leads to impaired growth and development in animals, suggesting that this dietary micronutrient is required for the maintenance of organismal energy balance. However, the impact of Zn restriction on energy b

Multi-disciplinary collaboration in diagnosing thymic hyperplasia in a Graves' disease patient.

ABSTRACT: Dietary zinc deficiency disrupts fertility in vivo by impairing oocyte and embryo development near ovulation. Acute treatment of newborn ovaries with a strong intracellular chelator (TPEN), which preferentially binds zinc, disrupts follicular development. However, the chronic effects of transition metal chelation on preantral follicle development are not known. In this study, the effect of the extracellular transition metal chelator, diethylenetriaminepentaacetic acid (DTPA), was used to examine more prolonged effects on preantral follicle development. Preantral granulosa cell-oocyte

Genetic determinants of zinc homeostasis and its role in cardiometabolic diseases.

Zinc is essential for many physiological processes and its deficiency is highly prevalent worldwide. Its complex homeostasis involves membrane transporters from the SLC39/ZIP and SLC30/ZnT protein families. We conducted a genome-wide association study (GWAS) meta-analysis of urinary zinc levels in three European-ancestry cohorts (N = 10,113), followed by in silico and in vivo studies to elucidate their underlying public health and physiological relevance. We identified eleven genome-wide significant signals with six mapping to SLC39/ZIP and SLC30/ZnT gene regions. The lead signal (rs3008217C>G

A Short-Term Zinc-Deficient Diet Maintains Serum Calcium Concentrations through Ca Absorption-Related Gene Expression in Rats.

We investigated the effects of short-term dietary zinc deficiency on zinc and calcium metabolism. Four-week-old male Wistar rats were divided into two pair-fed groups for a 1-wk treatment: zinc-deficient group (ZD, 1 ppm); control group (PF, 30 ppm). The mRNA expression of zinc transporters, such as Slc39a (Zip) 4, Zip5, Zip10, and Slc30a (ZnT) 1, in various tissues (liver, kidney, and duodenum) quickly responded to dietary zinc deficiency. Although there was no significant difference in serum calcium concentrations between the PF and ZD groups, serum 1,25-dihydroxycholecalciferol (1,25(OH)2D3

Zinc deficiency deteriorates ovarian follicle development and function by inhibiting mitochondrial function.

Zinc (Zn) is a crucial trace element essential for human growth and development, particularly for reproductive health. Previous research has shown a decrease in serum zinc concentration with age and individuals with conditions such as polycystic ovary syndrome (PCOS) and diabetes mellitus. However, the specific effects of zinc deficiency on the female reproductive system, especially ovarian function, are not fully understood. In our study, we observed a significant reduction in the total number of follicles and mature follicles in the zinc deficiency group. This reduction correlated with decre

Paternal zinc deficiency alters offspring metabolic status in Drosophila melanogaster.

BACKGROUND: This study delves into the understudied yet potentially crucial role of paternal zinc deficiency in programming offspring metabolic outcomes. By examining paternal zinc deficiency, we aim to shed light on a previously unexplored avenue with the potential to significantly impact future generations. We investigated the intergenerational effects of paternal zinc deficiency on metabolic parameters in Drosophila melanogaster. METHODS: Dietary zinc deficiency was induced by supplementing the diet of Drosophila F0 male flies with TPEN (N,N,N',N'-tetrakis(2-pyridylmethyl)ethylenediamine)

Dietary zinc deficiency promotes Acinetobacter baumannii lung infection via IL-13 in mice.

Dietary zinc deficiency is a major risk factor for pneumonia. Acinetobacter baumannii is a leading cause of ventilator-associated pneumonia and a critical public health threat due to increasing rates of multidrug resistance. Patient populations at increased risk for A. baumannii pneumonia are also at increased risk of zinc deficiency. Here we established a mouse model of dietary zinc deficiency and acute A. baumannii pneumonia to test the hypothesis that host zinc deficiency contributes to A. baumannii pathogenesis. We showed that zinc-deficient mice have significantly increased A. baumannii b

Proteomics analysis in rats reveals convergent mechanisms between major depressive disorder and dietary zinc deficiency.

BACKGROUND: Preclinical and clinical studies have shown that dietary zinc deficiency can lead to symptoms similar to those observed in major depressive disorder (MDD). However, the underlying molecular mechanisms remain unclear. To investigate these mechanisms, we examined proteomic changes in the prefrontal cortex (PFC) and hippocampus (HP) of rats, two critical brain regions implicated in the pathophysiology of depression. METHODS: Rats were fed diets either adequate in zinc (ZnA, 50 mg Zn/kg) or deficient in zinc (ZnD, <3 mg/kg) for four weeks. High-throughput proteomic analysis was used t

Single-cell RNA sequencing unravels the transcriptional network underlying zebrafish retina regeneration.

Mechanical ventilation is necessary to support patients with acute lung injury, but also exacerbates injury through mechanical stress-activated signaling pathways. We show that stretch applied to cultured human cells, and to mouse lungs in vivo, induces robust expression of metallothionein, a potent antioxidant and cytoprotective molecule critical for cellular zinc homeostasis. Furthermore, genetic deficiency of murine metallothionein genes exacerbated lung injury caused by high tidal volume mechanical ventilation, identifying an adaptive role for these genes in limiting lung injury. Stretch i

Identification of novel compound heterozygous variants in the SLC30A7 (ZNT7) gene in two French brothers with stunted growth, testicular hypoplasia and bone marrow failure.

Zinc is an essential trace mineral. Dietary zinc deficiency results in stunted growth, skin lesions, hypogonadism and frequent infections in humans. Mice genetically lacking Slc30a7 suffer from mild zinc deficiency and are prone to development of prostate cancer and insulin resistance. Disease-causing variants or mutations in the human SLC30A7 (ZNT7) gene have not been previously reported. Here, we describe two-boy siblings from a French family with stunted growth, testicular hypoplasia and bone marrow failure. Exome sequencing revealed compound heterozygous variants in ZNT7 consisting of NM_1

Neurodevelopmental Consequences of Dietary Zinc Deficiency: A Status Report.

Zinc is a tightly regulated trace mineral element playing critical roles in growth, immunity, neurodevelopment, and synaptic and hormonal signaling. Although severe dietary zinc deficiency is relatively uncommon in the United States, dietary zinc deficiency is a substantial public health concern in low- and middle-income countries. Zinc status may be a key determinant of neurodevelopmental processes. Indeed, limited cohort studies have shown that serum zinc is lower in people diagnosed with autism spectrum disorder (ASD), attention-deficit/hyperactivity disorder (ADHD), and depression. These o

Dietary Zinc Deficiency Induces Cav3.2-Dependent Nociceptive Hypersensitivity in Mice.

Cav3.2 channels belong to the T-type calcium channel (T-channel) family, i.e., low voltage-activated calcium channels, and are abundantly expressed in the nociceptors, playing a principal role in the development of pathological pain. The channel activity of Cav3.2 is suppressed by zinc under physiological conditions. We thus tested whether dietary zinc deficiency would cause Cav3.2-dependent nociceptive hypersensitivity in mice. In the mice fed with zinc deficient diet for 2 weeks, plasma zinc levels declined by more than half, and mechanical allodynia developed. The dietary zinc deficiency-in
