Effect of dietary zinc intake on the hematological profile of the rat.

The effect of dietary zinc deficiency (imposed at weaning) on the hematological profile of the male rat was studied. Lack of dietary zinc resulted in elevated numbers of erythrocytes and segmented neutrophils and decreased reticulocyte number. The hematocrit of zinc-deficient rats was significantly elevated after the first week. The mean lifespan of erythrocytes from zinc-deficient rats (tested in adult control rats) was not significantly altered from that of controls. The lack of dietary zinc did not cause a major change in the erythrocyte density (age) distribution.

Role of zinc deficiency in carcinogenesis.

Zinc is a trace element required for the growth of normal and neoplastic tissues in a variety of species. Zinc deficiency is associated with alterations in the activity of zinc-dependent enzymes essential for cell replication. Dietary zinc deficiency also increases the incidence of certain tumors while decreasing the incidence of others. The mechanism by which zinc deficiency alters carcinogenesis is not fully understood. Among those tumors whose incidence is increased by dietary zinc deficiency are carcinomas induced by dialkylnitrosamines. This class of carcinogens requires microsomal cytoch

Effect of dietary zinc deficiency on the accumulation of cadmium and metallothionein in selected tissues of the rat.

The effect of continuous dietary zinc deficiency on the metabolism of the toxic heavy metal cadmium has not been widely studied. This investigation was designed to assess the effects of subadequate dietary zinc intake on the accumulation of dietary cadmium and on metallothionein (MT) and zinc concentrations in target organs of cadmium toxicity. Adult male Wistar rats (180-200 g) were allowed, ad libitum, diets either adequate (60 ppm) or deficient (7 ppm) in zinc for a total of 9 wk. The zinc-deficient diet resulted in an approximately 40% reduction in plasma zinc (assessed at 3, 6, and 9 wk)

Genes, manganese, and zinc in formation of otoconia: labeling, recovery, and maternal effects.

Published studies indicate that genes and dietary manganese deficiency cause vestibular defects and ataxic behaviors. Manganese deficiency during development causes otoconial defects in mice, rats, guinea pigs, and chicks. Mutant genes cause otoconial defects in mice, mink, and poultry. Manganese supplementation prevents the otoconial defects in some mutant mice and mink. Manganese is essential, before crystallization of the otoconia, for synthesis of mucopolysaccharides and otoconial matrix. Such defects can be induced, after otoconia are crystallized during fetal development, by dietary zinc

The effect of N-nitrosodimethylamine and N-nitroso-N-benzylmethylamine on [3H]thymidine incorporation into the DNA of target and non-target tissues in the zinc-deficient rat.

The influence of dimethylnitrosamine (NDMA), a liver carcinogen and nitrosobenzylmethylamine (NBMA) as esophageal carcinogen on [3H]thymidine incorporation into DNA was studied in the esophagus, liver, forestomach and gastric-stomach of fasted zinc-deficient and pair-fed zinc-sufficient rats, measured 1 h after the thymidine injection. In the untreated animals, dietary zinc deficiency significantly depressed [3H]thymidine incorporation (89%) into the DNA of forestomach only. NDMA, administered 4 h before death at 30 mg/kg, produced 50-55% inhibition in [3H]thymidine incorporation in the esopha

Influence of dietary zinc deficiency and parenteral zinc on rat liver fructose 1,6-bisphosphatase activity.

Fructose 1,6-bisphosphatase activity in liver of rats fed a zinc deficient diet was decreased to 60% of that in zinc adequate controls. Activity in the zinc deprived rats was not restored to control values by in vitro addition of EDTA. When a physiological dose of zinc was tube fed to the depleted rats, activity increased approximately 150% within 0.5 hr of the dose, and by 1 hr plateaued to a level seen in zinc adequate controls. A significant transient decrease in activity occurred following an intraperitoneal zinc load. This is reversible by in vitro addition of EDTA. These results suggest

Effect of dietary zinc on endogenous free radical production in rat lung microsomes.

The objective of this study was to investigate the effect of dietary zinc on endogenous production of free radicals in lung and liver microsomes. Male weanling rats were fed a zinc-deficient basal diet containing less than 1.1 ppm zinc, or were pair-fed or fed ad libitum a zinc-adequate diet supplemented with 100 ppm zinc. The isolated microsomes (100,000 X g precipitate) of lung and liver were incubated with 0.1 M PBN (spin trap) and 0.3 mM NADPH (cofactor) at 37 degrees C for 1.0 h. A carbon-centered free radical (aN = 16.0 G, aH beta = 3.4 G) was trapped in both lung and liver microsomes. T

Decreased glutathione transferase activities in zinc-deficient rats.

Weanling rats fed a zinc-deficient diet for 56 days exhibit a decrease in the total activities of glutathione transferases with 1-chloro-2,4-dinitrobenzene (CDNB) as substrate in the liver, esophagus and stomach. The level of acid-soluble sulfhydryl group, on the other hand, is decreased in the liver, but elevated in the esophagus and kidney. Deficient rats fed a zinc-replenished diet for just 7 days have a fully restored glutathione transferase activity in all tissues except the liver. In view of our earlier findings that the carcinogenic effects of nitrosamines are enhanced in zinc-deficient

Effects of dietary zinc and copper on free radical production in rat lung and liver.

The effects of dietary copper and zinc on free radical production in lung and liver microsomes were studied in male weanling rats. The rats were fed for 6 weeks on one of seven diets, with different copper and zinc concentrations representing low, adequate, and high dietary levels of copper and low and adequate levels of zinc. Rats were put on diets arranged in a 3 X 2 factorial design with copper and zinc supplementations of 0, 15, and 500 mg/kg and 0.5 or 100 mg/kg, respectively. The low copper diet depressed copper levels in both the lungs and liver, although zinc levels were unchanged in r

Histopathology of methylbenzylnitrosamine-induced esophageal carcinoma in the rat: comparison with cytomorphology.

The incidence of human carcinoma of the esophagus is increased in association with environmental nitrosamines and dietary zinc deficiency. For examination of the role of zinc deficiency in esophageal carcinogenesis, methylbenzylnitrosamine [(MBN) CAS: 937-40-6] was used to induce esophageal carcinoma in rats fed either a zinc-deficient or control diet. Histologic examination of the MBN-treated Sprague-Dawley rat esophagi revealed a significantly higher incidence of esophageal carcinoma in the zinc-deficient rats vs. controls (62 vs. 33%). In addition, three distinct cytologic and histologic pa

Requirement of zinc in human subjects.

Essentiality of zinc in nutrition of higher animals was established in 1934. Dietary zinc deficiency in humans was recognized in 1961. Dietary requirements for zinc have been estimated factorially and by balance studies. Factors that influence dietary zinc requirement include dietary and other substances that either facilitate or inhibit absorption and retention of zinc; and metabolic phenomena that influence retention or excretion of the element. These determinants must be considered in estimating the requirement and the recommended dietary allowance for this essential element. An approach th

The effects of zinc deficiency on developing photoreceptors in the rat retina: a scanning microscopic study.

A study was undertaken to observe the effects of dietary zinc deficiency on the topographical morphology of developing photoreceptor (rod) cells in the sensory retina of the rat. Observations were made by means of scanning electron microscopy (SEM). Pups were utilized from three groups including (1) zinc deficient (2) pair-fed controls and (3) ad libitum controls. Animals ranged in age from birth to twenty days. Femur zinc levels were measured by flame atomic absorption spectrophotometry. A statistically significant increase in the number of photoreceptor cells continued through the 6th day, h

Zinc deficiency and the development of esophageal and forestomach tumors in Sprague-Dawley rats fed precursors of N-nitroso-N-benzylmethylamine.

Nine-week-old zinc-sufficient (100 mg zinc/kg feed) and zinc-deficient (7 mg zinc/kg feed) noninbred male Sprague-Dawley rats were given free access 5 days a week to deionized drinking water containing low (0.05%) or high (0.25%) quantities of benzylmethylamine (BMA) and concurrently 0.5% NaNO2. In contrast to the action of the preformed carcinogen N-nitroso-N-benzylmethylamine, which almost invariably produced esophageal tumors, oral administration of its precursors, BMA and NaNO2, resulted in forestomach tumors as well. In both the high- and low-BMA groups given precursors for 16 weeks, the

Edema, hypoproteinemia, and zinc deficiency in low-birth-weight infants.

Three premature infants with zinc deficiency who had an unusual presentation with generalized edema and hypoproteinemia between 5 and 9 weeks of age are described. The infants were fed their own mother's milk, supplemented with a proprietary formula after the first 2 to 3 weeks of life. None of the infants had diarrhea, liver disease, or urinary protein loss. Treatment with oral zinc supplements led to rapid resolution of the edema, with an increase in values for serum proteins, alkaline phosphatase, and zinc. There was no recurrence of symptoms following discontinuation of zinc therapy 1 mont

Forestomach squamous papillomas in the rat: effect of dietary zinc deficiency on induction.

Administration of N-nitrosodimethylamine (NDMA) to zinc deficient rats intragastrically twice weekly for 8 weeks resulted in the development of forestomach tumors in 63% of treated animals. The tumors were frequently found at the junction between the fore and glandular stomach and were multiple. Smaller numbers of tumors were also present in the more proximal parts of the forestomach. All tumors were squamous papillomas. Similar tumors were not observed in zinc sufficient pair-fed rats given NDMA. These findings showed that NDMA, a non-esophageal and non-forestomach carcinogen in the rat, was

Changes in the levels of marker expression by mononuclear phagocytes in zinc-deficient mice.

To define the effects of dietary zinc deficiency on mononuclear phagocytes young adult A/J mice were fed zinc-deficient, zinc-adequate, or restricted amounts of a zinc-adequate diet for 28 days. The deficiency did not affect the total number or the percentage of nonspecific esterase (NSE)-positive cell populations in the blood or peritoneal cavity of the mice; however, the total numbers of leukocytes and NSE-positive cells in the spleen were reduced in the deficient and restricted mice to about 50% of the zinc-adequate group. In the peripheral blood of deficient mice, the percentages of adhere

Esophageal microsomal metabolism of N-nitrosomethylbenzylamine in the zinc-deficient rat.

Epidemiological studies in China suggest that dietary zinc deficiency and environmental exposure to N-nitrosamine carcinogens, such as N-nitrosomethylbenzylamine (NMBA), are among the factors associated with an increased incidence of esophageal carcinoma in humans. NMBA belongs to a class of nitrosamines which require activation by the cytochrome P-450-dependent mixed-function oxidases in order to be mutagenic. Rats maintained on a zinc-deficient diet exhibited an increased incidence of NMBA-induced esophageal carcinoma when compared to rats on a control diet. The increased tumor formation was
