More than just anemia: Uncovering celiac disease beneath the surface: A case report.

Celiac disease (CD) is an immune-mediated disorder triggered by gluten in genetically susceptible individuals. Although iron deficiency anemia is a common presentation, other micronutrient deficiencies may occur. We describe a 3.5-year-old female whose initial presentation of CD was severe, asymptomatic normocytic anemia secondary to copper (Cu) deficiency. She presented with mild abdominal symptoms, weight loss, and profound anemia (hemoglobin as low as 1.6 g/dL) requiring transfusions. Extensive infectious, inflammatory, hematologic, and oncologic evaluations were unrevealing. Workup demonst

Copper-molybdenum interactions in ruminant nutrition and reproductive physiology: mechanistic insights into growth, digestibility, and follicular development.

The copper-molybdenum-sulfur (Cu-Mo-S) interaction is one of the most important mineral antagonisms in ruminant nutrition because it can induce functional copper deficiency even when dietary copper intake appears adequate. In the rumen, dietary sulfur is reduced to sulfide, which reacts with molybdate to form thiomolybdates. These compounds bind copper within ruminal digesta and, when produced in excess, may also enter the circulation and interfere with systemic copper trafficking and cuproenzyme function. This review critically synthesizes evidence on ruminal thiomolybdate formation, post-abs

Iron Supplementation and Copper Homeostasis: Evidence for Potential Copper Depletion, Biomarker Challenges, and Clinical Implications.

Iron deficiency remains a major nutritional problem, particularly among women of reproductive age, pregnant women, infants, and children. Oral iron supplementation is the principal strategy for prevention and treatment, but the biological interdependence of iron and copper metabolism raises concern that prolonged or excessive iron exposure may impair copper availability in susceptible individuals. This review summarizes human and animal evidence linking iron supplementation or high dietary iron intake to altered copper homeostasis, with emphasis on underlying mechanisms, biomarker limitations,

Impact of copper deficiency in cows on immune system function in offspring: a study using lymphocyte immunophenotyping.

INTRODUCTION: Copper (Cu) deficiency is a common, often subclinical problem in dairy cattle herds that can adversely affect haematopoietic processes and immune function. This study aimed to evaluate the effect of hypocupraemia in cows on selected haematological parameters and identify the immunophenotype of calf lymphocytes. MATERIAL AND METHODS: The study included Holstein-Friesian cows and their calves aged two to six weeks, which were divided into two groups: a hypocupraemic group (17 cows and 17 calves) and a normocupraemic group (18 cows and 18 calves). Haematological and biochemical ana

Severe Anemia and Neutropenia Associated with Copper Deficiency in Three elderly Adults Residing in Nursing Homes.

This report describes three cases of severe anemia and neutropenia associated with copper deficiency. All patients were octogenarian women who presented with cognitive impairment and resided in nursing homes. The rapid response to low-dose oral copper supplementation indicates that their copper deficiency was caused by a poor intake rather than the malabsorption of copper. This case series demonstrates that severe copper deficiency can occur in the absence of typical underlying conditions, such as gastrointestinal surgery or an excessive zinc intake. Our experience suggests incorporating serum

Iron and Other Metal Ions in Human Health and Disease.

Iron is the most abundant trace element, driving both the commonest deficiency and ferroptosis. This review takes iron as the central theme and extends to copper, zinc, and calcium, spanning physiological functions, pathological mechanisms, risk factors, diagnostics, and therapeutics. We classify these ions into redox-active (iron, copper), which engage in catalytic electron transfer, and redox-inert (zinc, calcium), which serve structural and signaling roles without redox cycling. Iron deficiency remains the global leading micronutrient deficit, whereas iron overload triggers ferroptosis-medi

Restoring brain functions in the Mo-blo mouse model of Menkes disease with copper nanoclusters.

Menkes disease, caused by ATP7A mutations, leads to severe copper deficiency and fatal neurodegeneration. Current copper-histidine therapy fails to restore brain function. Copper nanoclusters (CuNCs) were designed for their ultrasmall size to cross physiological barriers and deliver copper directly to the whole organism, including the brain. In Mo-blo mice (ATP7A deficient), daily subcutaneous CuNCs markedly improved survival, normalized cuproprotein activities, and restored cognitive and motor functions. Unlike other treatments, CuNCs effectively penetrate the blood-brain barrier, deliver cop

Charge Ordering and Incommensurate Modulations in the Metamagnetic Layered Manganese Oxysulfide Sr2MnO2Cu3.5S3.

Sr2MnO2Cu3.5S3 contains mixed-valent Mn ions Mn2+/3+ in axially elongated MnO4S2 octahedra connected via apical sulfide anions to copper-deficient antifluorite-type Cu4-δS3 layers where δ ∼ 0.5. Copper deficiency is charge-compensated by oxidation of Mn 3d states resulting in mixed-valency. The compound is tetragonal in P4/mmm at ambient temperatures (a = 4.016345(1) Å, c = 11.40708(5) Å). Below 190 K, superlattice reflections in diffraction data and an increase in resistivity, signal checkerboard charge-ordering of Mn2+ and Mn3+. A C. elegans model of copper deficiency: Dietary interventions rescue CTR1/CHCA-1 copper transporter mutant phenotype.

Copper is an essential micronutrient for all living organisms. Mutations in the copper-importing transporter CTR1/CHCA-1 are associated with a severe copper deficiency disorder in humans, for which no effective cures are currently available. Here, we develop C. elegans as a model for copper deficiency. We show that chca-1 mutant worms fed HT115 bacterial diet exhibited a severe developmental phenotype resulting from copper deficiency, reminiscent of the symptoms observed in human patients. Remarkably, this phenotype can be rescued by switching to OP50 bacterial diet or by supplementing HT115 b

Diagnostic pitfalls of ESA-resistant anemia due to functional copper deficiency in a dialysis patient: a myelodysplastic syndrome mimic.

Copper deficiency is a rare yet reversible cause of anemia and bone marrow dysplasia that can mimic myelodysplastic syndrome (MDS), especially in patients on maintenance dialysis, who are at increased risk due to trace element losses, chronic inflammation, and impaired absorption. We report a 69-year-old woman on long-term hemodialysis with a history of partial gastrectomy who developed macrocytic anemia refractory to erythropoiesis-stimulating agents (ESAs). Bone marrow examination revealed erythroid dysplasia, hypocellularity, and ring sideroblasts. Although serum copper levels were initiall

Ambivalent Copper: Mechanistically Distinct Immune Effects Driving Innovation in Cancer Nanomedicine.

Copper (Cu) is an essential element required by all living cells, where it supports critical enzymatic and signaling functions. In cancer, this balance is often disrupted, creating vulnerabilities that can be therapeutically exploited. Changes in Cu availability have been shown to influence key immunoregulatory pathways, including those involved in inflammation, cell death, and immune evasion. Notably, Cu can drive expression of programmed death ligand 1 (PD-L1), contributing to immunosuppression, while also promoting immunogenic cell death, which stimulates adaptive immune responses. These du

Distinct signaling mechanisms and proteome phenotypes are elicited by compartment-specific genetic defects of copper homeostasis.

Impairments to the complex machinery regulating copper homeostasis lead to neurodevelopmental diseases, demonstrating the importance of copper for neuronal health and maintenance. The exact mechanisms by which the brain responds to copper deficiency following disruptions to the copper transporters ATP7A and CTR1 in conditions such as Menkes disease remain unclear, though failure to supply complex IV of the respiratory chain with copper is suspected to account for substantial pathology. Here, we studied mechanisms of copper deficiency using systems biology approaches to contrast isogenic CTR1-

Copper Replacement: A Life-Saving Treatment.

Anemia is the hematological hallmark of copper deficiency; however, the infrequency with which it is encountered makes copper deficiency a formidable diagnostic challenge. We present the case of a 71-year-old woman who is a Jehovah's Witness with a history of cardiomyopathy, end-stage renal disease on peritoneal dialysis, prior gastric bypass surgery, and chronic anemia, who presented with generalized weakness. She was initially diagnosed with anemia in 2003, with a hemoglobin level of 10 g/dL. Her anemia progressively worsened over time, prompting a thorough gastroenterology evaluation with n

SLC31A1 exon 1 methylation reduces intracellular copper ion and promotes diabetic cardiac fibrosis.

BACKGROUND: Metal homeostasis is linked to cellular organelles modification. Dynamic lipid droplet (LD)-mitochondria interactions contribute to diabetes-related complications. However, little is known about whether and how copper regulates LD-mitochondria contacts in diabetic cardiac fibrosis (CF). METHODS: Leptin receptor-deficient (db/db) mice were used to induce diabetic CF. To investigate target gene function in vivo, AAV9 vectors carrying the cardiac fibroblast-specific POSTN promoter-driven small hairpin RNAs were administered. SLC31A1 overexpression was achieved via AAV9 plasmid specifi

Evaluation of novel assays of non-ceruloplasmin copper to monitor chelation treatment in patients with Wilson disease.

BACKGROUND & AIMS: We examined the use of two newer measures of non-ceruloplasmin-bound copper (NCC) and 24-h urinary copper excretion (UCE) to monitor chelation therapy in clinically stable Wilson disease (WD). METHODS: We post hoc analyzed data from the CHELATE study, in which 77 patients with clinically stable WD taking penicillamine (DPA) entered a 12-week screening phase after which 53 were randomized to continued DPA or same dose trientine-tetrahydrochloride (TETA4) (weeks 12-60). Data included NCC measured by protein speciation (NCC-Sp), exchangeable copper (NCC-Ex), and UCE. RESULTS:

Dynamic PET Imaging of Elesclomol-Facilitated Copper Delivery in Ischemic Stroke Rats.

Copper is an essential trace element that is required at optimal levels to support physiological functions in the cerebrovascular system. Copper deficiency reduces vascular elasticity, potentially worsening the ischemic stroke. Epidemiological evidence has linked higher copper intake with reduced stroke risk. This study aimed to investigate Elesclomol (ES) as a vector for delivering copper to the brain in models of ischemic stroke. 64Cu-ES showed excellent serum stability in vitro. PET/CT imaging revealed that 64Cu-ES exhibited longer brain retention compared to 64Cu-ATSM, likely due to its hi

Copper deprivation reprograms antioxidant defense to suppress ferroptosis via SLC7A11.

Copper is an essential trace element that governs diverse cellular functions and influences cell fate. However, how cells adapt to copper deprivation remains poorly understood. Here, we identify a copper-ferroptosis regulatory axis mediated by the cystine transporter SLC7A11. We show that copper loss, induced either by silencing of the copper importer SLC31A1 or by pharmacological chelation, leads to a marked upregulation of SLC7A11. This adaptive response enhances glutathione synthesis, bolsters antioxidant defenses, and protects cells from ferroptosis. Mechanistically, copper deprivation act
