Magnesium at the Neurovascular Interface: A Narrative Review of Atherosclerosis, Peripheral Arterial Disease, and Neuropathic Pain.

Magnesium (Mg) is an essential divalent cation involved in more than 600 enzymatic reactions and plays a fundamental role in vascular, metabolic, and neural homeostasis. Although Mg is frequently discussed as an analgesic supplement, emerging evidence suggests that it acts as a neurovascular-metabolic modulator. Low magnesium status has been associated with endothelial dysfunction, atherosclerotic burden, impaired microcirculatory function, and overlapping ischemic and neuropathic pain phenotypes, although direct causal clinical evidence remains limited. This narrative review integrates mechan

Neuropsychiatric symptoms of chronic stress and pharmacist counseling.

Chronic stress negatively affects both mental and physical health. The most common manifestations are neuropsychiatric disorders, which can be characterized by their symptoms and assessed using the proposed "6N" concept. The growing prevalence of these disorders increases the need to address them in pharmacies as well. An effective and safe self-treatment for the neuropsychiatric symptoms of chronic stress involves treating magnesium deficiency with over-the-counter magnesium-containing medications as an etiopathogenetically oriented approach, given the vicious cycle of stress and magnesium, a

Association between low magnesium status and new-onset dementia in the general population: a propensity score-matched cohort study.

BACKGROUND: Low magnesium status has been linked to cerebrovascular conditions that overlap with pathways implicated in dementia, yet prior studies have relied on single magnesium measurements obtained in earlier population-based cohorts. Whether repeatedly documented low magnesium status is associated with incident dementia in real-world clinical populations is unclear. METHODS: This retrospective propensity score-matched cohort study used data from the TriNetX Global Collaborative Network (2016-2023). Adults aged ≥50 years with two serum magnesium values <1.7 mg/dL within a 1-year window we

Mallotus japonicus leaf extract restores TRPM6 magnesium channel expression and suppresses cellular senescence in colonic epithelial cells.

Age-related alterations in intracellular Mg2+ homeostasis can influence cellular bioenergetics and stress responses, yet the epithelial mechanisms linking aging and cellular senescence to impaired magnesium handling remain incompletely defined. Frailty is an emerging public health challenge in aging societies, and age-related magnesium deficiency has been proposed as one contributing factor. We investigated whether aging and cellular senescence impair epithelial magnesium handling in the colon and whether Mallotus japonicus (M. japonicus) leaf extract ameliorates these alterations in mouse col

Multi-omics analysis reveals galactose metabolism as a key regulatory pathway of stress adaptation to magnesium deficiency in passion fruit.

Magnesium (Mg) deficiency is a major nutritional constraint in acidic orchard soils and can impair both vegetative growth and fruit quality in passion fruits (Passiflora edulis). However, the physiological and molecular mechanisms underlying Mg deficiency responses in passion fruit remain insufficiently understood. This study employed a comprehensive approach, integrating physiological assessments with metabolomic and transcriptomic profiling, to explore the responses to Mg deficiency in the leaves and fruits of purple passion fruit. Our findings revealed that Mg deficiency detrimentally affec

The dietary calcium-to-magnesium ratio and its association with glycemic control in type 2 diabetes: a cross-sectional study.

Inadequate magnesium (Mg) and calcium (Ca) intake has been associated with impaired glucose homeostasis in type 2 diabetes mellitus (T2DM). An imbalanced dietary Ca/Mg ratio may affect the concentrations of these nutrients and contribute to poor disease control. This study investigated the association between the dietary Ca/Mg ratio, serum Ca and Mg levels, and glycemic biomarkers in individuals with T2DM. A cross-sectional observational study was conducted in adults with T2DM. Body weight, height, waist circumference, blood pressure, body fat percentage, and body mass index were assessed. Die

Magnesium: Hype and Health Benefits.

PURPOSE OF REVIEW: Magnesium plays an indispensable role in human health and is intricately involved in many crucial physiological and metabolic pathways. True biochemical magnesium deficiency is rarely encountered in clinical practice but subclinical and functional deficiency of magnesium appears to be more common and is reported in many chronic health disorders. RECENT FINDINGS: Estimating magnesium status is challenging and best evaluated by a combination of serum magnesium, urinary magnesium, and dietary magnesium intake. Many randomized controlled trials (RCTs) have demonstrated the effi

Magnesium depletion score and erectile dysfunction: A cross-sectional and Mendelian randomization study.

Erectile dysfunction (ED) is closely linked to vascular and metabolic disorders. Magnesium plays a crucial role in endothelial function, inflammation, and metabolism, but the relevance of chronic magnesium depletion to ED remains unclear. The magnesium depletion score (MDS) is a practical marker of long-term magnesium loss. This study examined the association between MDS and ED in United States adults and explored genetic evidence for magnesium metabolism using Mendelian randomization (MR). This cross-sectional study included 3692 men aged ≥ 20 years from National Health and Nutrition Examinat

Why Serum Magnesium Fails: A Narrative Review of Magnesium Biochemistry, Compartmental Exchange, and Endpoint Selection for Supplementation Trials.

Magnesium is an obligate cofactor in more than 600 enzymatic reactions, yet population-level inadequacy is widespread and rarely detected. The reason is largely methodological. Serum magnesium, used in routine care and in most supplementation trials, holds less than one percent of total body magnesium and is homeostatically defended by renal reabsorption and bone exchange, so individuals with substantial tissue depletion may present with values inside the reference range. This narrative review addresses a specific problem: the biochemical endpoints of oral magnesium supplementation trials are
