BACKGROUND: The COVID-19 pandemic underscored how hospital-acquired co-infections with antimicrobial resistance (AMR) bacteria and respiratory viruses can drive high mortality in critically ill patients. Despite antimicrobial stewardship and vaccines, effective prophylactic solutions remain lacking, highlighting the need for safe, antigen-independent strategies that provide rapid, broad-spectrum protection.
METHODS: We tested whether a single intraperitoneal pretreatment with n-dodecyl-β-D-maltoside (DDM) could provide broad prophylaxis. Mice were challenged with clinical isolates of carbapen

Patients with sepsis and concurrent methicillin-resistant Staphylococcus aureus (MRSA) bloodstream infection face a substantial risk of mortality. This study aimed to develop and validate a pragmatic, interpretable prediction model for in-hospital mortality in this high-risk population. We conducted a retrospective, single-center cohort study including 1,605 eligible patients, who were randomly divided into a training set (n = 1,124) and a validation set (n = 481). A rigorous, multi-stage feature selection pipeline—integrating univariate analysis, multi-model machine learning importance assess

BACKGROUND: Panton-Valentine leukocidin (PVL)-positive Staphylococcus aureus strains are associated with severe necrotising infections and may be linked to fulminant systemic inflammatory presentations. We report an exceptionally fulminant and rapidly fatal case of PVL-positive S. aureus sepsis with toxic shock-like features and early deterioration despite escalation to veno-arterial extracorporeal membrane oxygenation (VA-ECMO).
CASE PRESENTATION: A previously healthy 16-year-old boy presented with chest pain, dyspnoea and circulatory collapse. Initial evaluation demonstrated massive bilater

Hematology patients are highly susceptible to severe bacterial infections, particularly those caused by multidrug-resistant (MDR) gram-negative pathogens, which are associated with significant morbidity and mortality. Eravacycline, a novel fluorocycline antibiotic, demonstrates broad-spectrum activity against MDR bacteria. This real-world study aimed to evaluate the effectiveness and safety of eravacycline in Chinese hematology patients. In this multicenter, retrospective study, hematology patients receiving ≥3 days of eravacycline between September 2023 and September 2024 were included. The o

Cytokine storm (CS) is a severe hyperinflammatory syndrome characterized by the excessive production of pro-inflammatory cytokines, such as IL-1β, IL-6, and TNF-α. While CS can arise from various triggers, including viral infections, autoimmune disorders, and immunotherapies, in severe bacterial infections and sepsis, it is primarily initiated by bacterial pathogen-associated molecular patterns (PAMPs). This dysregulated immune response increases the risk of endothelial dysfunction, disseminated intravascular coagulation, acute respiratory distress syndrome, multi-organ failure, and mortality.

BACKGROUND: Staphylococcus aureus is a major pathogen responsible for both community-acquired and hospital-acquired infections. However, disseminated infections complicated by pulmonary abscesses and osteomyelitis are rare in otherwise healthy children.
CASE PRESENTATION: We report a case of a previously healthy 12-year-old boy who presented with a 7-day history of cough and a 5-day history of fever and progressive swelling of the left upper limb after self-manipulating a right forearm furuncle. Physical examination and imaging revealed multifocal pulmonary abscesses and osteomyelitis of the

Menstrual toxic shock syndrome (mTSS) is a rare yet life-threatening disease that is caused when the opportunistic pathogenic bacterium Staphylococcus aureus releases the toxic shock syndrome toxin 1 (TSST-1) superantigen, which triggers systemic inflammation. The main risk factor for mTSS is prolonged use of intravaginal products, which facilitates S. aureus growth and TSST-1 production in menstrual blood. However, some studies suggest that the vaginal microbiota may also play a role in mTSS occurrence. A previous study reported that the presence of S. aureus in menstrual fluids was correlate

Neutrophil extracellular traps (NETs) are crucial innate immune components that ensnare and neutralize pathogens. Inspired by this, we engineered a novel peptide, RFC, designed to mimic NETs' "trap-and-kill" strategy against Staphylococcus aureus infections. RFC integrates an antimicrobial peptide (KR12), a self-assembling motif (KLVFF), and a Staphylococcus-targeting sequence (CARGGLKSC). In vitro, RFC exhibited potent broad-spectrum activity (minimum inhibitory concentration (MIC) as low as 4 μM), fast bactericidal kinetics (>3-log10 reduction within 2 h at 1× MIC), inhibited biofilm formati

For pregnant patients with severe coronavirus disease 2019 (COVID-19) requiring veno-venous (V-V) extracorporeal membrane oxygenation (ECMO), the ideal timing of delivery is uncertain, particularly when complicated by cardiogenic shock. A 44-year-old patient experienced severe respiratory failure due to COVID-19 at 23 weeks of gestation and underwent V-V ECMO support. Due to severely depressed biventricular cardiac function caused by Staphylococcus aureus bacteremia, the ECMO mode was converted from V-V to veno-arteriovenous (V-AV) ECMO, resulting in improved maternal cardiac function and feta

Sepsis is a life-threatening state of disseminated infection, and treatment requires knowledge of the organism responsible. The gold standard for sepsis diagnosis is blood culture, which requires days of growth. Next-generation sequencing has been proposed as an alternative; however, existing methods may lack sensitivity. In this work, we explore the idea of genomic quasi-G-primes, which are short DNA sequences specific to a single species within a group of relevant species. We first validated the genomic quasi-G-prime classification in controlled Staphylococcus aureus sequencing experiments,

OBJECTIVE: This review aimed to provide a comprehensive overview of the types of ocular manifestations observed in patients suffering from systemic sepsis including modifications in management of sepsis with ocular involvement. INTRODUCTION: Sepsis is a life-threatening condition resulting in widespread organ dysfunction, driven by dysregulated host responses to infection. In patients with altered sensorium or impaired communication, timely detection of ocular involvement becomes particularly challenging. Delays in recognition can lead to irreversible visual loss, underscoring the need for ear

PURPOSE: To report a case of bilateral macular neovascularization (MNV) secondary to septic embolism from a shunt infection.
METHODS: Observational case report.
RESULTS: A 57-year-old female on maintenance hemodialysis experienced cardiac arrest due to sepsis and was resuscitated with an automated external defibrillator. Blood cultures grew methicillin-sensitive Staphylococcus aureus (MSSA). Echocardiography showed no evidence of infective endocarditis (IE); a pseudoaneurysm of the shunt vessel was identified as the likely source of infection. Chest computed tomography revealed a septic pulm

INTRODUCTION: Osteomyelitis of the pubic symphysis is rare, representing less than one percent of all osteomyelitis cases, and is frequently misdiagnosed as genitourinary pathology owing to overlapping symptoms and anatomical proximity. Tuberculous involvement at this site is exceptional, and concurrent infection with methicillin-resistant Staphylococcus aureus (MRSA) remains infrequently documented. This case contributes to orthopedic literature by demonstrating how reliance on blood and pus cultures alone can delay recognition of occult mycobacterial disease in culture-discordant pelvic oste

Previous Staphylococcus aureus vaccines have been unsuccessful, but it seems that including T cell immunity in the vaccine is essential for its success. This study used Metal ABC transporter substrate-binding protein (MABC), Nickel ABC transporter (NABC), and Phosphatidylinositol phosphodiesterase (PIc) B and T cell epitopes as a vaccine in a mouse bacteremia model. Mice immunized with PIc and epitope mixture groups showed robust humoral immunity and high survival rates in the bacteremia model. The highest protection level of mice was shown in the peptide mixture group, which was correlated wi

BACKGROUND: Although manipulation of host hemostasis is a central mechanism in Staphylococcus aureus virulence, clinically relevant phenotypes of S aureus coagulase activity and the variability thereof in the context of bloodstream infections are unknown.
METHODS: We created a high-throughput staphylothrombin activity and growth fitness assay to characterize fibrin formation and S aureus replication by adding bacterial culture supernatant to human blood plasma. Supernatant is rich in bacterial proteins, including coagulases secreted during growth, but it contains relatively few viable cells.

INTRODUCTION: Bloodstream infections (BSIs) are an important complication among injured patients, yet existing studies have focused on selected populations or specific settings, limiting the generalisability of the findings. In this study, we conducted a population-based study to examine the incidence, demographic and clinical variation of injury-related BSIs.
METHODOLOGY: The study population consisted of all residents of Queensland, Australia, who developed an injury-related BSI identified between 1 January 2000 and 31 December 2019. The linked data used for this study consisted of three st

INTRODUCTION: Antimicrobial Resistance (AMR) is a global health issue with profound implications for human health, economy, and agriculture worldwide. Maternal colonization with antibiotic-resistant bacteria serves as a surrogate marker for the prevalence of such bacteria in the community and also gives a clue to the likely bacteria that cause early onset neonatal sepsis. This study aims to determine the proportion of colonization with selected antibiotic-resistant bacteria, namely Methicillin Resistant Staphylococcus aureus (MRSA) and Extended spectrum beta-lactamase (ESBL) -producing E. coli
