Transient activation of ciliary CCR2 signaling promotes the initiation of adipogenesis.

Despite extensive evidence linking inflammation to obesity, a clear mechanistic model for how inflammatory signals regulate adipose tissue development remains lacking. Here, using in vitro and in vivo models, we demonstrate that transient primary cilium assembly and cilium-dependent inflammatory signaling in preadipocytes serve as key initiators of adipogenesis. Adipose stem cells transiently activate a ciliary gene program to form ciliated preadipocytes, which subsequently commit to adipogenesis and lose their cilia upon further differentiation. We further show that C-C motif chemokine recept

The impact of obesity and overweight on maternal and fetal health with conservative preconception interventions: a structured narrative review.

PURPOSE: Overweight and obesity before pregnancy are associated with adverse maternal, pregnancy-related, and longer-term health outcomes. This review aimed to summarize recent evidence on preconception overweight and obesity and to synthesize findings on conservative preconception or interpregnancy interventions, including lifestyle, dietary, behavioral, weight-management, and supplementation-based strategies. METHODS: A structured narrative review with a predefined database search was conducted. PubMed and Embase were searched for studies published between January 2019 and December 2023 usi

Constitutive UCP1 expression is associated with a glucose-dependent phenotype in murine mesenchymal stem cells.

Mesenchymal stem cells (MSCs) possess significant metabolic plasticity, enabling them to adapt dynamically to environmental stressors during tissue repair. Although UCP1 is associated with nonshivering thermogenesis in brown adipose tissue (BAT) through the uncoupling of oxidative phosphorylation, its impact on MSCs metabolism remains unexplored. Herein, we evaluated the ectopic expression of UCP1 in MSCs and its effects on classical surface markers, the redox state and glucose metabolism. MSCs were genetically modified to constitutively overexpress UCP1 using a lentiviral construct. Transduct

Healthcare costs of childhood overweight and obesity in Chile: evidence from an analysis of linked administrative records.

BACKGROUND: Childhood overweight and obesity are highly prevalent in Chile, affecting over 50% of children under the age of 12, yet evidence on the healthcare costs associated with them remains limited. In this study, we quantify the association between high body mass index (BMI)-for-age and annual health expenditures among school-aged children using linked national administrative data. METHODS: We linked anthropometric measurements from a national school-based surveillance system with health expenditure records from the public and private insurance systems. The sample included children in pr

BMI as a partitioning factor for estradiol and estrone reference intervals in middle-aged men and postmenopausal women.

INTRODUCTION: Estrogens play a large role in female and male physiology and are routinely determined in the clinical setting. For interpretation, distinct reference intervals are applied for men, pre---, and postmenopausal women. Adipose tissue is a potent producer of estrogens and influences the circulating estrogen concentrations, although its impact on estrogen reference intervals is currently unknown. METHOD: We determined estradiol and estrone concentrations in 6,671 middle-aged participants of a large cohort oversampled for persons with BMI >27 kg/m2. We investigated partitioning based

Midlife physical activity trajectories are associated with reduced physical pre-frailty/frailty in older age: insights from the prospective ATTICA sub-study 2002-2024.

BACKGROUND: Although physical activity in older adults has been associated with lower risk of frailty, it is unclear if midlife physical activity patterns are associated with pre-frailty/frailty status in older age. OBJECTIVES: To investigate whether longitudinal midlife physical activity trajectories are associated with physical pre-frailty/frailty development in older age, independent of cardiometabolic risk and other lifestyle factors. DESIGN: Population-based prospective cohort study. SETTINGS: In-person assessments and telephone interviews of persons living in urban communities in Atti

Clinical, Economic, and System-Level Outcomes of Bariatric Surgery in Adults: Protocol for a Systematic Review.

BACKGROUND: Obesity affects approximately 880 million adults worldwide and carries substantial health, economic, and social consequences. Although bariatric surgery achieves sustained weight loss and metabolic improvement, existing systematic reviews largely predate 2020 and rarely integrate clinical, economic, and system-level perspectives within a single synthesis. OBJECTIVE: This study aims to identify, critically appraise, and synthesize evidence published between 2020 and 2025 on the clinical effectiveness and economic impact of bariatric surgery in adults with obesity. A secondary, expl

Bridging the intention-behavior gap in physical activity among adults with obesity: A situation-specific theory.

BACKGROUND: Adherence to physical activity is a major challenge in the management of obesity. While research on physical activity among individuals with obesity has expanded, there is still a lack of a systematic, population-specific theoretical framework that addresses the unique behavioral mechanisms pertinent to this demographic. This study aims to develop a situation-specific theory of physical activity tailored for adults with obesity and applicable to nursing clinical practice. METHODS: An integrative approach was employed, combining theoretical analysis, a scoping review, qualitative i

Substantial preoperative weight loss increases odds of healthcare utilization after reduction mammoplasty in obese patients.

BACKGROUND: Obesity is associated with increased complications following reduction mammoplasty (RM). Preoperative weight loss is commonly recommended to reduce surgical risk, although it remains unclear whether obese patients who have recently lost weight experience outcomes comparable to their weight-stable obese counterparts. This study evaluated the impact of recent preoperative weight loss on postoperative outcomes following RM. METHODS: Epic COSMOS was used to identify obese patients undergoing RM. Patients who lost ≥5% body weight within two years before surgery were compared with weigh

Weight Regain After Incretin Treatment: The Lean Mass Deficit Hypothesis and a Role for Testosterone Combination Therapy in Men.

BACKGROUND: Glucagon-like peptide-1 receptor agonists (GLP-1RAs), dual glucose-dependent insulinotropic polypeptide (GIP)/GLP-1RA and triple (GIP)/GLP-1RA/glucagon agonists achieve placebo-subtracted weight loss of 15% to 22%. Their long-term value depends on how well the weight loss is maintained and on the composition of weight loss, not only amount of weight loss. Part of incretin-induced weight loss is lean tissue. It is unknown whether the proportion of lean mass, and especially, muscle mass loss is greater than with other weight-loss interventions, and whether it may cause clinically mea

Real-World Semaglutide Initiation Behaviours in the United States: Impact on Early Treatment Discontinuation and on 1-Year Persistence.

AIMS: Little is known about how glucagon-like peptide-1 receptor agonists (GLP-1RA) are initiated in real-world settings and whether prescribing behaviours in routine care contribute to the high discontinuation rates seen in practice. We examined prescribing behaviours for injectable semaglutide at treatment initiation and evaluated their impact on early medication discontinuation and on 1-year persistence among US adults. MATERIALS AND METHODS: Using MarketScan data (2018-2021), this retrospective cohort study identified US adults with diabetes and/or obesity on no prior incretin therapy sta

Current status of anti-obesity medications and performance, an EHR based survey.

BACKGROUND: While randomized clinical trials (RCTs) establish the efficacy of anti-obesity medications (AOMs), their real-world effectiveness, inter-individual variability, and weight regain post discontinuation remain poorly characterized across diverse, large scale populations. METHODS: Electronic health records with pharmacy dispensation of obesity or overweight individuals were retrieved from local UT-Physician and nationwide EPIC COSMOS databases. AOMs were analyzed for short to long-term effectiveness on obesity relevant metrics. RESULTS: From the two cohorts, 71,318 (11·7%) and 1 M (2
