[Sarcopenia, obesity, and limitations in daily activities in older adults from Elsi-Brazil: recommendations from the Sarcopenia Definition and Outcomes Consortium].

This study aimed to investigate the associations between isolated sarcopenia and its combination with general and abdominal obesity, stratified by sex, with limitations in basic (BADL) and instrumental (IADL) activities of daily living among Brazilian older adults. A cross-sectional study was conducted with 6,874 older adults (≥ 60 years) from the second wave of the Brazilian Longitudinal Study of Aging (2019-2021). Sarcopenia was defined according to the criteria of the Sarcopenia Definition and Outcomes Consortium, which involves the concomitant presence of low gait speed and low handgrip st

Effect of postnatal prebiotic fiber intake on obesity risk and insulin resistance in rat offspring exposed to a maternal obesogenic diet and low-dose aspartame.

OBJECTIVES: The objective of this study was to examine whether offspring postnatal prebiotic fiber (Pre) intake could mitigate metabolic risks associated with a maternal obesogenic diet plus aspartame (APM) consumed during pregnancy and lactation. METHODS: Following 11 weeks of obesity induction, female Sprague-Dawley rats (n = 42) were bred and then randomized to a high fat/sucrose (HFS) diet with water (control, CTR) or HFS with APM (7mg/kg/day in drinking water, APM) throughout pregnancy and lactation. From 3 to 12 weeks of age, offspring consumed a HFS or HFS+Pre (10% wt/wt) diet, resulti

Sexual dimorphism in high-fat diet-induced depression-like phenotypes in mice: astrocyte-centered molecular, circuit, and microbiota associations.

Metabolic disorders (MetD) are linked to depression, both of which exhibit sex differences in prevalence and clinical presentation; however, the mechanisms underlying sex-specific vulnerability to MetD-related depression remain unclear. Male and female C57BL/6 N mice were fed a high-fat diet (HFD) for 12 weeks (8 to 20 weeks of age) to induce MetD. Depression-like behaviors were assessed using the sucrose preference and forced swim tests. Ventral hippocampus (vHPC) astrocyte-centered molecular, structural, and circuit alterations were evaluated using Fluoro-Gold retrograde labeling, immunohist

Central epigenetic mechanisms of overeating and thermal regulation.

This chapter discusses epigenetic mechanisms underlying the central control of homeostasis, focusing on energy intake (and expenditure) and on body temperature. Research, mostly performed in rodents, shows a wide range of epigenetic mechanisms involved in regulation of feeding and body weight, which, when challenged, result in overeating and obesity in vulnerable individuals. Many studies found prominent sex differences and intergenerational transfer of overweight and obesity, with concomitant transfer of epigenetic marks. The anterior hypothalamic preoptic area has a pivotal role in homeostas

Assessing Behavioral and Environmental Facets of Obesity Unobtrusively Utilizing Digital Tools: Introducing the BETTER4U Indicators.

Understanding obesity involves evaluating numerous contributing factors. Collecting the necessary data for this can impose a significant burden on both research participants during clinical studies and real-life deployments of digital tools aiding obesity management. The BETTER4U project (European Union's Horizon Europe, Grant Agreement ID 101080117) introduces 36 behavioral and 9 living environment indicators designed to unobtrusively and efficiently capture daily life through passive monitoring using market-available smartphones and smartwatches while accounting for environmental confounders

Incremental value of DXA-derived visceral versus total adiposity beyond routine anthropometric assessment for hypertension discrimination: a population-based study.

Hypertension is strongly linked to obesity, yet it remains uncertain whether DXA-derived measures of adiposity improve hypertension discrimination beyond routine anthropometric assessment and whether any incremental value reflects visceral rather than total adiposity. This study examined whether DXA-derived visceral and total adiposity provide incremental discriminatory value beyond routine anthropometric assessment for hypertension discrimination. We analyzed 7817 adults aged 20-59 years from the 2013 to 2018 National Health and Nutrition Examination Survey with valid DXA measurements. Discri

A Multi-Component High-Fibre Meal Replacement Containing Pyrus bretschneideri Rehd. Pomace Alleviates Diet-Induced Obesity by Modulating the Gut Microbiota, Bile Acid Metabolism and Hepatic AMPK Signalling.

This study employed an integrated multi-omics approach to investigate the regulatory effects and molecular mechanisms of a multi-component, high-dietary-fibre meal replacement powder containing Pyrus bretschneideri Rehd. pomace (PDMR) in high-fat-diet-induced obese mice. The results showed that PDMR significantly reduced body weight gain, Lee's index, serum lipid and inflammatory cytokine levels, and improved adipocyte morphology. Mechanistically, PDMR activated the AMPK signalling pathway, suppressed lipogenesis via the ACC-SREBP1c-FAS axis, and promoted fatty acid β-oxidation and thermogenes

Gjb2 V37M Modifies Metabolic Responses to Chronic High-Fat Feeding in Mice.

GJB2 encodes connexin 26 (Cx26), and pathogenic GJB2 variants are among the most common genetic causes of hereditary hearing loss; because Cx26 is expressed in metabolically relevant tissues including liver, pancreas, and adipose tissue, whether the hearing-loss-associated c.109G>A (p.V37M) variant also modifies systemic metabolic responses to high-fat diet (HFD) exposure remains unclear. Three-month-old male wild-type (WT) and homozygous Gjb2 V37M mice were assigned to low-fat diet (LFD) or high-fat diet (HFD) groups, generating four study groups: WT-LFD, WT-HFD, V37M-LFD, and V37M-HFD. Do Nutritional Status and Dietary Intake Influence the Ghrelin Response?

BACKGROUND/OBJECTIVES: Ghrelin is a key orexigenic hormone involved in the regulation of appetite and energy homeostasis, and its fasting circulating concentrations may reflect long-term energy balance and nutritional status. The aim of the study was to assess the associations between habitual daily energy and macronutrient intake from a conventional diet and the fasting total ghrelin concentrations in individuals with different nutritional status and to evaluate the postprandial ghrelin response following consumption of a standardized meal. An additional aim was to identify body composition p
