Effects of Vitamin B12, Folic Acid, and L-Carnitine on Congenital Malformations Induced by Vitamin A Hypervitaminosis.

BACKGROUND: Excessive intake of exogenous vitamin A during pregnancy has been associated with congenital malformations. This experimental study aimed to evaluate whether vitamin B12, folic acid, and L-carnitine exert a protective or contributory effect on malformations induced by hypervitaminosis A. METHODS: Thirty-five healthy adult female Wistar albino rats and seven male rats were used. Pregnant rats received vitamin A alone or in combination with vitamin B12, folic acid, or L-carnitine for 21 consecutive days (GD 0-21). At the end of gestation, neonates were evaluated for gross congenital

Effect of vitamin A supplementation and the role of infections and inflammation on vitamin A status in children aged 36-59 months in rural Burkina Faso: results of a community-based pre-post intervention study.

BACKGROUND: Several interventions are implemented in Burkina Faso to tackle vitamin A (VA) deficiency. This study assessed the effects of high-dose VA supplementation (VAS) on indicators of VA status in preschool children and examined whether infections and inflammation modified VA status post-supplementation. METHODS: A community-based pre-post intervention study was conducted during the dry season in rural Burkina Faso among 60 children aged 36–59 months. After baseline assessment, each child received a high-dose oral VAS, and an endline assessment was performed one month later. VA total bod

Gut microbiota and metabolic disruption induced by food chain-transferred nanoplastics: A mediating role of CYP26A1 in retinoic acid toxicity.

Polystyrene nanoplastics (PS-NPs) pollution has emerged as critical environmental and public health concerns due to their potential for bioaccumulation and systemic toxicity. While our previous work demonstrated the trophic transfer of micro- and nanoplastics from insect yellow mealworms (Tenebrio molitor) to mammals, the biological consequences remain poorly understood. This study aimed to investigate the impact of food chain-transferred PS-NPs (FCT-NPs) on the gut-liver axis in a mouse model. We demonstrate that FCT-NPs induce gut microbiota dysbiosis, disrupt metabolic homeostasis, and alte

Enhanced Calcium Intake Increases Bone Mineral Density on the Background of Vitamin A Toxicity in a Swine Model.

BACKGROUND: Hypervitaminosis A inhibits bone mineralization. Additional calcium (Ca) may mitigate the negative impacts of excessive dietary vitamin A (VA). OBJECTIVES: This study determined the effects of high dietary Ca on bone formation and growth in swine receiving varying concentrations of dietary VA, including the National Research Council (NRC) recommendation (2200 IU VA/kg feed for 5-11 kg pigs). METHODS: Pigs were weaned on day 17 and randomly assigned by sex, weight, and litter to 5 treatments (n = 12/group): -VA=Ca, low VA, control Ca; =VA=Ca, NRC control; +VA-Ca, high VA, low Ca;

Intracranial hypertension: the hidden risk of supplements containing vitamin A.

Hypervitaminosis A is relatively uncommon but may develop with excessive supplementation or medication use. We report a 17-year-old non-obese male presenting with symptomatic papilloedema and normal neuroimaging. Investigations excluded alternative causes, but a detailed history identified that he used a protein supplement containing significant amounts of vitamin A. Although there are potential confounding factors, the combination of the clinical presentation, deranged liver function tests, resolution following the diagnostic lumbar puncture, starting acetazolamide and stopping the supplement

Clinical Spectrum and Management of Hypervitaminosis A: A Systematic Review of Case-Based Evidence.

BACKGROUND: Hypervitaminosis A, caused by excessive intake or pathological accumulation of vitamin A, can lead to severe and diverse adverse effects across multiple organ systems. With rising availability of over-the-counter supplements and expanding food fortification programmes, the evidence base remains fragmented and primarily reliant on sporadic case reports. This systematic review synthesizes available evidence on the adverse effects and management of hypervitaminosis A. METHODS: This systematic review was conducted according to the PRISMA 2020 guidelines. The literature was searched us

Supraphysiological Doses of Vitamin A Promote Changes in Hippocampal Oscillation Bands and Cardiac Activity in Wistar Rats.

Vitamin A is an essential micronutrient involved in growth, immune regulation, and central nervous system function; however, excessive intake may disrupt systemic homeostasis. This study investigated the electrophysiological effects of experimental hypervitaminosis A on hippocampal activity and cardiac function in male Wistar rats. Animals received a supraphysiological dose of vitamin A (50,000 IU/kg, intraperitoneally) once daily for 3, 7, or 14 days. Serum calcium levels were quantified, and electrophysiological recordings were obtained from the CA1 hippocampal region (EEG) and through elect

Vitamin A concentration in bovine liver and milk does not only depend on characteristics of the farming system.

The study examined the vitamin A status in commercially managed suckler cows and lactating dairy cows and identified primary influencing factors. Liver retinyl ester concentrations were higher in multiparous than primiparous cows (p < 0.01). Pasture availability was associated with higher β-carotene concentrations (p < 0.001). In dairy cows, pasture access during the dry period did not affect any of the parameters assayed. β-Carotene and retinol in milk increased with parity. No vitamin A deficiency or hypervitaminosis A was detected. Liver and milk retinol and retinyl ester concentrations tha

Association between vitamin A status, inflammations, and infections in children 36-59 months of age in rural Burkina Faso: A cross - sectional study.

BACKGROUND: Assessing vitamin A (VA) status using retinol and retinol-binding protein (RBP) in the presence of infection and inflammation remains challenging, as both markers prove to be unreliable during such physiological disturbances. OBJECTIVE: This study aimed to assess the association between common infections and inflammation and VA status of children in rural Burkina Faso. METHODS: Two community-based cross-sectional studies were conducted in the villages of Sourkoudougou and Banakeledaga, in Southwestern Burkina Faso, one during the dry season (November 2016- January 2017) and the s

Anencephaly and palatoschisis in 2 newborn puppies.

Anencephaly is a congenital condition characterized by incomplete brain development and malformations of cranial bones and cerebral hemispheres. Such defects arise from improper neural tube closure during embryogenesis. Although well documented in humans, they are infrequently reported in veterinary medicine. The etiology of anencephaly in dogs is not fully understood, but a strong genetic predisposition has been observed, particularly among brachycephalic breeds. In addition, studies in humans have implicated factors such as folic acid deficiency, hypervitaminosis A, and exposure to teratogen

Metabolomics profiling of hypervitaminosis A in South African preschoolers is characterised by modified serum lysophospholipids and oxylipins.

Evidence indicates hypervitaminosis A may be attributed to overconsumption of natural preformed vitamin A (VA) and overlapping VA intervention strategies. Hypervitaminosis A can disrupt metabolic processes; however, the extent and mechanisms of these impacts are not well understood. This study aims to assess metabolic differences related to hypervitaminosis A and VA supplementation by performing metabolomics analysis. A subsample of South African preschoolers participating in the country's VA supplementation programme was selected. Participants were divided into two groups: adequate VA (n 15;

Vitamin A toxicity and hepatic pathology: A comprehensive review.

Vitamin A is essential for vision, immunity, and cellular function, but excessive intake, known as hypervitaminosis A, leads to liver toxicity. Toxicity can be acute (from high single doses) or chronic (from prolonged overconsumption), causing symptoms like nausea, bone pain, and liver damage. The normal values of vitamin A in adults, measured as serum retinol, can range from 0.3 mg/L to 1.2 mg/L. The liver, which stores vitamin A in hepatic stellate cells, becomes overwhelmed, leading to retinoid accumulation, oxidative stress, and inflammation. Pathologically, vitamin A toxicity progresses f

Hypervitaminosis A in type 2 diabetes and its relation with renal function and cardiovascular complications.

Vitamin A is a fundamental micronutrient in the diet. In recent years, hypervitaminosis A has been described in subjects, mainly children, with renal dysfunction. The aim of the present study was to estimate the prevalence of hypervitaminosis A in adult patients affected by type 2 diabetes and to evaluate the associated factors. This cross-sectional observational study included 200 ambulatory patients affected by type 2 diabetes regularly attending the Diabetes Clinic of Verona (Italy). Eligible patients were consecutively enrolled. Vitamin A was measured by reverse phase high performance liqu

The effects of preformed vitamin A and provitamin A carotenoid supplementation on tadpoles of the poison frog Phyllobates vittatus.

Understanding the nutritional requirements of captive animals is necessary for proper animal husbandry, however, the specific dietary requirements for many amphibian species commonly kept in captivity are unknown. Like most vertebrates, frogs cannot synthesize carotenoids and must therefore obtain these essential nutrients through diet. It is unclear if amphibians can cleave provitamin A carotenoids to form vitamin A metabolically within the body, so common practice is to supplement their captive diets with both preformed vitamin A and provitamin A carotenoids. We carried out a feeding experim

A Rare Case Report of Benign Intracranial Hypertension Caused by Hypervitaminosis A.

Benign intracranial hypertension (BIH) in children is recognized as elevated intracranial pressure without hydrocephalus or intracranial mass. It manifests differently in adults, with no apparent predilection for sex or weight. Headache, papilledema, and possibly sixth nerve palsy with visual field defects are the typical symptoms of this syndrome. Vitamin A toxicity is a rare cause of BIH. We report the case of a previously healthy 13-year-old girl presenting with photophobia, a frontal headache, and vomiting. She had bilateral papilledema discovered by fundoscopy. Both magnetic resonance ima

Mechanistic perspective on the actions of vitamin a in autism spectrum disorder etiology.

Vitamin A (VA) has many functions in the body, some of which are key for the development and functioning of the nervous system, while some others might indirectly influence neural function. Both hypovitaminosis and hypervitaminosis A can lead to clinical manifestations of concern for individuals and for general global health. Scientific evidence on the link between VA and autism spectrum disorder (ASD) is growing, with some clinical studies and accumulating results obtained from basic research using cellular and animal models. Remarkably, it has been shown that VA deficiency can exacerbate aut
