Genetic evidence for a potential role of cytoskeleton-associated protein 2 in lung squamous cell carcinoma: a bidirectional mendelian randomization study.

BACKGROUND: Cytoskeleton-associated protein 2 (CKAP2) plays a role in tumor proliferation, but its causal relationship with lung cancer remains unclear. This study employed a bidirectional two-sample Mendelian randomization (MR) analysis to investigate this association. METHODS: Using genome-wide association and FinnGen data, we assessed the potential causal relationship between genetically predicted plasma CKAP2 levels and lung cancer risk, including its subtypes. The inverse-variance weighted (IVW) method served as the primary analysis, supplemented by MR-Egger and weighted median. Sensitiv

The zebrafish model tackles anti-P2Y12 variability in humans: a translational approach.

Antiplatelet drugs are a pillar in the treatment strategy to prevent ischemic events in patients with cardiovascular disease. However, the action of existing therapies is nonuniform, and the causative mechanism for this variability remains poorly understood. The differences in the microRNA (miRNA) profiles among individuals have been suggested to impact platelet reactivity and treatment outcomes. In several previous clinical reports, miR-150 has been associated with platelet function variability and cardiovascular events. Therefore, we initiated our analysis by examining the mechanistic role o

The prognostic potential of an immunology score in patients treated with curative surgery for non-small cell lung cancer.

PURPOSE: Curative surgery is the best treatment for patients with early-stage non-small cell lung cancer (NSCLC), but around 40% experience recurrence impacting survival. Does the individual immunological status affect the prognosis? This study investigated four immunology markers in relation to recurrence and survival after curative surgery for NSCLC. METHODS: Natural Killer cell activity (NKA), C-reactive protein (CRP), Interleukin 6 (IL6) and Transforming growth factor beta (TGFβ) were analysed before surgery and at three months after surgery for NSCLC. RESULTS: Of the 103 enrolled patien

Astrocyte lipid metabolism: From physiological homeostasis to reactive phenotype plasticity in neurodegeneration.

Astrocytes are one of the most abundant types of glial cells in the central nervous system (CNS) and play pivotal roles in metabolic support, synaptic regulation, and neurotransmitter homeostasis. Astrocytic dysfunction is closely associated with the pathogenesis of neurodegenerative disorders such as Alzheimer's disease (AD), and lipid metabolism constitutes a central axis for maintaining astrocyte function and CNS homeostasis. Here, we provide a systematic review of recent advances in astrocyte lipid metabolism, summarizing the fundamental roles of astrocytes in the CNS and focusing on the b

Inulin- and Butyrate-Mediated HIF-1α Butyrylation Prevent Diabetic Ferroptosis and Cardiomyopathy by Maintaining Iron Homeostasis.

UNLABELLED: Abnormal iron metabolism and ferroptosis contribute to diabetic cardiomyopathy (DCM); however, the regulatory mechanisms remain unclear. Inulin improves systemic metabolism and cardiac function, but whether it alleviates myocardial ferroptosis and DCM is unknown. Using two distinct type 2 diabetes (T2D) mouse models-the high-fat diet combined with streptozotocin-induced T2D model and the db/db mouse model-in combination with an iron chelator, we found that imbalanced iron homeostasis and ferroptosis drive DCM. Inulin restored cardiac iron homeostasis, inhibited ferroptosis and DCM,

What Proportion of Antibody-Negative Type 1 Diabetes Cases Have a Nonautoimmune Etiology? A Genetics-Based Estimate.

UNLABELLED: Among patients with young-onset type 1 diabetes, 10-15% are autoantibody (aAb) negative. How many of these are autoimmune versus nonautoimmune is unknown. To estimate this proportion, we used the only high-confidence evidence of autoimmunity in these cases, the presence of an aAb(+) sibling. In the 2,500 affected sibling pairs of the Type 1 Diabetes Genetics Consortium cohort, we compared the Type 1 Diabetes Genetic Risk Score (GRS2) between 379 aAb(-) patients with an aAb(+) sibling (autoimmune cause) and 253 aAb(-) patients with no aAb(+) siblings (unknown cause), under the assum

Mass Spectrometry-Based Structural Proteomics and Analytical Ultracentrifugation Characterize the Structures of Oligomeric Apolipoprotein E4 and Its Variants.

Apolipoprotein E4 (ApoE4), one of the three common isoforms of ApoE, differs from ApoE3 and ApoE2 by just one amino acid substitution each. Yet these changes in ApoE4 profoundly impact protein structure and function to make it the strongest genetic risk factor for late-onset Alzheimer's disease (AD). ApoE4 exhibits a tendency to form oligomers (e.g., tetramers) that complicate high-resolution structural characterization in aqueous environments. Very little is known about the self-associated oligomeric structures. In this study, we employed ultracentrifugation, native mass spectrometry (MS), an

Suppression of pain transmission and behavior by inhibition of peripheral diacylglycerol metabolism.

Diacylglycerol lipase (DAGL) produces 2-arachidonoylglycerol (2-AG) and other proinflammatory lipids. Inactivation of DAGLs reduces the production of 2-AG, arachidonic acid (AA) and eicosanoids and elicits antinociceptive and anti-(neuro)inflammatory effects in rodents. However, inhibitors that enter the brain can cause significant central nervous system (CNS) side effects. Using activity-based protein profiling (ABPP), we report the discovery of A1480LS, a potent, in vivo active, small molecule dual inhibitor of DAGLα/β that is functionally biased to the periphery. We demonstrate that A1480LS

Exploring protein glycosylation across GlyGen, UniProtKB, and related knowledgebases for data integration.

Protein glycosylation plays a crucial role in cellular function, protein stability, and disease mechanisms, making the accurate annotation and integration of glycobiology data essential for biological research. However, the inherent complexity of glycosylation and the heterogeneity of glycans present significant challenges for data integration, annotation, and accessibility across biological databases. In this paper, we explore the protein glycosylation information present in GlyGen, a glycoscience knowledgebase, Universal Protein Resource Knowledgebase (UniProtKB), a proteomics knowledgebase,

Association between Yo-yo intermittent recovery test, physiological markers of endurance performance, and buffering capacity in professional soccer players.

The Yo-Yo Intermittent Recovery Test Level 1 (YYIR1) is widely recognized as a reliable tool for assessing intermittent endurance and aerobic capacity in team sports athletes. This study aims to verify the association between YYIR1 and physiological markers of endurance performance in professional soccer players, as well as the relationship between metabolic buffering capacity, metabolic acidosis tolerance, and YYIR1 total distance. Twenty-six professional soccer athletes (26.3±4.98 years, 76.4±8.96 kg, 178±6.52 cm, 54.1±4.85 mL·kg-1·min-1 V̇O2max) performed the YYIR1 and underwent a cardiopul

Effects of feeding a Bacillus-based probiotic to grazing Nellore bulls finished on tropical pasture.

Interest in probiotics as an alternative to ionophores has increased, particularly in pasture-based systems using energy supplementation. While these additives aim to improve feed efficiency and mitigate acidosis risk, probiotics may also modulate rumen function by enhancing fiber digestion. This study evaluated the effects of including a Bacillus-based probiotic in an energy supplement on intake, performance, total tract digestibility, and rumen and cecum morphometrics of finishing Nellore cattle. Twenty-eight bulls (502.6 ± 7.0 kg) were blocked by initial body weight and assigned to four pad

Randomized Clinical Trial of Ringer's Lactate Versus Chloride-Reduced Saline-Dextrose in Postoperative Kidney Transplantation.

INTRODUCTION: Normal saline administration induces hyperchloremic metabolic acidosis, potentially impairing kidney allograft recovery. The comparative efficacy of balanced crystalloids against actively chloride-reduced saline protocols remains unclear. METHODS: This randomized, open-label trial compared Ringer's Lactate Solution (RLS) with a dynamic dual-infusion protocol of normal saline and 5% dextrose (NSS + D5W) in adult kidney transplant recipients. Using a prespecified modified intention-to-treat analysis, 134 patients who received ≥2 L of study fluid were evaluated. Fluids were strictl

Occult hypoxemia in the ICU: Clinical risk factors, mortality and implications for nursing practice.

BACKGROUND: Pulse oximetry (SpO₂) is the primary tool used by Intensive Care Unit (ICU) nurses to monitor oxygenation. Occult hypoxemia - a reassuring SpO₂ despite low arterial saturation (SaO₂) - is a critical patient-safety concern. OBJECTIVES: To measure SpO₂-SaO₂ discordance in ICU patients, identify situations in which SpO₂ cannot be trusted, and assess its impact on ICU mortality. METHODS: Retrospective single-centre cohort study (September 2019 - January 2026), reported according to STROBE. SpO₂ was charted hourly; each arterial blood gas was paired with the most recent preceding entr

Advancing osteosarcoma treatment: the impact of intratumoral metabolic reprogramming on immunotherapy.

Osteosarcoma (OS) remains difficult to cure once it becomes metastatic, recurrent, or resistant to chemotherapy, and immune checkpoint blockade has provided limited clinical benefit. The metabolic-immune tumor microenvironment (TME) is one barrier to effective immunity because malignant, stromal, and immune cells compete for nutrients and adapt to hypoxia, acidosis, and matrix-related constraints. This review examines how cancer-associated fibroblasts, extracellular matrix, myeloid and lymphoid populations interact with glucose/lactate, glutamine, arginine, and lipid metabolism in OS. Public O

Incidence of Pediatric Hyperglycemic Hyperosmolar State: A Prospective National Surveillance Study.

INTRODUCTION: Hyperglycemic hyperosmolar state (HHS) is a severe diabetic emergency, typically seen in adults with type 2 diabetes, but with high morbidity described in the limited pediatric cases available in the literature. Existing estimates of pediatric HHS incidence are primarily derived from administrative datasets and may be affected by diagnostic misclassification. We sought to determine the minimum incidence, clinical characteristics, management, and outcomes of pediatric HHS. METHODS: Approximately 2,700 pediatricians and pediatric subspecialists were surveyed monthly from June 2023

A cow individual milk fat percentage prediction method based on low-rank multimodal fusion of time-series, numerical, and textual data.

Milk fat percentage (MFP) precise prediction is paramount for precision dairy farming and early warning of health issues. However, existing prediction methods primarily rely on single lactation time-series data or statistical models, which often fail to effectively fuse multi-source heterogeneous information and frequently overlook the impact of individual variability. To address current limitations, long short-term memory(LSTM), Stacking ensemble learning, bidirectional encoder representations from transformers(BERT) and low-rank multimodal fusion(LMF) were employed to explore a hierarchical m
