A Reproducible Electroporation Strategy for CRISPR-Cas9 RNP and mRNA Delivery in Fish Embryos.

This study presents a streamlined electroporation-based method for efficient macromolecular delivery into zebrafish embryos. We first characterized the physical barrier posed by the chorion using TEM (Transmission Electron Microscope) and established that its removal is prerequisite for effective delivery. A systematic optimization of electroporation parameters for dechorionated embryos identified optimal conditions (e.g., poring pulse: 25 V, 20 ms; transfer pulse: 5 V, 50 ms) that ensured high embryo survival and robust eGFP mRNA transfection. Applying this protocol, we achieved targeted gene

Patterns of Preimplantation Genetic Testing Utilization During In Vitro Fertilization in a Nigerian Fertility Centre: Sociodemographic Characteristics and Clinical Indications.

BACKGROUND: Preimplantation genetic testing (PGT) is increasingly integrated into in-vitro fertilization (IVF) to detect chromosomal abnormalities and monogenic disorders before embryo transfer. Although its use is primarily intended to prevent genetic disease and improve reproductive outcomes, PGT is also applied for non-medical indications such as elective sex selection in some settings. The objective of this study is to characterize the sociodemographic profile of couples undergoing PGT, the clinical indications for testing, and patterns of maternal and paternal age, parity, and geographic

Mild form of aromatic L-amino acid decarboxylase deficiency.

Aromatic L-amino acid decarboxylase (AADC) deficiency (Online Mendelian Inheritance in Man #608643) is a rare autosomal recessive neurometabolic disorder caused by pathogenic variants in the DDC gene, leading to impaired enzyme activity. Affected individuals typically develop symptoms in early infancy, including truncal hypotonia, global developmental delay and oculogyric crises, and the condition is generally associated with a severe clinical course and poor prognosis. We report a patient with a mild and atypical phenotype of AADC deficiency complicated by sensorineural hearing loss and oculoc

Pigmentation anomalies in marine fishes: novel records from Spanish coastal waters and a global synthesis of wild flatfish ambicolouration.

This study documents unusual pigmentation patterns in marine fishes from Spanish coastal waters and provides a global synthesis of published records of ambicolouration in wild flatfishes. A mixed pigmentation anomaly was recorded in Diplodus sargus (Sparidae), severe hypopigmentation compatible with oculocutaneous albinism was documented in Halobatrachus didactylus (Batrachoididae), and near-complete ambicolouration was observed in Solea senegalensis (Soleidae). Three additional unusual pigmentation patterns involving Labrus bergylta (Labridae) and D. sargus were documented photographically du

Homozygous 11q14.3 deletion causing oculocutaneous albinism and multisystem disorder.

Oculocutaneous albinism (OCA) is characterised by hypopigmentation of the skin, hair and eyes. Developmental delay is not a commonly reported feature in OCA. A female toddler was diagnosed with OCA, global developmental delay, hypotonia and congenital heart disease.Given the coexisting neurodevelopmental and cardiac abnormalities, chromosomal microarray (CMA) analysis was performed, which revealed a homozygous deletion at chromosome 11q14.3 involving the TYR, GRM5 and NOX4 genes. Parental segregation analysis using CMA demonstrated heterozygous deletions in both parents, with the proband's hom

Early Genotype-Driven Diagnosis of Hermansky-Pudlak Syndrome Type 4 in a Child With Oculocutaneous Albinism: An Ophthalmic Case Report.

Hermansky-Pudlak syndrome (HPS) is a rare inherited multisystem condition characterized by oculocutaneous albinism, bleeding diathesis, and subtype-specific systemic complications, including pulmonary fibrosis and granulomatous colitis. Early diagnosis is essential because pulmonary involvement, particularly in HPS type 4, may be life-threatening. We report a boy who was first referred for ophthalmologic evaluation at seven weeks of age due to generalized hypopigmentation and congenital nystagmus. Examination revealed marked iris translucency, diffuse fundus hypopigmentation with prominent cho

CsZAT12 interacts with CsNAC87 to repress chlorophyll accumulation in albino leaves of tea plants.

The albino leaves of cold-sensitive tea plants exhibited a marked reduction in chlorophyll content. However, the underlying regulatory mechanism remains elusive. In this study, we determined lower chlorophyll levels in tender leaves of cold-sensitive 'Baiye 1' compared to green leaves during the spring. Comparative transcriptome analyses identified the chlorophyll biosynthesis gene CsPORA, which was significantly downregulated in albino leaves. Functional assays confirmed that CsPORA positively regulates chlorophyll content. Electrophoretic mobility shift assay, yeast one-hybrid, dual-lucifera

[Differential diagnosis of infantile nystagmus].

Infantile nystagmus syndrome is classified into two main categories: idiopathic infantile nystagmus, in which no obvious structural or functional abnormalities of the eyes are present and infantile nystagmus associated with ocular pathologies (sensory defect nystagmus). In the latter, a defect located in the visual pathway but anterior to the lateral geniculate nucleus is responsible for the nystagmus, e.g., pathologies of the anterior segment, retinal diseases or optic neuropathies. Furthermore, oculocutaneous or ocular albinism is a common cause of infantile nystagmus syndrome. Acquired nyst

Beyond Appearances: The Hidden Coat Diversity of the Sicilian Mastiff Revealed by Integrated Phenotypic and Genetic Analyses.

Coat colour is one of the most recognizable and traditionally selected traits in dog breeds. However, despite its apparent simplicity, it represents a biologically complex phenotype, shaped by the interaction of multiple genetic loci, modifier genes, and variable phenotypic expression, challenging standardized phenotypic classification. This study presents the first characterization of coat colour and texture in the Sicilian Mastiff, integrating pedigree records, photographic assessment, and genetic analyses. Breeder-declared pedigree coat colour was examined for 760 dogs to evaluate terminolo

Discovery of urinary metabolite biomarkers of psychiatric disorders using two-sample Mendelian randomization.

BACKGROUND: Mental health disorders cause substantial patient suffering, which could be alleviated through early diagnostic biomarkers. While biomarker discovery is costly, genetic methods utilizing data from large-scale studies, such as Mendelian randomization, may provide a cost-effective approach. METHODS: A two-sample Mendelian randomization analysis was conducted to identify potential urinary biomarkers of seven psychiatric disorders using summary statistics from GWAS data. RESULTS: The analysis revealed 67 analyte-disorder associations, of which 21 were exclusive to a single disorder.

The ability of branched-chain amino acid-to-tyrosine ratio (BTR) to assess preoperative liver function of patients with hepatocellular carcinoma.

Assessment of hepatic functional reserve is critically important for preventing serious complications after hepatectomy such as liver failure. While the indocyanine green clearance test (ICG) is a liver-specific test that is not affected by other organs and is commonly used to evaluate liver reserve capacity. We lack knowledge regarding what test should be performed for patients with jaundice, portal shunts, or intolerance whose liver function cannot be accurately assessed by ICG. To close this gap, we focused on changes in amino acid metabolism associated with impaired liver function. The bra

Low Fischer's ratio is associated with increased mortality in patients with kidney failure.

Metabolic changes in kidney failure, including an altered ratio of plasma branched-chain amino acids (BCAAs) to aromatic amino acids (AAAs), also known as Fischer’s ratio (FR), could potentially affect survival. We analyzed associations of FR with mortality in patients with kidney failure. In 328 kidney failure patients starting dialysis (median age 54 years, 60% men), plasma concentrations of BCAAs (valine, isoleucine, and leucine) and AAAs (phenylalanine, tyrosine) were measured using HPLC. We used Fine-Gray competing-risk regression analysis with renal transplantation as competing risk to c

Gut microbiota-tryptophan-serotonin axis drives anxiety-like behavior via NLRP3-mediated neuronal pyroptosis in the medial prefrontal cortex.

The gut-brain axis plays a critical role in anxiety disorders, yet the underlying mechanisms remain incompletely understood. Using a mouse model of radiofrequency radiation (RFR)-induced anxiety-like behaviors, we employed gut microbiota intervention, regulation of tryptophan metabolites, and other methods to investigate the impact of the gut-brain axis on brain function changes. It was found that gut microbiota dysbiosis disrupts tryptophan metabolism, leading to reduced serotonin (5-HT) levels and NLRP3 inflammasome-mediated neuronal pyroptosis in the medial prefrontal cortex (mPFC). Probiot

Structural determinants of ligand response specificity in the mast cell activating GPCR, MRGPRX2.

The mast cell-specific G-protein-coupled receptor (GPCR) MRGPRX2 (Mas-Related G Protein-coupled Receptor X2) has roles in itch and pain, and it mediates clinically relevant allergy-like responses to a diverse assortment of drugs. The varied responses of individuals to MRGPRX2 agonists, leading to drug hypersensitivity reactions in some cases, suggests the presence of consequential variants in the population. However, genetic associations with drug responses are poorly understood. We used heterologously-expressed MRGPRX2 to investigate the effect of 18 naturally occurring non-synonymous single

Predictive validation of the repeated low-dose reserpine rodent model of parkinsonism.

L-DOPA (LD) is the gold-standard treatment of motor symptoms in Parkinson's disease (PD), conventionally used to prove the predictive value of PD animal models. This study aimed to investigate the predictive validity of an adaptation of the conventional reserpine model of PD: the repeated low-dose reserpine administration in rodents, which promotes progressive motor impairment in catalepsy, vacuous chewing behavior tests, and neurochemical deficits. Specifically, we investigated the effects of acute and chronic treatment with LD on motor alterations induced in mice by this modified protocol. S
