Parkinson's disease as a somato-cognitive action network disorder.

Parkinson's disease (PD) is an incurable neurological disorder that often begins insidiously with sleep disturbances and somatic symptoms, progressing to whole-body motor and cognitive symptoms1-5. Dysfunction of the somato-cognitive action network (SCAN)-which is thought to control action execution6,7 by coordinating arousal, organ physiology and whole-body motor plans with behavioural motivation-is a potential contributor to the diverse clinical manifestations of PD. To investigate the role of the SCAN in PD pathophysiology and treatments (medications, deep-brain stimulation (DBS), transcran

The abnormal thyroxine signals triggers podocyte apoptosis in DN mice.

Podocyte injury is a pivotal factor in the advancement of diabetic nephropathy (DN). The present study aimed to delineate the influence of disrupted thyroxine signaling on podocyte apoptosis in DN mouse models. We employed bioinformatics analyses, coupled with Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment assessments, to identify differentially expressed genes (DEGs) associated with thyroxine signaling in both human and murine DN datasets. Subsequently, we elucidated the function of thyroid hormone receptor α1 (THRA1) and nuclear receptor co-repressor 1 (NCOR

Interferon-Driven Tryptophan Metabolism Links Inflammation and Mental Health in Juvenile Dermatomyositis.

BACKGROUND: Children with juvenile dermatomyositis (JDM) experience significant mental health burdens, including anxiety and depression. Inflammatory activation can alter tryptophan metabolism, particularly through IDO-driven kynurenine pathway induction, which has been implicated in diseases such as mood disorders and SLE. We therefore investigated skewed tryptophan metabolism and its association with disease activity and mental health symptoms in JDM. METHODS: Serum samples from JDM, juvenile idiopathic arthritis (JIA), and healthy controls (HC) were analysed for tryptophan metabolites by EL

BLOC1S1 variants cause lysosomal and autophagic defects resulting in a hypomyelinating leukodystrophy with epileptic encephalopathy.

BLOC1S1 encodes a subunit shared by the BLOC-1 and BLOC-one-related complex (BORC) hetero-octameric complexes that regulate various endolysosomal processes. Here, we report the identification of seven distinct variants in BLOC1S1 in 11 individuals from seven independent families presenting with early psychomotor delay, hypotonia, spasticity, epileptic encephalopathy, optic atrophy, and leuko-axonopathy with hypomyelination. A subset of the affected individuals also have features of hypopigmentation and ocular albinism that are similar, although milder, than those of individuals with BLOC-1-rel

Assessing the Cardiovascular Effects of Levothyroxine Use in an Ageing UK Population with Subclinical Hypothyroidism: Emulated Target Trial (ACEL-UK-ETT).

BACKGROUND: Thyrotropin levels increase with age, but standard reference intervals do not account for this, potentially leading to overdiagnosis of subclinical hypothyroidism (SCH) and overuse of levothyroxine in older adults. METHODS: Using data from The Health Improvement Network, this observational emulated target trial study assessed 10-year outcomes in adults over 50 years with SCH (thyrotropin 4.1-10.0mU/L, free thyroxine 10.0-24.0 pmol/L) who were prescribed levothyroxine versus those who were not. Subgroup analyses were limited to patients with age-specific thyrotropin levels. The pri

The role of gut microbes in production of aromatic carboxaldehydes.

Gut microbes play an important role in maintaining our health through the alteration of available nutrients and by the production of small molecules/metabolites. Indole-3-carboxaldehyde (I3A) is an aromatic carboxaldehyde (ArA) synthesized by gut microbes from the aromatic amino acid tryptophan, and has been mechanistically linked to antitumor activity, intestinal homeostasis, and metabolic syndrome. However, the capacity of gut microbes to produce other ArAs and their associations with host health remains largely unexplored due to a lack of methods for their detection and quantification. A st

Haplotype-Based Analysis of OCA2 Variants in Oculocutaneous Albinism.

OCA2, a melanosome transmembrane spanning protein, functions to regulate melanosomal pH, optimizing production of melanin pigment. OCA2 is one of eight non-syndromic autosomal recessive oculocutaneous albinism (OCA) loci and is the second most common cause of OCA worldwide. Genome wide association studies (GWAS) have identified OCA2 coding and regulatory variants linked to common skin and eye color pigment variation, skin cancer susceptibility, and retinal pigment epithelium tissue metrics. Within a cohort of 106 OCA2 probands with two biallelic OCA2 variants, a total of 74 distinct OCA2 rare

A microbiota-IPA axis facilitates intestinal stem cell-mediated regeneration in colitis through a Hopx-associated program.

The gut microbiota plays a crucial role in maintaining intestinal stem cell (ISC) homeostasis and epithelial barrier integrity. Here, we report that Blautia coccoides (B. coccoides) is significantly reduced in inflammatory bowel disease (IBD) patients and dextran sulfate sodium (DSS)-induced colitis mice. Through an integrated approach combining RNA sequencing, metabolomic profiling, and ISC lineage tracing across multiple mucosal injury models, we demonstrate that B. coccoides colonization enhances β-hydroxybutyrate (BHB) production in intestinal epithelial cells (IECs), which activates HOPX⁺

Microbial tryptophan metabolism activates host lysosomal activity to facilitate lipid breakdown.

Lysosomes are central to lipid metabolism, yet how gut microbiota-derived metabolites regulate lysosomal function to influence host lipid homeostasis remains unknown. Here, we identify a mechanism in which bacterial tryptophan metabolism activates lysosomal activity to promote lipid breakdown in Caenorhabditis elegans, and show that the bacterial tryptophan metabolite indole recapitulates these effects in mammalian hepatocytes. By developing a lysosomal-responsive lipid reporter in C. elegans to screen for bacterial metabolic states that modulate host lipid storage, we discover that Escherichi

Levodopa in retinal disease: Dopamine pathways, neuroprotective mechanisms, and clinical evidence.

We review the role of levodopa (L-DOPA), a dopamine precursor used to treat Parkinson disease, as a candidate for retinal neuroprotection, focusing on retinal dopamine biology, mechanisms of action, and evidence from preclinical and clinical studies. Selected experimental and clinical studies indicate that L-DOPA and related dopaminergic interventions influence multiple retinal pathways relevant to degeneration. L-DOPA, synthesized in the pigmented retinal pigment epithelium, activates GPR143 signaling and promotes the release of neuroprotective factors. In animal studies, L-DOPA supplementati

Renoprotection by 5-Methoxytryptophan in Kidney Disease.

Kidney disease, be it acute or chronic, has a complex pathology and is a significant human health problem. Increasing interest has been focused on exploring therapeutic targets that can be used to safeguard kidney function under a variety of detrimental conditions. In this article, we review the protective effects of 5-methoxytryptophan (5-MTP), a tryptophan metabolite, on kidney injury. Published studies indicate that serum 5-MTP level is decreased in patients with chronic kidney disease (CKD), suggesting that 5-MTP is a biomarker for CKD and has therapeutic values. Indeed, rodent models of k

Plasma neurotransmitter-related metabolite alterations in adolescent major depressive disorder with and without psychotic features.

BACKGROUND: Whether adolescent major depressive disorder (MDD) with psychotic features has a distinct peripheral metabolite signature remains uncertain. We compared targeted plasma neurotransmitter-related metabolites in psychotic MDD, non-psychotic MDD, and healthy controls. METHODS: We enrolled 220 adolescents (53 psychotic MDD, 67 non-psychotic MDD, 100 controls) and quantified 14 metabolites spanning tryptophan, glutamate-GABA, and catecholamine pathways using LC-MS/MS. Age- and sex-adjusted ANCOVA tested group effects on metabolites, biologically informed ratios, and pathway composites w

Human IDO2 exhibits unique binding affinities distinct to those of human IDO1.

Indoleamine 2,3-dioxygenase 2 (IDO2) is a heme enzyme in the kynurenine pathway that shares high structural similarity with IDO1 but exhibits markedly lower catalytic activity. To clarify the molecular basis of this difference, we performed spectroscopic, biochemical, and crystallographic analyses of human IDO2. We found that IDO2 binds L-tryptophan (L-Trp) in a flipped orientation stabilized by the IDO2-specific residue His143, which results in inefficient catalysis. Replacement of His143 with tyrosine, the corresponding residue in IDO1, restored an IDO1-like binding mode of L-Trp and enhance

The role of the FT3/FT4 ratio in predicting remission and relapse in pediatric Graves' disease.

UNLABELLED: Long-term antithyroid drug (ATD) therapy is often required in pediatric Graves' disease, yet adherence can be challenging. Therefore, identifying reliable predictors of remission and relapse is crucial for optimizing disease management. This study aimed to evaluate the role of the baseline FT3/FT4 ratio in predicting remission and relapse, alongside other potential markers, including TRAb levels, thyroid volume, and body mass index (BMI). This retrospective, single-center study included pediatric patients diagnosed with Graves' disease and treated with ATDs. Age, sex, BMI, thyroid

Thyrotropin Controversy in Subclinical Thyroid Disorders.

AIM: The thyrotropin controversy in subclinical thyroid disorders (STDs) is among the most common endocrine disorders globally. In India, approximately 42 million people suffer from thyroid disorders, with subclinical hypothyroidism (SCH) affecting about 9.4% of the population. SCH is more prevalent in females (11.4%) compared to males (6.2%). DISCUSSION: The diagnosis and treatment of SCH and subclinical hyperthyroidism (SHT) are controversial. SCH is often diagnosed based on biochemical markers, as patients may be asymptomatic or exhibit vague symptoms. Thyrotropin (TSH) levels may be eleva

Structural determinants for the monomeric and phospholipid-transport P4B-ATPase via comprehensive in silico analysis of P4-ATPase structures.

The P4-ATPase family of phospholipid flippases plays a critical role in maintenance of membrane asymmetry and cellular protein traffic and eukaryotic homeostasis. Several structures of these phospholipid flippases have been resolved, along with extensive biochemical characterization of the substrate transport properties. However, an essential subfamily of monomeric phospholipid flippases, the P4B-ATPases, remains to be characterized in depth. While P4B-ATPases appear to share similar lipid transport properties to their heterodimeric counterparts, the P4A-ATPases, the basis of their substrate t
