Periodontal inflammation and tryptophan-kynurenine metabolism in Parkinson's disease.

OBJECTIVE: The kynurenine pathway(KP) of tryptophan catabolism is a major regulator of the immune response. Metabolites of this pathway may have protective or degenerative effects on the nervous system. Based on our recent studies, we tested the hypothesis that KP metabolites play a role in the pathogenesis of the link between Parkinson’s disease (PAD) and Periodontitis (PD). MATERIALS AND METHODS: Saliva and serum samples were collected from Stage III, Grade B periodontitis patients with PAD (Parkinson+periodontitis group, n = 20) and without PAD (periodontitis group, n = 24), and 24 periodon

Gut-kidney axis: Dysbiosis and renal disease.

According United States renal data system the morbidity rate for chronic kidney disease (CKD) is 2.5 times than patients not affected by CKD and the mortality rate is 144.9 per 1000 persons-years. The gut microbiota is involved in uremic toxins (UTs) production. This fact was demonstrated by experiments in rats, which revealed better survival in CKD rats that were deprived of the gut microbiota. In men, UT levels are low in CKD patients without a colon. Diet may affect the gut microbiota through food additives such as prebiotics, probiotics and post biotics. Conservation processes and food pro

A drug-microbiome-drug interaction impacts co-prescribed medications for Parkinson's disease.

Simultaneous prescription of multiple drugs is widespread in medicine. Although the gut microbiome is implicated in drug responses, its role in mediating drug-drug interactions is unexplored. Catechol-O-methyltransferase inhibitors (COMT-I), a class of drugs used alongside levodopa (L-DOPA) to treat Parkinson's disease symptoms, can alter microbiome composition in patients. Here we characterize the antibiotic properties of COMT-I drugs in vitro, ex vivo and in vivo and dissect how these interactions alter microbiome-mediated L-DOPA metabolism in vitro and ex vivo. Notably, in vitro iron availa

Menopause-Related Health Outcomes in Women with Differentiated Thyroid Carcinoma Receiving Long-Term TSH Suppression after Total Thyroidectomy.

BACKGROUND: There is uncertainty regarding whether thyroid-stimulating hormone (TSH) suppression therapy in postmenopausal women with differentiated thyroid cancer (DTC) is associated with menopause-specific health concerns. This study aimed to determine whether prolonged TSH suppression confers an additional burden on postmenopausal health by comprehensively evaluating cardiovascular health (CVH), quality of life (QoL), muscle mass, and bone health using menopause-specific assessment tools. METHODS: A cross-sectional study was conducted involving three groups of postmenopausal women 107 pati

Tryptophan metabolism in embryo implantation and decidualization.

Tryptophan is an essential amino acid for the human body and is crucial for protein synthesis. It is primarily metabolized through the kynurenine, 5-hydroxytryptamine, and indole pathways. The various bioactive compounds produced through tryptophan metabolism can regulate a range of physiological functions, including digestive, metabolic, immune responses, and neurological functions. New evidence supports a close relationship between tryptophan metabolism and embryo implantation; the levels or ratios of tryptophan metabolites, such as kynurenine, serotonin, and melatonin, significantly correla

Post-synthetic modification strategy for constructing dual-emission fluorescent sensor toward visual detection and quantification of 3-nitrotyrosine biomarker.

BACKGROUND: As the biomarker of oxidative stress (OS), 3-nitrotyrosine (3-NT) levels in serum is closely associated with disease progression, and the development of rapid and reliable fluorescent sensor is therefor urgent. Metal-organic frameworks (MOFs) are attractive for their diversified, easily designed and functionalized structures, yet most MOF-based 3-NT fluorescent sensors are single-signal depended, resulting in inadequate anti-interference capability and insufficient reliability. Developing MOF-based dual-emission fluorescent sensors for precise detection of serum 3-NT levels thus re

AXL prevents amyloid-β-induced microglial ferroptosis by sustaining SLC2A3-mediated mitochondrial respiration.

Dysregulated iron metabolism is a pivotal driver of Alzheimer's disease (AD). Excess iron promotes Aβ aggregation and tau hyperphosphorylation, thereby accelerating disease progression. Serving as the primary iron reservoir in the central nervous system, microglia are intrinsically susceptible to ferroptosis, thereby amplifying neurotoxicity to neighboring neurons. While plaque-associated receptors (e.g., TREM2, AXL, MERTK) govern microglial responses, their precise contribution to metabolic susceptibility to ferroptosis remains elusive. Here, we identify the receptor tyrosine kinase AXL as a

A frontotemporal dementia-like phenotype in schizophrenia: links to striatal dopamine and iron accumulation.

Some schizophrenia patients share characteristics with behavioral variant frontotemporal dementia (bvFTD) including gray matter volume (GMV) similarities, suggesting overlapping brain mechanisms that may contribute to disease heterogeneity in schizophrenia. However, it is not yet understood whether schizophrenia patients with a bvFTD-like GMV signature also show additional features of bvFTD. Seventy-six patients with schizophrenia (mean age 31.2 years; 71% male) from a previous clinical trial (NCT04038957) underwent structural magnetic resonance imaging (MRI) to measure GMV. Healthy controls (

Cuttlefish ink nanoparticle hydrogels ameliorate osteoarthritis via concurrent inhibition of macrophage pyroptosis and chondrocyte senescence.

Osteoarthritis (OA) is a debilitating joint disorder characterized by synovial inflammation and cartilage degradation; however, current therapeutic options fail to simultaneously address these dual pathological progressions. Here, we present clinical evidence identifying synovial macrophage (Mφ) pyroptosis and chondrocyte senescence as pivotal features of OA, which amplify joint damage through sustained inflammatory cascades and metabolic dysfunction. To address this, we developed cuttlefish ink-derived melanin nanoparticle (CIMNP) hydrogels designed to target the ROS/HIF-1α/glycolysis axis, t

Blood-based biomarkers of ferroptosis in Parkinson's disease.

BACKGROUND: As Parkinson's disease progresses, patients require second-line treatments such as subthalamic stimulation, the benefits of which may be diminished by the onset of non-dopaminergic axial motor and cognitive disorders. This study aimed to identify blood biomarkers of ferroptosis for predicting the progression of Parkinson's disease at the stage of L-DOPA-related complications. METHODS: We analyzed 598 blood samples from patients with PD enrolled in the French multicentric PREDISTIM study. Putamen dopamine synthesis, vesicular storage, and metabolism in Parkinson disease.

BACKGROUND: Severe putamen dopamine depletion in Parkinson disease (PD) has been attributed to nigrostriatal denervation; however, there are also functional abnormalities in extant terminals (the "sick-but-not-dead" phenomenon). Rates of intra-neuronal processes of synthesis, storage, and metabolism of dopamine complexly influence releasable dopamine stores but have not yet been systematically estimated. METHODS: Post-mortem empirical data were available about putamen tissue contents of 7 reactants, including the autotoxic dopamine metabolite 3,4-dihydroxyphenylacetaldehyde (DOPAL). We constr

Natural history of thyroid function in ageing: an individual participant data analysis of 137 488 participants from 31 prospective cohort studies.

BACKGROUND: Evidence regarding thyroid function changes with ageing remains inconsistent and the implications of potential changes are unclear. We aimed to investigate ageing-related thyroid function changes and their associations with mortality. METHODS: In this individual participant data (IPD) analysis, prospective population-based cohorts were eligible for inclusion when data on thyroid function measurements and mortality were available in individuals aged 18 years and older. Eligible datasets were identified through a systematic search of PubMed. We excluded cohorts of participants with

Stage-Dependent Changes in Kynurenine Pathway Enzyme Expression Suggest Immune-Related Involvement in Bladder Cancer Progression.

UNLABELLED: L-tryptophan (Trp) metabolism through the L-kynurenine (Kyn) pathway contributes to immune escape in neoplasms, including bladder cancer (BC). Indoleamine 2,3-dioxygenase-1 (IDO1) is a key enzyme, and its catabolites-Kyn, 3-hydroxykynurenine (3HK), and 3-hydroxyanthranilate (3HAA)-depend on other enzymes and play a crucial role in immunomodulation. OBJECTIVES: We investigated the association between Kyn pathway enzyme expression and BC prognostic factors. METHODS: Data from 165 bc patients in the GEO DataSets were analyzed, comparing staging, tumor grade, progression, and recurre

Convergent evolution of albinism in cave animals is driven by shared mechanisms and adaptive advantages.

Loss of pigmentation is a hallmark of cave-dwelling animals, yet the mechanisms and evolutionary forces driving this convergence remain unclear. Here, we investigate the cellular and biochemical basis of albinism across phylogenetically diverse cave species, including annelids, mollusks, and vertebrates. Across all taxa, melanin loss consistently results from disruption of the first step in the biosynthetic pathway. Despite this metabolic block, cells capable of melanin synthesis are retained showing distributions similar to pigmented surface relatives and contributing to innate immune respons

Analysis of multi-trait evolution across independently evolved cavefish populations reveals shared and independent evolution of suites of traits.

Environmental perturbations often lead to the evolution of multiple traits. Determining whether shared genetic factors underlie multi-trait evolution is a central question in evolutionary biology. In the Mexican tetra, Astyanax mexicanus, cave-dwelling populations have repeatedly evolved multiple traits. The repeated evolution of these traits, paired with robust environmental differences between the surface and cave habitats, provide an opportunity to investigate the genetic basis of multi-trait evolution. Here, we investigate the extent to which shared genetic mechanisms underlie the repeated

European perspectives on albinism: Epidemiology, clinical features, and management.

Albinism is a genetically heterogeneous group of disorders characterized by reduced or absent melanin synthesis, leading to variable hypopigmentation of the skin, hair, and eyes, and associated ocular abnormalities such as foveal hypoplasia, nystagmus, and reduced visual acuity. Although considered rare, the prevalence of albinism varies widely across regions and is likely underestimated in Europe, particularly because mild phenotypes in fair-skinned populations may be overlooked. In people with albinism in Europe (PWA), reduced cutaneous pigmentation increases susceptibility to ultraviolet ra

Oculo-Cutaneous and syndromic albinisms: Epidemiology, clinical spectrum and diagnosis.

BACKGROUND AND OBJECTIVES: Oculo-Cutaneous Albinism and Ocular Albinism are worldwide autosomal and X-linked recessive disorders, respectively, resulting from mutations in at least 21 genes that affect melanin production or transport in the skin, hair and eyes. The purpose of this review is to assist clinicians in recognizing its main clinical manifestations. The review is structured into three main sections: general manifestations common to all albinism subtypes, the distinct features of non-syndromic subtypes (oculocutaneous albinism types 1-8), and the syndromic forms (Chediak-Higashi syndr

Comparative Transcriptome and Gene-Editing Analyses Reveal the Molecular Mechanism of Albinism in the Taiwanese Loach.

The Taiwanese loach (Paramisgurnus dabryanus ssp. Taiwanese), known for its nutritional and ornamental qualities, exhibits a rare form of albinism. Specifically, certain individuals display golden-pink eyes and a golden-red body color devoid of blotches, thereby attracting consumers. However, the principal genes regulating this phenotype remain unidentified. Consequently, this study conducted a transcriptome comparative analysis of the skin between wild-type (M) and mutant (N) Taiwanese loaches. A total of 6,152 differentially expressed genes (DEGs) were identified, comprising 2,467 down-regul
