Loss-of-function variants in the melanocortin 1 receptor (MC1R) gene are a well-established cause of red hair in European populations. However, the genetic spectrum and phenotypic effects of MC1R variants in East Asians remain poorly defined, as red hair is exceedingly rare in this population. To characterize the clinical and genetic features of MC1R-associated red hair and hypopigmentation in Chinese pediatric patients and to expand the genotypic spectrum of MC1R in East Asian populations, four unrelated Chinese children with red or reddish-brown hair underwent comprehensive dermatologic and

DOPA decarboxylase (DDC) is a key enzyme that connects melanin production and innate immunity in insects, yet its regulatory network in major lepidopteran pests remains poorly understood. We combined CRISPR/Cas9 gene editing with multi-omics analysis to explore DDC's role in immune responses in the invasive pest Spodoptera frugiperda. Knocking out DDC caused complete albinism, severe developmental delays, pronounced developmental defects, and increased susceptibility to bacterial challenge. Following the E. coli challenge, DDC-mutant larvae showed substantially lower final survival than infect

Oculocutaneous albinism-characterized by absent or decreased melanin in the skin, eyes and hair-often co-occurs with sensory, skin and immunity disorders. The genetic basis of albinism in humans is complex, with many loci implicated in multiple forms of the disorder. Less is known about the underlying genetic causes of albinism and leucism in other species, and cross-species studies of the molecular correlates of hypopigmentation could highlight common and conserved pathways underlying mammalian pigmentation disorders. We characterize the putative causal loci of reduced pigmentation in an Egyp

Albinism is typically lethal in autotrophic plants due to the absence of photosynthetic pigments and functioning chloroplasts. Yet, rare exceptions occur where achlorophyllous individuals persist in natural ecosystems. We investigated the physiological, anatomical and isotopic characteristics of naturally occurring albino European beech (Fagus sylvatica L.) trees in the Moravian Karst, Czechia. One albino individual, estimated to be ~30 years old, represents an unprecedented case of long-term survival without photosynthesis in a woody angiosperm. Using a multi-parameter approach-including stab

GPR143, originally identified as the gene product of ocular albinism 1 (OA1), a G protein-coupled receptor (GPCR), can function as a receptor for l-3,4-dihydroxyphenylalanine (DOPA), a precursor of dopamine (DA). To examine the physiological and pathophysiological roles of GPR143, we analyzed the behavior of Gpr143 gene-deficient (Gpr143-/y) mice. We performed comprehensive behavioral analyses including the prepulse inhibition test, sucrose preference test, light-dark exploration test, etc. and microarray analysis. Gpr143-/y mice displayed a mixed psychosis-like phenotype: impaired prepulse in

PURPOSE: To present a case of type II oculocutaneous albinism (OCA2) diagnosed in infancy following the finding of nystagmus, and to review the diagnostic process and the management of this disorder.
OBSERVATION: A 4-month-old female presented with subtle, roving eyes that were initially attributed to normal development. A subsequent evaluation by a pediatric ophthalmologist, prompted by a high index of suspicion, confirmed the findings of nystagmus, mild foveal hypoplasia, and astigmatism. Genetic testing confirmed the presence of pathological variants of the OCA2 gene, leading to a diagnosi

We present a comprehensive molecular and epidemiological analysis of albinism in a Russian cohort, comprising 177 probands with isolated (OCA1, OCA2, OCA3, OCA4, and OA) and syndromic (HPS-associated) forms of the disease. The comparative analysis of population frequencies between the National Genetic Initiative "100,000 + Me" (NGI) project and GnomAD (v3.1.2) revealed variants in the TYR gene (NM_000372.5):c.650G>A and c.1037-7 T>A, which are specifically prevalent in the Russian population. The functional analysis was provided for variants in the TYR gene, which can probably affect splicing.

Only a few records of pigmentation anomalies, particularly hypopigmentation conditions such as albinism, have been reported among Neotropical non-human primates. In Brazil, there are only 14 documented cases of pigmentation anomalies, with just two instances of albinism recorded within the Cebidae family. To our knowledge, no pigmentation polymorphisms have been documented in the Pitheciidae family to date. In this study, we report the first documented case of albinism in Callicebus nigrifrons (the black-fronted titi monkey) within the largest remnant of the Atlantic Forest in the State of Min

RATIONALE: Griscelli syndrome (GS) is a rare autosomal recessive disorder marked by partial oculocutaneous albinism, immunodeficiency, and neurological issues. It has 3 types based on genetic mutations. This report focuses on a patient with GS type 2, characterized by immune abnormalities and neurological symptoms, with only 160 cases documented globally.
PATIENT CONCERNS: A 1-year-old boy presented with hyperthermia, diarrhea, and vomiting, revealing hypopigmented skin and silvery-gray hair. He exhibited tachycardia, abdominal distension, hepatosplenomegaly, and signs of immunocompromise. Ne

While pigmentation disorders such as albinism have been documented in a range of elasmobranch species, including the American cownose ray (Rhinoptera bonasus), the implications of these rare conditions for behavior, social dynamics, and fitness remain speculative and unverified. Here, we report a case of aberrant pigmentation in a single cownose ray observed schooling among conspecifics in the nearshore waters of Chesapeake Bay, Virginia, USA. The individual displayed pronounced white coloration against a notably dark dorsal surface of both pectoral fins, contrasting sharply with the otherwise

Oculocutaneous albinism type 1 (OCA1) caused by pathogenic variants of the TYR gene is an autosomal recessive disorder of pigmentation characterized by reduced biosynthesis of melanin pigment in skin, hair, and eyes. We had the opportunity to examine five East Cameroon Baka rainforest hunter-gatherers (historically called "Pygmies") with albinism and belonging to three different families. Screening of known albinism genes revealed a homozygous missense variant in the TYR gene, NM_000372.5: c.1109T>C; p.Met370Thr. In addition, one patient was also hemizygous for a variant in GPR143, the gene in

The SLC45 family emerges as a proton-coupled sugar transport system that complements the classical SGLT (SLC5) and GLUT (SLC2) transporters in mammals. This review synthesizes recent advances on four family members (SLC45A1–A4), covering their structural features (12-pass transmembrane topology with a pH-sensing cytoplasmic loop), genomic organization across chromosomes 1p36.1, 5p13.3, 1q32.1, and 8q24.3, and diverse physiological roles in brain glucose homeostasis, melanogenesis, lipid metabolism, and cancer metabolic reprogramming. We highlight disease associations, including SLC45A1 mutatio

Dopa decarboxylase (DDC) is a crucial enzyme that regulates melanin synthesis and plays an essential role in cuticular pigmentation and insect development. However, the function of DDC in the fall armyworm, Spodoptera frugiperda, is not yet well understood. In this study, we knocked out the DDC gene and found that mosaic mutants displayed an albino phenotype across all life stages, including larval, pupal, and adult stages. Notably, homozygous DDC mutants showed complete larval albinism, exhibited developmental delays, and frequently failed to pupate, ultimately leading to death. Histological

Artemisia argyi is a significant medicinal plant in China. To date, no viruses have been reported to infect A. argyi. In this study, using the RNA-seq technique, we identified a novel marafivirus in A. argyi plants showing leaf mottle and albinism symptoms, which we have tentatively named “Artemisia argyi albinism-associated virus” (AAAaV). The AAAaV genome is a positive-sense single-stranded RNA with a length of 6749 nt, excluding the poly(A) tail, containing a single open reading frame (ORF) encoding a large polyprotein. Phylogenetic analysis based on the full-length genome and polyprotein a

Body color is the most prominent phenotypic trait in animals. To investigate the molecular regulatory mechanisms underlying skin pigmentation and body color in Channa argus, single-nucleus RNA sequencing technology was employed to analyze cell diversity and functional changes in the skin of normal and albino C. argus. Three pigment-related cell types, seven immune-related cell types, and nine other skin-related structural and functional cell types were identified. The skin of albino C. argus, which appears white to the naked eye, contains numerous melanocytes and iridophores with reflective si
