Human recombinant tyrosinase destabilization caused by the double mutation R217Q/R402Q.

Oculocutaneous albinism is an autosomal recessive inherited disorder associated with mutations in the TYR gene. A single missense change in the tyrosinase (Tyr) could result in partial or complete loss of catalytic activity. The effect of two genetic mutations in the same Tyr as the molecule is less studied. Here, we study single mutation variants, R217Q, R402Q, and a double mutant variant, R217Q/R402Q, to establish a link between alterations at the level of the atomic model of the protein and the disease phenotype. Human recombinant intra-melanosomal Tyr domains of Tyr and three mutant varian

Truncated SPAG9 as a novel candidate gene for a new syndrome: Coarse facial features, albinism, cataract and developmental delay (CACD syndrome).

Sperm-associated antigen 9 (SPAG9) is a member of cancer-testis antigen, having characteristics of a scaffold protein, which is involved in the c-Jun N-terminal kinase JNK signaling pathway, suggesting its key involvement in different physiological processes, such as survival, apoptosis, tumorigenesis, and cell proliferation. We identified two families (A and B) having multisystem features like coarse facial features, albinism, cataracts, skeletal abnormalities, and developmental delay. Whole genome sequencing (WGS) in families A and B revealed a homozygous frameshift variant (c.903del; p.Phe3

Advancing Insights into Pediatric Macular Diseases: A Comprehensive Review.

Pediatric macular disorders are a diverse group of inherited retinal diseases characterized by central vision loss due to dysfunction and degeneration of the macula, the region of the retina responsible for high-acuity vision. Common disorders in this category include Stargardt disease, Best vitelliform macular dystrophy, and X-linked retinoschisis. These conditions often manifest during childhood or adolescence, with symptoms such as progressive central vision loss, photophobia, and difficulty with fine visual tasks. Underlying mechanisms involve genetic mutations that disrupt photoreceptor a

Dyadic adjustment of persons with albinism and their significant other: A Bayesian mediation model of the Actor-Partner Interdependence (APIMeM).

BACKGROUND: Limited standardized empirical research exists in France on the psychosocial implications of albinism, prompting an investigation into how affected persons adapt to their disabilities. Recent advancements in health psychology have led researchers to adopt a systemic approach, considering disabilities and involving close relatives in the adaptation process. The aim of this study was to explore the mediating role of dyadic coping within family dyads, where one is a person with albinism (PWA) and the other is a close relative. METHODS: The study included 38 family dyads with each mem

A Recessive oca2 Mutation Underlies Albinism in Xiphophorus fish.

Oculocutaneous albinism (OCA) is a group of genetic disorders characterized by impaired melanin production, leading to reduced pigmentation in the skin, hair, and eyes. Xiphophorus , a genus of small freshwater fish, has been a pivotal model organism in pigmentation disorder research, providing key findings in the genetic pathways governing physiological and pathological pigment cell biology. Leveraging the well-established research framework provided by Xiphophorus , we have identified a spontaneously occurring albinism phenotype in swordtail fish Xiphophorus hellerii . Genetic mapping of alb

Development and Evaluation of a Rapid Visualisation Detection Method for Ameson portunus Based on RPA-LFD.

The Ameson portunus is a prevalent pathogen affecting Portunus trituberculatus, which can infect P. trituberculatus and cause albinism, seriously damaging its economic value. Therefore, there is a pressing need for an efficient detection platform to rapidly and sensitively identify A. portunus. In this study, we have developed a method known as recombinase polymerase amplification combined with lateral flow dipstick (RPA-LFD) for the swift detection of A. portunus. Three sets of specific primer pairs and a probe were designed according to the spore wall protein (SWP) sequence of A. portunus, i

Chiasmal Decussation in Oculo-Cutaneous Albinism Type 8.

PURPOSE: Albinism is a genetic disorder characterized by a defect in melanin biosynthesis. Ophthalmological and dermatological impairments vary according to the patient genotype and are highly heterogenous. Recently, variants in the DCT gene were showed to be responsible for a new type of oculocutaneous albinism (OCA) named OCA8. We report the ophthalmological, electrophysiological, and dermatological characteristics of three patients with genetically confirmed OCA8. METHODS: This is a retrospective study of three patients with OCA8. Complete dermatological, ophthalmological, and orthoptic ex

Characterization of a novel green-revertible albino mutant in alfalfa (Medicago sativa L.).

High-temperature-sensitive leaf color mutants are ideal materials for studying photosynthetic pigment biosynthesis and corresponding response mechanisms under heat stress. Here, we provide the first report of albinism occurrence in alfalfa and characterize the high-temperature albino regreen (har) mutant of alfalfa, which presents albino leaves when exposed to temperatures ≥35 °C and is not specific to developmental stage. Genetic analysis demonstrated that the albino trait exhibits dominant inheritance. Agronomic trait evaluations revealed that the har mutants were slightly but negatively aff

Sequence Context-Agnostic TadA-Derived Cytosine Base Editors for Genome-Wide Editing in Zebrafish.

Single-nucleotide variants (SNVs) represent a significant form of genetic variation linked to various diseases. CRISPR-mediated base editing has emerged as a powerful method for modeling diseases caused by SNVs, particularly in zebrafish, which serve as an excellent platform for investigating disease mechanisms and conducting drug screenings. However, existing cytosine base editors (CBEs) for zebrafish often have broad editing windows and strong sequence preferences, limiting their effectiveness. In this study, zebrafish (z) TadA-derived cytosine base editors, termed zTadA-CBEs, are developed

Emergence of inflammatory fibroblasts with aging in Hermansky-Pudlak syndrome associated pulmonary fibrosis.

The longitudinal cellular interactions that drive pulmonary fibrosis are not well understood. To investigate the disease underpinnings associated with fibrosis onset and progression, we generated a scRNA-seq atlas of lungs from young and aged mouse models of multiple subtypes of Hermansky-Pudlak syndrome (HPS), a collection of rare autosomal recessive diseases associated with albinism, platelet dysfunction, and pulmonary fibrosis. We have identified an age-dependent increase in SAA3+ inflammatory lung fibroblasts in HPS mice, including in double-mutant HPS1-2 mice which develop spontaneous fib

Synonymous but Significant: New Findings of Pathological Variants in Hermansky-Pudlak Syndrome.

Hermansky-Pudlak syndrome (HPS) is a rare autosomal recessive disorder characterized by oculocutaneous albinism and systemic complications, including bleeding tendencies. While 11 genes associated with HPS have been identified, cases of HPS5 remain exceedingly rare, particularly in Japan. Here, we report two Japanese patients with novel pathological HPS5 variants, expanding the genetic spectrum of this disorder. Both patients exhibited typical features of mild skin and hair hypopigmentation, and significant ocular involvement. Genetic analysis revealed a heterozygous nonsense variant, NG_00887

Proteome and Metabolome Analyses of Albino Bracts in Davidia involucrata.

Although the mechanisms underlying albino phenotypes have been examined in model plants and major crops, our knowledge of bract albinism is still in its infancy. Davidia involucrata, a relic plant called dove tree, is best known for the intriguing trait with a pair of white bracts covering the capitula. Here, comparative physiological, cytological, proteomic, and metabolomic analyses were performed to dissect the albinism mechanism of D. involucrata bracts. The bracts exhibited low chlorophyll and carotenoid contents, reduced photosynthetic efficiency, and impaired chloroplast structure. The s

Cerebellar control of targeted tongue movements.

The cerebellum is critical for coordinating movements related to eating, drinking and swallowing, all of which require proper control of the tongue. Cerebellar Purkinje cells can encode tongue movements, but it is unclear how their simple spikes and complex spikes induce changes in the shape of the tongue that contribute to goal-directed movements. To study these relations, we recorded and stimulated Purkinje cells in the vermis and hemispheres of mice during spontaneous licking from a stationary or moving water spout. We found that Purkinje cells can encode rhythmic licking with both their si

The small molecule ML233 is a direct inhibitor of tyrosinase function.

Melanogenesis is the biological process regulating the synthesis of melanin pigments in melanocytes. Defective melanogenesis is associated with numerous human skin diseases, including, but not limited to, albinism, vitiligo, melasma, and hypo- and hyperpigmentation disorders. Tyrosinase is the rate-limiting enzyme controlling melanogenesis, and hence tremendous efforts have been made to identify potent and safe inhibitors of tyrosinase function. However, despite decades of research, currently there is no effective treatment that inhibits melanogenesis or tyrosinase activity with no adverse sid

Unifying Structure and Function Towards a Comprehensive Macular Evaluation in Eye Disorders: A Multi-Modal Approach Using Microperimetry and Optical Coherence Tomography.

OBJECTIVE: We present a new automatic and comprehensive framework to evaluate retinal sub-layer thickness and visual sensitivity at precise retinal locations through multi-modal registration of confocal scanning laser ophthalmoscope (cSLO) images obtained separately from an optical coherence tomography (OCT) device and a microperimeter. METHODS: We map consecutive B-scans onto the cSLO images after accounting for eye motion. Next, we coarsely register the SLO-microperimetry and cSLO-OCT images using SIFT, followed by precise elastic image registration. Subsequently, we ensure the quality of t
