Kynurenine metabolites unchanged after multinutrient supplementation in children with ADHD: a secondary data analysis from the MADDY study.

Attention-deficit/hyperactivity disorder (ADHD) affects up to 10% of children, often presenting with emotional dysregulation (irritability, anger). The kynurenine pathway, by which the amino acid tryptophan is converted into both neuroprotective and neurotoxic metabolites, was investigated for its role in ADHD pathophysiology. The MADDY study was an 8-week, multi-site, randomized controlled trial of a multinutrient supplement versus placebo in children 6-12 years old with symptoms of ADHD and emotional dysregulation. Participants from two U.S. sites (n = 84) provided baseline and week 8 urinar

Targeting Oxidative Stress and Apoptosis with Levothyroxine in Experimental Parkinson's Disease in Rats.

Parkinson’s disease is a long-term, progressive condition that affects both movement and other parts of life that aren’t motor-related. Cognitive decline is one of the most impactful non-motor symptoms, as it can seriously affect overall quality of life. Although existing dopamine replacement therapies, including levodopa, mainly focus on alleviating motor symptoms, they do not adequately address issues such as dyskinesia, non-motor deficits, and the need for neuroprotective treatments. This research aimed to explore the neuroprotective properties of levothyroxine (L-T4) in a PD animal model.

Diabetes-associated modifications in gut microbiota and tryptophan metabolism: implications for macrophage polarization and wound repair in mice.

BACKGROUND: Diabetic wound (DW) is a severe complication of diabetes with poor healing, linked to gut microbiota dysbiosis, metabolic imbalance, and macrophage polarization disorder. This study aimed to systematically explore the role of the gut-skin axis in DW. METHODS: A type 2 diabetes mellitus (T2DM) mouse model was established via high-fat diet feeding and streptozotocin injection. Wound healing was evaluated by histological and immunofluorescence analyses. Gut microbiota composition (16 S rRNA sequencing), serum non-targeted metabolomics (GC-MS), and targeted tryptophan metabolite detec

Tyrosine supplementation with high-protein diet as a therapeutic strategy for YARS1 deficiency.

PURPOSE: Biallelic pathogenic variants in YARS1 cause tyrosyl-tRNA synthase (TyrRS) deficiency that compromises the loading of tyrosine to its tRNA. YARS1 deficiency is characterized by impairment of neurological development, growth, liver function, and hematopoiesis. For other aminoacyl-tRNA synthetase deficiencies, supplementation of the respective amino acid and high-protein diet improved outcome. Whether tyrosine supplementation is effective in YARS1 deficiency is not known. METHODS: Nine individuals with YARS1 deficiency received tyrosine (7 with and 2 without a high-protein diet). Amino

TYK2 and IL-23 Pathway Therapies in Psoriasis: Associations With IL-6, Tryptophan Metabolism, and Depressive Symptoms.

BACKGROUND: Psoriasis and depression frequently coexist, creating a complex, bidirectional relationship that complicates treatment. This study, integrating clinical assessments with transcriptomic and metabolomic analyses, hypothesizes that TYK2 (tyrosine kinase 2) inhibitors possess a dual therapeutic potential to simultaneously address both dermatological manifestations of psoriasis and the frequently accompanied depressive symptoms. METHODS: In a cohort of 298 psoriasis patients evaluated using the Hospital Anxiety and Depression Scale (HADS), participants were categorized into a TYK2 inhi

Multi-omics reveals co-regulation of hepatic bile acid metabolism in laying hens by host genetics and the cecal Anaerostipes.

Bile acids (BA) are central regulators of lipid metabolism and key signaling molecules within the gut-liver axis. Dysregulation of BA is implicated in fatty liver hemorrhagic syndrome (FLHS), a prevalent metabolic disorder in laying hens characterized by hepatic lipid accumulation, hemorrhage, reduced egg production, and increased mortality. However, the mechanisms governing BA regulation in poultry remain poorly understood. Here, we integrated multi-omics analyses to dissect how host genetics and gut microbiota interact to modulate hepatic total bile acids (HTBA) levels. A total of 686 hens a

Honokiol attenuates diabetes by enriching Akkermansia muciniphila andregulating tryptophan metabolism in mice.

Diabetes mellitus (DM) is a chronic disease influenced by gut microbiome disturbances. Honokiol (HON), a low oral bioavailability compound from Magnolia officinalis bark, has demonstrated potential as a treatment for DM. This research investigates the effects of HON on gut microbiota and host metabolism to elucidate its mechanism of action in DM. After 8 weeks of intervention through fecal microbiota transplantation (FMT) or antibiotic treatment, HON improved glucose tolerance and lipid metabolism in a gut microbiota-dependent manner. Specifically, HON administration significantly increased Ak

Phenylalanine as a biomarker of disease in animals: Current evidence and future perspectives.

Phenylalanine (Phe), an essential aromatic amino acid, is emerging as a potential biomarker of metabolic and health status in animals. Altered Phe concentrations reflect changes in metabolic, inflammatory, and disease processes. In companion animals, shifts in serum or biofluid Phe have been linked to hepatopathies, inflammatory enteropathies, and neoplastic conditions. In livestock, variations in milk or serum Phe accompany mastitis, metabolic stress, lameness, and other production-related disorders. Evidence for cats, horses, poultry, and aquaculture species remains limited. Interpretation i

Testing dopaminergic markers of problematic social media use using neuromelanin-sensitive MRI.

BACKGROUND: Neuromelanin-sensitive MRI (NM-MRI) has demonstrated potential as a proxy measure of dopamine functioning in the brain. Altered NM-MRI has been associated with substance use disorders, yet remains unexplored in problematic social media use (PSMU). The current study aims to determine whether higher PSMU is linked to lower NM-MRI signal intensity in the substantia nigra and ventral tegmental area (SN-VTA). METHODS: Seventy-two young adults (18-35 years of age) completed the Bergen Social Media Addiction Scale to measure PSMU and underwent an NM-MRI scan. Half of the participants had

Development of a Fluorophore-Bound l-Tryptophan Derivative for Evaluating Indoleamine 2,3-Dioxygenase Activity by HPLC with Fluorescence Detection: An In Vivo Microdialysis Study Using Rat Kidney.

Evaluating the activity of indoleamine 2,3-dioxygenase (IDO), the rate-limiting enzyme in tryptophan (Trp) metabolism, is important because IDO is involved in immune tolerance and drives the production of Trp metabolites implicated in psychiatric disorders and cancer. This study aimed to design and develop a novel fluorescent l-Trp derivative to fluorometrically monitor Trp-catabolizing enzyme activity via IDO. To evaluate IDO activity in vivo, 7-N,N-dimethylamino-2,1,3-benzoxadiazole (DBD), a fluorophore, was covalently bound at the 5-position of the indole ring in Trp to produce 5-DBD-l-Trp.

Selective peroxynitrite-mediated protein nitration catalyzed by glyoxalase domain containing protein 4.

Tyrosine nitration alters the structure, function, and cellular localization of proteins and is implicated in the pathology of multiple diseases [G. Ferrer-Sueta et al., Chem. Rev. 118, 1338-1408 (2018), H. Ischiropoulos, Arch. Biochem. Biophys. 356, 1-11 (1998), I. Griswold-Prenner et al., J. Biol. Chem. 299, 105038-10554 (2023)]. Although protein nitration is assumed to proceed via nonspecific chemical mechanisms, it is highly selective, suggesting the possibility of enzymatic catalysis. Here, we showed that glyoxalase domain-containing protein 4 (GLOD4), a previously uncharacterized protein

Parkinson's disease as a somato-cognitive action network disorder.

Parkinson's disease (PD) is an incurable neurological disorder that often begins insidiously with sleep disturbances and somatic symptoms, progressing to whole-body motor and cognitive symptoms1-5. Dysfunction of the somato-cognitive action network (SCAN)-which is thought to control action execution6,7 by coordinating arousal, organ physiology and whole-body motor plans with behavioural motivation-is a potential contributor to the diverse clinical manifestations of PD. To investigate the role of the SCAN in PD pathophysiology and treatments (medications, deep-brain stimulation (DBS), transcran

The abnormal thyroxine signals triggers podocyte apoptosis in DN mice.

Podocyte injury is a pivotal factor in the advancement of diabetic nephropathy (DN). The present study aimed to delineate the influence of disrupted thyroxine signaling on podocyte apoptosis in DN mouse models. We employed bioinformatics analyses, coupled with Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment assessments, to identify differentially expressed genes (DEGs) associated with thyroxine signaling in both human and murine DN datasets. Subsequently, we elucidated the function of thyroid hormone receptor α1 (THRA1) and nuclear receptor co-repressor 1 (NCOR

Interferon-Driven Tryptophan Metabolism Links Inflammation and Mental Health in Juvenile Dermatomyositis.

BACKGROUND: Children with juvenile dermatomyositis (JDM) experience significant mental health burdens, including anxiety and depression. Inflammatory activation can alter tryptophan metabolism, particularly through IDO-driven kynurenine pathway induction, which has been implicated in diseases such as mood disorders and SLE. We therefore investigated skewed tryptophan metabolism and its association with disease activity and mental health symptoms in JDM. METHODS: Serum samples from JDM, juvenile idiopathic arthritis (JIA), and healthy controls (HC) were analysed for tryptophan metabolites by EL

Assessing the Cardiovascular Effects of Levothyroxine Use in an Ageing UK Population with Subclinical Hypothyroidism: Emulated Target Trial (ACEL-UK-ETT).

BACKGROUND: Thyrotropin levels increase with age, but standard reference intervals do not account for this, potentially leading to overdiagnosis of subclinical hypothyroidism (SCH) and overuse of levothyroxine in older adults. METHODS: Using data from The Health Improvement Network, this observational emulated target trial study assessed 10-year outcomes in adults over 50 years with SCH (thyrotropin 4.1-10.0mU/L, free thyroxine 10.0-24.0 pmol/L) who were prescribed levothyroxine versus those who were not. Subgroup analyses were limited to patients with age-specific thyrotropin levels. The pri

The role of gut microbes in production of aromatic carboxaldehydes.

Gut microbes play an important role in maintaining our health through the alteration of available nutrients and by the production of small molecules/metabolites. Indole-3-carboxaldehyde (I3A) is an aromatic carboxaldehyde (ArA) synthesized by gut microbes from the aromatic amino acid tryptophan, and has been mechanistically linked to antitumor activity, intestinal homeostasis, and metabolic syndrome. However, the capacity of gut microbes to produce other ArAs and their associations with host health remains largely unexplored due to a lack of methods for their detection and quantification. A st
