Hereditary syndromes with congenital deafness and hypothyroidism have been reported from many countries. The large majority of them are believed to form a single genetic entity, known as Pendred's syndrome, and inherited as a recessive trait. The anatomical basis of deafness is not known. It is generally believed that deafness and hypothyroidism are not causally related, but are independent effects of the gene. Highly specific antisera to triiodothyronine (T(3)) were prepared by immunization of rabbits with T(3)-bovine serum albumin conjugates. Antisera with T(3) binding capacity of up to 600 ng/ml were obtained. The ability of various thyronine derivatives to inhibit the binding of T(3-) (125)I to anti-T(3) serum was found to vary considerably. l-T(3), d-T(3) and several triiodoanalogues were potent inhibitors of the reaction. Studies of immunoglobulin synthesis, total body tumor cell number, and tumor kinetics were carried out in a series of patients with IgG multiple myeloma. The changes in tumor size associated with tumor growth or with regression were underestimated when the concentration of serum M-component was used as the sole index of tumor mass. Partial thyroxine-binding globulin (TBG) deficiency was observed in a euthyroid boy. A family study was conducted in order to trace the defect. Of the 15 members, 3 male subjects were found to be affected, whereas 6 female subjects were shown to be carriers of the trait. The inheritance pattern of the partial TBG deficiency was found to meet the criteria for X chromosome linked transmission. A new procedure for the radioimmunoassay of l-triiodothyronine (T(3)) in human plasma is described in which the iodothyronines are separated from the plasma proteins before incubation with a specific antiserum to T(3). The antibody bound and free T(3) are separated with dextran-coated charcoal. An analysis of 51 patients with Parkinsonism who have developed L-dopa induced dyskinesias is presented. The cause has not been proven, although various hypotheses are discussed. One third of the total number of patients treated developed dyskinesia. These patients tend to respond better to L-dopa than the other group. There is a tendency for the older patient or the patient with long-standing disease to develop dyskinesias. A double-blind cross-over study was carried out in 54 patients with Parkinson's disease to evaluate the efficacy of amantadine hydrochloride as compared to a lactose placebo in the management of this illness. Amantadine proved to be a useful and safe addition to the armamentarium when given in daily doses of 200 mg. Forty-eight per cent of patients experienced moderate to good results while 31% showed no measurable improvement.
