Blood-based biomarkers of ferroptosis in Parkinson's disease.

BACKGROUND: As Parkinson's disease progresses, patients require second-line treatments such as subthalamic stimulation, the benefits of which may be diminished by the onset of non-dopaminergic axial motor and cognitive disorders. This study aimed to identify blood biomarkers of ferroptosis for predicting the progression of Parkinson's disease at the stage of L-DOPA-related complications. METHODS: We analyzed 598 blood samples from patients with PD enrolled in the French multicentric PREDISTIM study. Neurofilament light chain, 4-hydroxy-2-nonenal, glutathione peroxidase activity, ferritin, alp

Putamen dopamine synthesis, vesicular storage, and metabolism in Parkinson disease.

BACKGROUND: Severe putamen dopamine depletion in Parkinson disease (PD) has been attributed to nigrostriatal denervation; however, there are also functional abnormalities in extant terminals (the "sick-but-not-dead" phenomenon). Rates of intra-neuronal processes of synthesis, storage, and metabolism of dopamine complexly influence releasable dopamine stores but have not yet been systematically estimated. METHODS: Post-mortem empirical data were available about putamen tissue contents of 7 reactants, including the autotoxic dopamine metabolite 3,4-dihydroxyphenylacetaldehyde (DOPAL). We constr

Stage-Dependent Changes in Kynurenine Pathway Enzyme Expression Suggest Immune-Related Involvement in Bladder Cancer Progression.

UNLABELLED: L-tryptophan (Trp) metabolism through the L-kynurenine (Kyn) pathway contributes to immune escape in neoplasms, including bladder cancer (BC). Indoleamine 2,3-dioxygenase-1 (IDO1) is a key enzyme, and its catabolites-Kyn, 3-hydroxykynurenine (3HK), and 3-hydroxyanthranilate (3HAA)-depend on other enzymes and play a crucial role in immunomodulation. OBJECTIVES: We investigated the association between Kyn pathway enzyme expression and BC prognostic factors. METHODS: Data from 165 bc patients in the GEO DataSets were analyzed, comparing staging, tumor grade, progression, and recurre

Photodistributed Hyperpigmentation Associated With COVID-19 Vaccination.

BACKGROUND: Although drugs are a common cause of hyperpigmentation, the pathogenesis is unclear and varies based on the offending agent. Classic medications associated with hyperpigmentation secondary to increased melanin deposition include tetracyclines, prostaglandins, nicotine, and antimalarial medications. We report the case of a 70-year-old female who developed photodistributed hyperpigmentation due to increased melanin deposition following COVID-19 vaccination. CASE REPORT: A 70-year-old female presented with a one-year history of diffuse blue-grey hyperpigmentation in a photodistribute

Impaired sleep microarchitecture is associated with locus coeruleus degeneration in Parkinson's disease.

STUDY OBJECTIVES: Sleep disorders are common non-motor symptoms of Parkinson's disease (PD) that significantly impact patients' quality of life. Specifically, alterations in sleep microstructure - such as reduced slow-wave activity and sleep spindles - are prevalent in PD. The locus coeruleus (LC), the brain's primary source of noradrenaline, plays a pivotal role in regulating sleep and wakefulness and is highly vulnerable to neurodegeneration in PD. This study explores whether disruptions in sleep microarchitecture in PD are linked to LC degeneration. METHODS: We assessed polysomnography for

Cell Signaling by Tryptophan Catabolism.

Tryptophan (Trp) metabolism follows three main branches: the kynurenine (KP), serotonin, and indole (IP) pathways. These pathways generate bioactive metabolites that regulate immune responses, redox balance, neurotransmission, metabolic homeostasis, inflammation, and circadian rhythms. A common theme across these pathways is the activation of the aryl hydrocarbon receptor (AhR). Several metabolites from KP, IP, and serotonin act as endogenous AhR ligands or interact indirectly with AhR, but the downstream consequences of this interaction depend on the cellular environment and inflammatory cont

Akkermansia muciniphila reduces neuroinflammation and Aβ deposition via tryptophan metabolism in the APP/PS1 mouse model of Alzheimer's disease.

Akkermansia muciniphila (A. muciniphila), a beneficial gut bacterium, has increasingly attracted interests in Alzheimer's disease (AD) research, its specific role in the microbiota-gut-brain axis still remains unclear. In this study, we demonstrated that A. muciniphila administration improve cognitive deficits and reduce amyloid-beta (Aβ) deposition in APP/PS1 mice, a transgenic model of AD. Subsequently, it is revealed that A. muciniphila administration significantly alters gut microbiota diversity and composition. Mechanically, our metabolomics analysis of cecal contents indicates A. mucinip

Serum metabolite patterns of adipose tissue distribution and body composition subphenotypes.

BACKGROUND: Obesity prevalence is increasing globally, accompanied by increases in obesity-related diseases. While obesity is usually defined by BMI, the development of obesity-related disease might be better characterized by specific adipose tissue (AT) distributions or body composition subphenotypes. Serum metabolite patterns reflecting AT distribution could provide insights into potential underlying pathophysiological pathways and the interplay between AT depots. We therefore aim to identify metabolite signatures associated with specific AT depots and body composition subphenotypes. METHOD

Discovery of urinary metabolite biomarkers of psychiatric disorders using two-sample Mendelian randomization.

BACKGROUND: Mental health disorders cause substantial patient suffering, which could be alleviated through early diagnostic biomarkers. While biomarker discovery is costly, genetic methods utilizing data from large-scale studies, such as Mendelian randomization, may provide a cost-effective approach. METHODS: A two-sample Mendelian randomization analysis was conducted to identify potential urinary biomarkers of seven psychiatric disorders using summary statistics from GWAS data. RESULTS: The analysis revealed 67 analyte-disorder associations, of which 21 were exclusive to a single disorder.

The ability of branched-chain amino acid-to-tyrosine ratio (BTR) to assess preoperative liver function of patients with hepatocellular carcinoma.

Assessment of hepatic functional reserve is critically important for preventing serious complications after hepatectomy such as liver failure. While the indocyanine green clearance test (ICG) is a liver-specific test that is not affected by other organs and is commonly used to evaluate liver reserve capacity. We lack knowledge regarding what test should be performed for patients with jaundice, portal shunts, or intolerance whose liver function cannot be accurately assessed by ICG. To close this gap, we focused on changes in amino acid metabolism associated with impaired liver function. The bra

Low Fischer's ratio is associated with increased mortality in patients with kidney failure.

Metabolic changes in kidney failure, including an altered ratio of plasma branched-chain amino acids (BCAAs) to aromatic amino acids (AAAs), also known as Fischer’s ratio (FR), could potentially affect survival. We analyzed associations of FR with mortality in patients with kidney failure. In 328 kidney failure patients starting dialysis (median age 54 years, 60% men), plasma concentrations of BCAAs (valine, isoleucine, and leucine) and AAAs (phenylalanine, tyrosine) were measured using HPLC. We used Fine-Gray competing-risk regression analysis with renal transplantation as competing risk to c

Gut microbiota-tryptophan-serotonin axis drives anxiety-like behavior via NLRP3-mediated neuronal pyroptosis in the medial prefrontal cortex.

The gut-brain axis plays a critical role in anxiety disorders, yet the underlying mechanisms remain incompletely understood. Using a mouse model of radiofrequency radiation (RFR)-induced anxiety-like behaviors, we employed gut microbiota intervention, regulation of tryptophan metabolites, and other methods to investigate the impact of the gut-brain axis on brain function changes. It was found that gut microbiota dysbiosis disrupts tryptophan metabolism, leading to reduced serotonin (5-HT) levels and NLRP3 inflammasome-mediated neuronal pyroptosis in the medial prefrontal cortex (mPFC). Probiot

Structural determinants of ligand response specificity in the mast cell activating GPCR, MRGPRX2.

The mast cell-specific G-protein-coupled receptor (GPCR) MRGPRX2 (Mas-Related G Protein-coupled Receptor X2) has roles in itch and pain, and it mediates clinically relevant allergy-like responses to a diverse assortment of drugs. The varied responses of individuals to MRGPRX2 agonists, leading to drug hypersensitivity reactions in some cases, suggests the presence of consequential variants in the population. However, genetic associations with drug responses are poorly understood. We used heterologously-expressed MRGPRX2 to investigate the effect of 18 naturally occurring non-synonymous single

Predictive validation of the repeated low-dose reserpine rodent model of parkinsonism.

L-DOPA (LD) is the gold-standard treatment of motor symptoms in Parkinson's disease (PD), conventionally used to prove the predictive value of PD animal models. This study aimed to investigate the predictive validity of an adaptation of the conventional reserpine model of PD: the repeated low-dose reserpine administration in rodents, which promotes progressive motor impairment in catalepsy, vacuous chewing behavior tests, and neurochemical deficits. Specifically, we investigated the effects of acute and chronic treatment with LD on motor alterations induced in mice by this modified protocol. S

COMMD3 mediates melanin synthesis through both clusterin-PAX3 axis and copper-dependent tyrosinase activity in skin pigmentation.

BACKGROUND: Pigmentation-related disorders mainly revolve around abnormal melanogenesis, but the regulatory mechanisms remain unclear. Recent reports have found that COMMD3 plays a role in promoting melanin synthesis. OBJECTIVES: This study aims to investigate the molecular mechanisms and relation with copper homeostasis of a novel pigment-promoting gene, COMMD3, in skin pigmentation. METHODS: A UV-induced pigmentation murine model was created by UVB irradiation for examining the function of COMMD3 in acquired hyperpigmentation. Intracellular copper content was measured to determine the effe

A Combination of the Tryptophan Hydroxylase Inhibitor Telotristat with the mTOR Inhibitor Everolimus as an Effective Strategy Against Neuroendocrine Tumors.

BACKGROUND AND OBJECTIVES: Neuroendocrine tumors are a rare and heterogeneous group of neoplasms that frequently cause carcinoid syndrome, characterized by diarrhea, flushing, and carcinoid heart disease. A recent therapy for carcinoid syndrome involves inhibition of tryptophan hydroxylase, the rate-limiting enzyme in serotonin synthesis, using telotristat ethyl. Telotristat has demonstrated clinical control of carcinoid syndrome; however, its potential antitumoral effects remain unclear. This study aimed to evaluate the antitumor activity of telotristat ethyl alone and in combination with eve
