Genetics of selenoproteins and selenoprotein metabolism - An overview of current concepts and emerging aspects.

Selenoproteins are proteins containing the 21st proteinogenic amino acid selenocysteine in their peptide chain. Selenocysteine lends specific properties to selenoproteins, and selenoenzymes are often catalytically superior to enzymes that would contain cysteine instead. Degradation of peroxides, in particular by GPX4, the master regulator of ferroptosis, is currently receiving a lot of attention owing to its potential in cancer or degenerative disorders. In this context, selenium metabolism has recently become again an active field of research based on the importance to provide selenium to GPX

Supranutritional Selenomethionine During Different Trimesters of Gestation in Beef Cows Alters Gene Expression Patterns in Peripheral Blood Mononuclear Cells at Parturition.

Selenium (Se) is an essential micronutrient. We have previously shown that supranutritional Se-yeast supplementation improves growth and immune function in Se-replete cattle and sheep despite limited impact on selenoprotein and oxidoreductase gene expression, suggesting effects beyond the selenocysteine proteome. This study evaluated the impact of supranutritional Se-yeast during each trimester of pregnancy on transcriptome-wide gene expression in peripheral blood mononuclear cells (PBMCs) at parturition, and the underlying functional categories affected. We administered Se-yeast containing bo

Isoform differentiation studies of arginase: From structural function to design strategies for specific ligands and their application prospects.

Arginase (ARG), a binuclear manganese metalloenzyme, hydrolyzes L-arginine to L-ornithine and urea, competitively regulating NO bioavailability with NOS. Mammals express two isoforms, ARG1 (cytosolic) and ARG2 (mitochondrial), that share a conserved catalytic core yet exert distinct physiological and pathological effects. ARG1 is essential for hepatic urea detoxification, as systemic Arg1 knockout mice develop hyperammonemia within 10-12 days after birth; in contrast, Arg2 knockout mice show only slightly elevated plasma arginine levels with no overt phenotypic abnormalities, this significant

Precision inflammation targeting via myeloperoxidase mediated antioxidant melanin nanomedicines for chronic inflammation therapy.

Chronic inflammation represents a great threat to human health, yet the lack of site-specific therapies often leads to suboptimal efficacy and serious side effects. Here we report an inflammatory targeting strategy leveraging myeloperoxidase (MPO), a well-established pathological marker elevated in inflamed tissues, mediated accumulation of melanin nanomedicines (MNMs) in inflammatory tissues. We demonstrate that MPO mediates the precise targeting of MNMs to inflammatory tissues. MNMs treatment significantly improved survival rate of hepatopulmonary syndrome (HPS) rat model compared with the s

Pantetheinase Vanin-1 potentiates agonist-induced STING activation via S-cysteaminylation.

Stimulator of interferon genes (STING) is a key player in innate immunity, and its dysregulation has been implicated in autoimmune and inflammatory diseases. The activation of STING is regulated by multiple factors, including post-translational modifications (PTMs), but the regulatory network of these PTMs remains incompletely characterized. Here, we report that vascular non-inflammatory molecule-1 (Vanin-1) facilitates STING activation and provide evidence supporting S-cysteaminylation as a previously unrecognized modification of STING. Mechanistically, Vanin-1 hydrolyzes pantetheine to panto

Tai Chi modulating multimodal connectivity patterns and cognitive function in cerebral small vessel disease.

Cerebral small vessel disease (CSVD) significantly contributes to cognitive decline and poses potential risk factors for both dementia and acute cerebrovascular events. Tai Chi has been recognized as an effective mind-body intervention for enhancing cognitive function and promoting neural plasticity. However, the neurobiological mechanisms that underlie how Tai Chi exercises affect cognitive improvement in patients with CSVD remain unclear. We recruited 51 patients with CSVD, randomly assigning them to either a 24-week Tai Chi exercise group (n = 26) or a health education control group (n = 25

LncPTEC mediated homocysteine accumulation elevates oxidative stress via UBQLN1-dependent MTHFD1 ubiquitination in DKD.

Diabetic kidney disease (DKD) is a leading cause of end-stage kidney disease and chronic kidney disease. Oxidative stress, a key driver of renal fibrosis and a hallmark of DKD pathological changes, has been extensively studied for its role in DKD progression. However, its specific mechanisms remain unclear. Here, we show that homocysteine (Hcy) accumulation in proximal tubular epithelial cells (PTECs) is a significant contributor to mitochondrial oxidative stress in DKD. Through single-cell RNA sequencing (scRNA-seq) screening, we identify lncPTEC, a DKD-associated long non-coding RNA (lncRNA)

A near-infrared fluorescent probe for monitoring hepatic cysteine dynamics in drug-induced liver injury guided by cholic acid group.

In this work, a liver-targeted NIR fluorescent probe CA-Cys for Cys was developed by integrating benzothiazole hemicyanine, an acrylate moiety responsive to Cys, and a cholic acid (CA) ligand. The CA group of CA-Cys recognizes the sodium-taurocholate cotransporting polypeptide (NTCP) specifically expressed on hepatocytes, enabling specific liver targeting and excellent resistance to disease interference. CA-Cys exhibits an excellent linear response to Cys with a detection limit of 0.54 μM, rapid response within 30 s, and favorable selectivity, without interference by other reactive sulfur spec

Targeting Cystine Addiction Suppresses Breast Cancer Lung Metastasis.

UNLABELLED: Metastatic disease remains a major cause of cancer-related mortality. Recent studies suggest that dissemination to other organs comes with metabolic changes that allow the metastasizing cancer cells to adapt to new microenvironments. A deeper knowledge of these specific metabolic features and associated vulnerabilities could lead to the development of more effective therapies against metastasis. We used in vivo and ex vivo models of MYC-driven breast tumorigenesis to explore the key metabolic pathways that change when mammary gland tumor (MGT) cells metastasize to the lung. Stable

A smartphone-integrated colorimetric sensor array based on chiral carbon dot nanozymes for rapid biothiol discrimination.

The discrimination of structurally similar biothiols remains a critical challenge in clinical diagnostics, as conventional nanozyme-based sensor arrays often suffer from limited signal diversity and poor discriminative ability. To address this, a colorimetric sensor array using two rationally designed chiral carbon dots (CDs) was constructed. The role of chirality was primarily reflected in regulating the catalytic heterogeneity and enriching the multi-channel response diversity rather than direct enantioselective recognition. The two chiral CDs possess markedly distinct oxidase-like and lacca

Multi-omics reveals regulatory networks and critical early-warning factors for severe disease progression in diabetic patients infected with SARS-CoV-2.

BACKGROUND: Diabetic patients face elevated risks of severe COVID-19, yet the molecular underpinnings of disease progression, particularly for Omicron subvariants, which have predominated since late 2021, remain poorly defined. This observational study leverages a cohort recruited during China's Omicron peak (December 2022-February 2023) to delineate multi-omic signatures underlying severe diabetic COVID-19. METHODS: We enrolled 55 patients across five clinical strata: non-diabetic mild/severe, diabetic mild/severe/fatal. Integrated 4D-DIA proteomics and LC/GC-MS metabolomics were applied, ad

The early postnatal maternal environment influences prepulse inhibition and brain one-carbon metabolites in male mice.

Maternal nutritional environments during early development are thought to influence neurodevelopmental and psychiatric disease susceptibility. While prenatal nutritional effects have been extensively studied within the developmental origins of health and disease (DOHaD) framework, the contribution of postnatal maternal environments remains less understood. In the present study, we investigated whether the postnatal maternal environment influences sensorimotor gating-related behavioral phenotypes and one-carbon metabolism-related metabolites using a cross-fostering paradigm between C57BL/6N (B6

Hepatic methionine-cycle dysregulation is associated with immune-inflammatory and fibrogenic remodeling in biliary atresia.

BACKGROUND: Biliary atresia (BA) is a progressive fibroinflammatory cholangiopathy leading to liver failure in infancy. The contribution of methionine metabolism to BA remains incompletely defined. This study investigated hepatic methionine-cycle alterations and the potential therapeutic effects of S-adenosylmethionine (SAMe). METHODS: Public bulk transcriptomic and single-cell RNA sequencing datasets were analyzed to characterize methionine-cycle-related alterations in BA. Selected genes were validated in clinical liver tissues. Methionine-cycle metabolites, oxidative stress indicators, and

Glutathione-depleting self-immolative nanoparticles boost cuproptosis-driven metalloimmunotherapy for triple-negative breast cancer.

Despite advances in immunotherapy, triple-negative breast cancer (TNBC) remains a major therapeutic challenge owing to its marked aggressiveness, low immunogenicity, and limited responsiveness to immune checkpoint blockade (ICB). Herein, the esterase-responsive polymeric nanoparticles (EsCu@NPs) are developed. The polymer backbone covalently integrates an L-buthionine sulfoximine prodrug to inhibit glutathione biosynthesis, and the nanoparticle matrix encapsulates an elesclomol-copper complex to promote intratumoral copper accumulation. By concurrently depleting glutathione and perturbing copp

N-Homocysteinylation of Cofilin-1 Aggravates Epileptic Pathophysiology via Disruption of Actin Dynamics.

Elevated homocysteine (Hcy) levels contribute to the onset and progression of childhood epilepsy. However, Hcy-lowering therapies provide limited benefits, suggesting additional pathogenic mechanisms. Here, we identified N-homocysteinylated Cofilin-1 (N-Hcy-CFL1) as a molecular link between elevated Hcy levels and childhood epilepsy. We detected N-Hcy-CFL1 in patients with epilepsy and in a mouse model of hyperhomocysteinemia. Mass spectrometry identified Hcy-dependent lysine modifications in CFL1, and functional assays, including actin depolymerization, the F/G-actin ratio, and axonal elongat

Escitalopram and N-acetylcysteine alleviate major depressive disorder with cognitive impairment through YTHDC1/ATF4/CHOP-mediated pyroptosis.

BACKGROUND: Cognitive impairment (CI) is an important factor contributing to poor prognosis in major depressive disorder (MDD). Endoplasmic reticulum stress and microglial inflammatory response are closely associated with the occurrence and development of MDD with CI. Escitalopram (ESC) and N-acetylcysteine (NAC) are two drugs used to improve depression. However, the therapeutic efficacy and underlying molecular mechanisms of ESC and NAC in treating MDD with CI are still not fully understood. METHODS: The depression rat was established using the chronic unpredictable mild stress (CUMS) protoc

Selenomethionine regulates macrophage polarization through mitochondrial dynamics via NF-κB signaling to attenuate sepsis-induced cardiomyopathy.

BACKGROUND: Sepsis frequently leads to multiple organ dysfunction, among which sepsis-induced cardiomyopathy (SIC) is a severe complication associated with high mortality and limited targeted therapies. Selenomethionine (SeMet) possesses potent antioxidant and anti-inflammatory properties; however, its role in SIC remains unclear. METHODS AND RESULTS: SIC models were established in C57BL/6J mice by intraperitoneal injection of LPS at 10 mg/kg or by cecal ligation and puncture (CLP). Mice were pretreated with SeMet (7.5 μg/20 g) by gavage for one week, followed by LPS challenge for 24 h. Cardi

Association of Vitamin B6 and Homocysteine-ACTH Coupling With Depressive and Anxiety Symptoms in Burning Mouth Syndrome: An Exploratory Case-Control Study.

BACKGROUND: Burning mouth syndrome (BMS) is a chronic, idiopathic and debilitating orofacial pain disorder, increasingly discussed within oral dysaesthesia and nociplastic pain frameworks, in which psychological distress and stress-related mechanisms are frequently implicated. Vitamin B6, homocysteine and adrenocorticotropic hormone (ACTH) reflect biologically interconnected micronutrient, one-carbon-metabolic and neuroendocrine pathways, but their relevance to BMS and particularly their interrelationships within patients remains unclear. OBJECTIVES: To compare plasma vitamin B6, homocysteine

Naringenin ameliorates neuropsychiatric deficits in mice fed a methionine-choline-deficient diet.

Neuropsychiatric dysfunction is increasingly being acknowledged as a disabling complication of non-alcoholic steatohepatitis (NASH), but there are no therapeutic approaches. We investigated in the present study the neuroprotective effectiveness of naringenin, a citrus flavonoid with known anti-inflammatory and neurotrophic effects, in a murine NASH model induced by an 8-week methionine-choline-deficient (MCD) diet. Male C57BL/6 mice (n = 8/group) were treated with naringenin (50 mg/kg/day, i.p.) during the final 4 weeks. In behavioral tests, naringenin counteracted cognitive impairment in nove
